  • B-fields in Star-forming Region Observations (BISTRO): Magnetic Fields in the Filamentary Structures of Serpens Main

    • We present 850 mu m polarimetric observations toward the Serpens Main molecular cloud obtained using the POL-2 polarimeter on the James Clerk Maxwell Telescope as part of the B-fields In STar-forming Region Observations survey. These observations probe the magnetic field morphology of the Serpens Main molecular cloud on about 6000 au scales, which consists of cores and six filaments with different physical properties such as density and star formation activity. Using the histogram of relative orientation (HRO) technique, we find that magnetic fields are parallel to filaments in less-dense filamentary structures where NH2 < 0.93 x 10(22) cm(-2) (magnetic fields perpendicular to density gradients), while they are perpendicular to filaments (magnetic fields parallel to density gradients) in dense filamentary structures with star formation activity. Moreover, applying the HRO technique to denser core regions, we find that magnetic field orientations change to become perpendicular to density gradients again at NH2 approximate to 4.6 x 10(22) NH2 approximate to 16 x 10(22) cm(-2), magnetic fields change back to being parallel to density gradients once again, which can be understood to be due to magnetic fields being dragged in by infalling material. In addition, we estimate the magnetic field strengths of the filaments (B-POS = 60-300 mu G)) using the Davis-Chandrasekhar-Fermi method and discuss whether the filaments are gravitationally unstable based on magnetic field and turbulence energy densities.

    • A new species of Redudasys (Macrodasyida: Redudasyidae) isdescribed from Texas and Oregon, U.S.A. A single Texas specimen wasreported (but not formally described and named) in 2014 from an artesianoutflow in Spring Lake, San Marcos, Texas. The Texas specimen provided thefirst record of Redudasys from outside Brazil. Fifteen specimens were foundand studied in 2015 and 2016 from sand pockets among boulders in the LittleNestucca River, near Pacific City, Oregon. Redudasys neotemperatus n. sp. ismorphologically and genetically distinguishable from Redudasys forneriseKisielewski, 1987a, the only formally described species in the genus up untilnow, and morphologically distinguishable from ‘‘Redudasys sp.’’, an unnamedform reported by Garraffoni et al. in 2010 without genetic information. TheTexas and Oregon specimens agree well morphologically and genetically.

  • Impact of cytogenetic abnormalities on outcomes of adult Philadelphia-negative acute lymphoblastic leukemia after allogeneic hematopoietic stem cell transplantation : a study by the Acute Leukemia Working Committee of the Center for International Blood and Marrow Transplant Research

    • Cytogenetic risk stratification at diagnosis has long been one of the most useful tools to assess prognosis in acute lymphoblastic leukemia (ALL). To examine the prognostic impact of cytogenetic abnormalities on outcomes after allogeneic hematopoietic cell transplantation, we studied 1731 adults with Philadelphia-negative ALL in complete remission who underwent myeloablative or reduced intensity/non-myeloablative conditioning transplant from unrelated or matched sibling donors reported to the Center for International Blood and Marrow Transplant Research. A total of 632 patients had abnormal conventional metaphase cytogenetics. The leukemia-free survival and overall survival rates at 5 years after transplantation in patients with abnormal cytogenetics were 40% and 42%, respectively, which were similar to those in patients with a normal karyotype. Of the previously established cytogenetic risk classifications, modified Medical Research Council-Eastern Cooperative Oncology Group score was the only independent prognosticator of leukemia-free survival (P=0.03). In the multivariable analysis, monosomy 7 predicted post-transplant relapse [hazard ratio (HR)=2.11; 95% confidence interval (95% CI): 1.04-4.27] and treatment failure (HR=1.97; 95% CI: 1.20-3.24). Complex karyotype was prognostic for relapse (HR=1.69; 95% CI: 1.06-2.69), whereas t(8;14) predicted treatment failure (HR=2.85; 95% CI: 1.35-6.02) and overall mortality (HR=3.03; 95% CI: 1.44-6.41). This large study suggested a novel transplant-specific cytogenetic scheme with adverse [monosomy 7, complex karyotype, del(7q), t(8;14), t(11;19), del(7q), tetraploidy/near triploidy], intermediate (normal karyotype and all other abnormalities), and favorable (high hyperdiploidy) risks to prognosticate leukemia-free survival (P=0.02). Although some previously established high-risk Philadelphia-negative cytogenetic abnormalities in ALL can be overcome by transplantation, monosomy 7, complex karyotype, and t(8;14) continue to pose significant risks and yield inferior outcomes.

    • Toroidal computations are performed using the MARS-F code (Liu et al 2000 Phys. Plasmas 7 3681), in order to understand correlations between the plasma response and the observed mitigation of the edge localized modes (ELM) using resonant magnetic perturbation fields in ASDEX Upgrade. In particular, systematic numerical scans of the edge safety factor reveal that the amplitude of the resonant poloidal harmonic of the response radial magnetic field near the plasma edge, as well as the plasma radial displacement near the X-point, can serve as good indicators for predicting the optimal toroidal phasing between the upper and lower rows of coils in ASDEX Upgrade. The optimal coil phasing scales roughly linearly with the edge safety factor , for various choices of the toroidal mode number n = 1-4 of the coil configuration. The optimal coil phasing is also predicted to vary with the upper triangularity of the plasma shape in ASDEX Upgrade. Furthermore, multiple resonance effects of the plasma response, with continuously varying , are computationally observed and investigated.

  • Incidence of Psoriatic Arthritis in Patients with Skin Psoriasis and Associated Risk Factors: A Retrospective Population-based Cohort Study in Swedish Routine Clinical Care

    • The incidence of psoriatic arthritis in patients with psoriasis is unclear; existing estimates differ by a factor of ten. Complete population-level data is needed to provide accurate estimates with high confidence. A total of 123,814 adults with psoriasis, free from pre-existing psoriatic arthritis, were identified in population-based data from secondary care in Sweden during 2007 to 2017. Incidence was calculated as the number of psoriatic arthritis diagnosis events per 100 patient-years. Time to diagnosis was assessed using cumulative incidence and Cox proportional hazards models to identify risk factors. Incidence of psoriatic arthritis in patients with psoriasis was 1.69 per 100 patient-years (95% confidence interval 1.65-1.72) overall, and 1.48, 3.00, and 5.49 per 100 patient-years in patients with mild, moderate and severe psoriasis, respectively. Risk of psoriatic arthritis was 3.2 times higher amongst patients with severe psoriasis compared with mild disease. Dermatologists should regularly assess risk factors for psoriatic arthritis in clinical practice in order to improve the detection of psoriatic arthritis.

    • The penetration dynamics of the resonant magnetic perturbation (RMP) field is sim-ulated in the full toroidal geometry, under realistic plasma conditions in MAST experiments.The physics associated with several aspects of the RMP penetration - the plasma responseand rotational screening, the resonant and non-resonant torques and the toroidal momentumbalance - are highlighted. In particular, the plasma response is found to significantly amplifythe non-resonant component of the RMP field for some of the MAST plasmas. A fast rotatingplasma, in response to static external magnetic fields, experiences a more distributed electro-magnetic torque due to the resonance with continuum waves in the plasma. At fast plasmaflow (such as for the MAST plasma), the electromagnetic torque is normally dominant overthe neoclassical toroidal viscous (NTV) torque. However, at sufficiently slow plasma flow,the NTV torque can play a significant role in the toroidal momentum balance, thanks to theprecession drift resonance enhanced, so called superbanana plateau regime.

    • The plasma response to the vacuum resonant magnetic perturbation (RMP) fields, produced by the ELM control coils in ASDEX Upgrade experiments, is computationally modelled using the MARS-F/K codes (Liu et al 2000 Phys, Plasmas 7 3681, Liu et al 2008 Phys. Plasmas 15 112503), A systematic investigation is carried out, considering various plasma and coil configurations as in the ELM control experiments. The low q plasmas, with q95 3.8 (q95 is the safety factor q value at 95% of the equilibrium poloidal flux), responding to low a (n is the toroidal mode number) field perturbations from each single row of the ELM coils, generates a core kink amplification effect. Combining two rows, with different toroidal phasing, thus leads to either cancellation or reinforcement of the core kink response, which in turn determines the poloidal location of the peak plasma surface displacement, The core kink response is typically weak for the a = 4 coil configuration at low q, and for the n = 2 configuration but only at high q (q(95) similar to 5.5). A phase shift of around 60 degrees for low q plasmas, and around 90 degrees for high q plasmas, is found in the coil phasing, between the plasma response field and the vacuum RMP field, that maximizes the edge resonant field component, This leads to an optimal coil phasing of about 100 (-100) degrees for low (high) q plasmas, that maximizes both the edge resonant field component and the plasma surface displacement near the X-point of the separatrix. This optimal phasing closely corresponds to the best ELM mitigation observed in experiments. A strong parallel sound wave damping moderately reduces the core kink response but has minor effect on the edge peeling response. For low q plasmas, modelling shows that both the resonant electromagnetic torque and the neoclassical toroidal viscous (NTV) torque (due to the presence of 3D magnetic field perturbations) contribute to the toroidal flow damping, in particular near the plasma edge region. For high q plasmas, however, significant amount of torque is also produced in the bulk plasma region, and the contributions from the electromagnetic, the NTV, and the torque associated with the Reynolds stress, all

  • Tumour-associated macrophages drive stromal cell-dependent collagen crosslinking and stiffening to promote breast cancer aggression

    • Stromal stiffening accompanies malignancy, compromises treatment and promotes tumour aggression. Clarifying the molecular nature and the factors that regulate stromal stiffening in tumours should identify biomarkers to stratify patients for therapy and interventions to improve outcome. We profiled lysyl hydroxylase-mediated and lysyl oxidase-mediated collagen crosslinks and quantified the greatest abundance of total and complex collagen crosslinks in aggressive human breast cancer subtypes with the stiffest stroma. These tissues harbour the highest number of tumour-associated macrophages, whose therapeutic ablation in experimental models reduced metastasis, and decreased collagen crosslinks and stromal stiffening. Epithelial-targeted expression of the crosslinking enzyme, lysyl oxidase, had no impact on collagen crosslinking in PyMT mammary tumours, whereas stromal cell targeting did. Stromal cells in microdissected human tumours expressed the highest level of collagen crosslinking enzymes. Immunohistochemical analysis of biopsies from a cohort of patients with breast cancer revealed that stromal expression of lysyl hydroxylase 2, an enzyme that induces hydroxylysine aldehyde-derived collagen crosslinks and stromal stiffening, correlated significantly with disease specific mortality. The findings link tissue inflammation, stromal cell-mediated collagen crosslinking and stiffening to tumour aggression and identify lysyl hydroxylase 2 as a stromal biomarker.
