    • Separating wounded serosa by physical barriers is the only clinically approved adjunct for postoperative adhesion prevention. Since the optimal adhesion barrier has not been found, it is essential to improve our pathogenic understanding of adhesion formation and to compare the effects of different barrier materials on tissue and cells. Wistar rats underwent standardized peritoneal damage and were treated either with Seprafilm, Adept, Intercoat, Spraygel, SupraSeal or remained untreated as a control. 14 days postoperatively, the lesions were explanted and histomorphologically analyzed using the European ISO score to evaluate material implants. Striking differences between the material groups were present regarding the inflammation, fibrosis, and foreign body reaction. According to the ISO score, Intercoat and Spraygel were considered as nonirritating to tissue. Adept, Seprafilm, and SupraSeal were assessed as mild-irritating materials. Interestingly, the most effective material in adhesion prevention revealed moderate inflammation accompanied by minor fibrosis. The degree of inflammation to barrier materials does not predict the efficacy in the prevention of adhesions. Histopathological investigations are crucial to improve our understanding of the cellular mechanisms during adhesion formation and elucidate the tissue response to material approaches used in adhesion prevention. This will lead to improved antiadhesive strategies and the development of functional barrier biomaterials.

  • The peculiar radio galaxy 4C 35.06 : a case for recurrent AGN activity?

    • Using observations obtained with the LOw Fequency ARray (LOFAR), the Westerbork Synthesis Radio Telescope (WSRT) and archival Very Large Array (VLA) data, we have traced the radio emission to large scales in the complex source 4C 35.06 located in the core of the galaxy cluster Abell 407. At higher spatial resolution (~ 4″), the source was known to have two inner radio lobes spanning 31 kpc and a diffuse, low-brightness extension running parallel to them, offset by about 11 kpc (in projection). At 62 MHz, we detect the radio emission of this structure extending out to 210 kpc. At 1.4 GHz and intermediate spatial resolution (~ 30″), the structure appears to have a helical morphology. We have derived the characteristics of the radio spectral index across the source. We show that the source morphology is most likely the result of at least two episodes of AGN activity separated by a dormant period of around 35 Myr. The outermost regions of radio emission have a steep spectral index (α< − 1), indicative of old plasma. We connect the spectral index properties of the resolved source structure with the integrated fluxdensity spectral index of 4C 35.06 and suggest an explanation for its unusual integrated flux density spectral shape (a moderately steep power law with no discernible spectral break), possibly providing a proxy for future studies of more distant radio sources through inferring their detailed spectral index properties and activity history from their integrated spectral indices. The AGN is hosted by one of the galaxies located in the cluster core of Abell 407. We propose that it is intermittently active as it moves in the dense environment in the cluster core. In this scenario, the AGN turned on sometime in the past, and has produced the helical pattern of emission, possibly a sign of jet precession/merger during that episode of activity. Using LOFAR, we can trace the relic plasma from that episode of activity out to greater distances from the core than ever before. Using the the WSRT, we detect H I in absorption against the center of the radio source. The absorption profile is relatively broad (FWHM of 288 kms-1), similar to what is found in other clusters. The derived column density is NHI ~ 4 × 1020 cm-2 for a Tspin = 100 K. This detection supports the connection – already suggested for other restarted radio sources – between the presence of cold gas and restarting activity. The cold gas appears to be dominated by a blue-shifted component although the broad H I profile could also include gas with different kinematics. Understanding the duty cycle of the radio emission as well as the triggering mechanism for starting (or restarting) the radio-loud activity can provide important constraints to quantify the impact of AGN feedback on galaxy evolution. The study of these mechanisms at low frequencies using morphological and spectral information promises to bring new important insights in this field.

  • LOFAR discovery of a quiet emission mode in PSR B0823+26

    • PSR B0823+26, a 0.53-s radio pulsar, displays a host of emission phenomena over time-scales of seconds to (at least) hours, including nulling, subpulse drifting, and mode-changing. Studying pulsars like PSR B0823+26 provides further insight into the relationship between these various emission phenomena and what they might teach us about pulsar magnetospheres. Here we report on the LOFAR (Low-Frequency Array) discovery that PSR B0823+26 has a weak and sporadically emitting ‘quiet’ (Q) emission mode that is over 100 times weaker (on average) and has a nulling fraction forty-times greater than that of the more regularly-emitting ‘bright’ (B) mode. Previously, the pulsar has been undetected in the Q mode, and was assumed to be nulling continuously. PSR B0823+26 shows a further decrease in average flux just before the transition into the B mode, and perhaps truly turns off completely at these times. Furthermore, simultaneous observations taken with the LOFAR, Westerbork, Lovell, and Effelsberg telescopes between 110 MHz and 2.7 GHz demonstrate that the transition between the Q mode and B mode occurs within one single rotation of the neutron star, and that it is concurrent across the range of frequencies observed.

    • While Bayesian Optimization (BO) is a very popular method for optimizing expensive black-box functions, it fails to leverage the experience of domain experts. This causes BO to waste function evaluations on bad design choices (e.g., machine learning hyperparameters) that the expert already knows to work poorly. To address this issue, we introduce Bayesian Optimization with a Prior for the Optimum (BOPrO). BOPrO allows users to inject their knowledge into the optimization process in the form of priors about which parts of the input space will yield the best performance, rather than BO’s standard priors over functions, which are much less intuitive for users. BOPrO then combines these priors with BO’s standard probabilistic model to form a pseudo-posterior used to select which points to evaluate next. We show that BOPrO is around 6.67 × faster than state-of-the-art methods on a common suite of benchmarks, and achieves a new state-of-the-art performance on a real-world hardware design application. We also show that BOPrO converges faster even if the priors for the optimum are not entirely accurate and that it robustly recovers from misleading priors.

  • Exclusion from spheroid formation identifies loss of essential cell-cell adhesion molecules in colon cancer cells

    • Many cell lines derived from solid cancers can form spheroids, which recapitulate tumor cell clusters and are more representative of the in vivo situation than 2D cultures. During spheroid formation, a small proportion of a variety of different colon cancer cell lines did not integrate into the sphere and lost cell-cell adhesion properties. An enrichment protocol was developed to augment the proportion of these cells to 100% purity. The basis for the separation of spheroids from non-spheroid forming (NSF) cells is simple gravity-sedimentation. This protocol gives rise to sub-populations of colon cancer cells with stable loss of cell-cell adhesion. SW620 cells lacked E-cadherin, DLD-1 cells lost α-catenin and HCT116 cells lacked P-cadherin in the NSF state. Knockdown of these molecules in the corresponding spheroid-forming cells demonstrated that loss of the respective proteins were indeed responsible for the NSF phenotypes. Loss of the spheroid forming phenotype was associated with increased migration and invasion properties in all cell lines tested. Hence, we identified critical molecules involved in spheroid formation in different cancer cell lines. We present here a simple, powerful and broadly applicable method to generate new sublines of tumor cell lines to study loss of cell-cell adhesion in cancer progression.

  • Efficacy and safety of adjunctive perampanel for the treatment of refractory partial seizures: A pooled analysis of three phase III studies

    • Purpose Three phase III studies (304 [ClinicalTrials.gov identifier: NCT00699972], 305 [NCT00699582], 306 [NCT00700310]) evaluated perampanel, an α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor antagonist, as adjunctive therapy for refractory partial seizures. We report post hoc analyses of pooled study data by randomized dose. Methods Patients with partial seizures despite receiving 1-3 antiepileptic drugs were randomized to once-daily placebo, perampanel 8 or 12 mg (studies 304, 305), or placebo, perampanel 2, 4, or 8 mg (study 306). Studies included a 6-week baseline period and double-blind treatment phase (6-week titration; 13-week maintenance). Primary end points were median change in partial seizure frequency (baseline vs. double-blind phase) and percentage of patients achieving ≥50% reduction in seizure frequency (baseline vs. maintenance). Here, these end points, together with secondary, exploratory, and safety end points, were assessed using pooled study data. Key Findings The pooled intent-to-treat analysis set (randomized, treated patients with any seizure data) included 1,478 patients. Median changes in partial seizure frequency were greater with perampanel than placebo (perampanel 4 mg, -23.3%; 8 mg, -28.8%; 12 mg, -27.2%; placebo, -12.8%; p < 0.01, each dose vs. placebo), as were 50% responder rates (perampanel 4 mg, 28.5%; 8 mg, 35.3%; 12 mg, 35.0%; placebo, 19.3%; p < 0.05, each dose vs. placebo). In addition, median changes in complex partial plus secondary generalized seizure frequency were also greater with perampanel than placebo (perampanel 4 mg, -31.2%; 8 mg, -35.6%; 12 mg, -28.6%; placebo, -13.9%). Perampanel was generally well tolerated. The most frequent treatment-emergent adverse events (TEAEs) were dizziness, somnolence, and headache. Most TEAEs were mild/moderate; relatively few patients experienced severe TEAEs (placebo, 5.4%; perampanel, 8.9%) or serious TEAEs (placebo, 5.0%; perampanel, 5.5%). There were no deaths and no clinically important mean changes in laboratory values, electrocardiography (ECG) findings, or vital signs. Significance Perampanel reduced partial seizure frequency and improved responder rates compared with placebo, with an acceptable tolerability profile. © 2013 International League Against Epilepsy.
