    • Delivery time and delivery reliability are two top-level measures of delivery performance, and they both influence customers perception of service value. However, the classic queue-pricing literature emphasizes the former and ignores the latter. In order to bridge the gap between research and practice, this study addresses the interactive impact of price, delivery time, and delivery-reliability level on the equilibrium behaviour of rational customers and the optimal decisions of a revenue-maximizing service provider. We assume that the customers sensitivity to the delivery-reliability level is characterized by an increasing concave service value function. We model the operations of the service provider as an M/M/1 queue. Two cases are investigated: homogeneous customers and heterogeneous customers. For the homogeneous customers case, we analytically characterize the service providers optimal price, delivery time, and delivery reliability level decisions. We show how the service providers decisions on whether to provide faster or more reliable service are affected when different problem parameters are subject to variation. For instance, when customers become more sensitive to the delivery-reliability level, the service provider increases the delivery-reliability level at the expense of a longer delivery time. However, the optimal price may either increase or decrease depending on a benchmark value for the delivery-reliability level. For the heterogeneous customers case, our results suggest that when the potential arrival rate is sufficiently high, the service provider always benefits from markets with higher levels of customer heterogeneity.   • Large scale multifactorial likelihood quantitative analysis of BRCA1 and BRCA2 variants : An ENIGMA resource to support clinical variant classification

    • The multifactorial likelihood analysis method has demonstrated utility for quantitative assessment of variant pathogenicity for multiple cancer syndrome genes. Independent data types currently incorporated in the model for assessing BRCA1 and BRCA2 variants include clinically calibrated prior probability of pathogenicity based on variant location and bioinformatic prediction of variant effect, co-segregation, family cancer history profile, co-occurrence with a pathogenic variant in the same gene, breast tumor pathology, and case-control information. Research and clinical data for multifactorial likelihood analysis were collated for 1395 BRCA1/2 predominantly intronic and missense variants, enabling classification based on posterior probability of pathogenicity for 734 variants: 447 variants were classified as (likely) benign, and 94 as (likely) pathogenic; 248 classifications were new or considerably altered relative to ClinVar submissions. Classifications were compared to information not yet included in the likelihood model, and evidence strengths aligned to those recommended for ACMG/AMP classification codes. Altered mRNA splicing or function relative to known non-pathogenic variant controls were moderately to strongly predictive of variant pathogenicity. Variant absence in population datasets provided supporting evidence for variant pathogenicity. These findings have direct relevance for BRCA1 and BRCA2 variant evaluation, and justify the need for gene-specific calibration of evidence types used for variant classification.   • A global view of aspen : conservation science for widespread keystone systems

    • Across the northern hemisphere, six species of aspen (Populus spp.) play a disproportionately important role in promoting biodiversity, sequestering carbon, limiting forest disturbances, and providing other ecosystem services. These species are illustrative of efforts to move beyond single-species conservation because they facilitate hundreds of plants and animals worldwide. This review is intended to place aspen in a global conservation context by focusing on the many scientific advances taking place in such biologically diverse systems. In this manner, aspen may serve as a model for other widespread keystone systems where science-based practice may have world implications for biodiversity conservation. In many regions, aspen can maintain canopy dominance for decades to centuries as the sole major broadleaf trees in forested landscapes otherwise dominated by conifers. Aspen ecosystems are valued for many reasons, but here we highlight their potential as key contributors to regional and global biodiversity. We present global trends in research priorities, strengths, and weaknesses based on, 1) a qualitative survey, 2) a systematic literature analysis, and 3) regional syntheses of leading research topics. These regional syntheses explore important aspen uses, threats, and research priorities with the ultimate intent of research sharing focused on sound conservation practice. In all regions, we found that aspen enhance biodiversity, facilitate rapid (re)colonization in natural and damaged settings (e.g., abandoned mines), and provide adaptability in changing environments. Common threats to aspen ecosystems in many, but not all, regions include effects of herbivory, land clearing, logging practices favoring conifer species, and projected climate warming. We also highlight regional research gaps that emerged from the three survey approaches above. We believe multi-scale research is needed that examines disturbance processes in the context of dynamic climates where ecological, physiological, and genetic variability will ultimately determine widespread aspen sustainability. Based on this global review of aspen research, we argue for the advancement of the "mega-conservation" strategy, centered on the idea of sustaining a set of common keystone communities (aspen) that support wide arrays of obligate species. This approach contrasts with conventional preservation which focuses limited resources on individual species residing in narrow niches.

    • The recurrent extreme El Niño events are commonly linked to reduced vegetation growth and the land carbon sink over many but discrete regions of the Northern Hemisphere (NH). However, we reported here a pervasive and continuous vegetation greening and no weakened land carbon sink in the maturation phase of the 2015/2016 El Niño event over the NH (mainly in the extra-tropics), based on multiple evidences from remote sensing observations, global ecosystem model simulations and atmospheric CO2 inversions. We discovered a significant compensation effect of the enhanced vegetation growth in spring on subsequent summer/autumn vegetation growth that sustained vegetation greening and led to a slight increase in the land carbon sink over the spring and summer of 2015 (average increases of 23.34% and 0.63% in net ecosystem exchange from two independent datasets relative to a 5-years average before the El Niño event, respectively) and spring of 2016 (6.82%), especially in the extra-tropics of the NH, where the water supply during the pre-growing-season (November of the previous year to March of the current year) had a positive anomaly. This seasonal compensation effect was much stronger than that in 1997 and 1998 and significantly alleviated the adverse impacts of the 2015/2016 El Niño event on vegetation growth during its maturation phase. The legacy effect of water supply during the pre-growing-season on subsequent vegetation growth lasted up to approximately six months. Our findings highlight the role of seasonal compensation effects on mediating the land carbon sink in response to episodic extreme El Niño events.

  • High potential of stable carbon sequestration in phytoliths of China's grasslands

    • Phytolith carbon (C) sequestration plays a key role in mitigating global climate change at a centennial to millennial time scale. However, previous estimates of phytolith-occluded carbon (PhytOC) storage and potential in China's grasslands have large uncertainties mainly due to multiple data sources. This contributes to the uncertainty in predicting long-term C sequestration in terrestrial ecosystems using Earth System Models. In this study, we carried out an intensive field investigation (79 sites, 237 soil profiles [0-100 cm], and 61 vegetation assessments) to quantify PhytOC storage in China's grasslands and to better explore the biogeographical patterns and influencing factors. Generally, PhytOC production flux and soil PhytOC density in both the Tibetan Plateau and the Inner Mongolian Plateau had a decreasing trend from the Northeast to the Southwest. The aboveground PhytOC production rate in China's grassland was 0.48 x 10(6) t CO2 a(-1), and the soil PhytOC storage was 383 x 10(6) t CO2. About 45% of soil PhytOC was stored in the deep soil layers (50-100 cm), highlighting the importance of deep soil layers for C stock assessments. Importantly, the Tibetan Plateau had the greatest contribution (more than 70%) to the PhytOC storage in China's grasslands. The results of multiple regression analysis indicated that altitude and soil texture significantly influenced the spatial distribution of soil PhytOC, explaining 78.1% of the total variation. Soil phytolith turnover time in China's grasslands was mainly controlled by climatic conditions, with the turnover time on the Tibetan Plateau being significantly longer than that on the Inner Mongolian Plateau. Our results offer more accurate estimates of the potential for phytolith C sequestration from ecological restoration projects in degraded grassland ecosystems. These estimates are essential to parameterizing and validating global C models.

    • BackgroundConfined ongoing ischemia after ischemia-reperfusion injury (IRI) may alter myocardial recovery. We evaluated in a rat cardiac transplantation model whether distal persistent myocardial ischemia (dMI) and remote preconditioning (RPreC) have a remote myocardial impact after IRI.Material and methodsSyngeneic heterotopic cardiac transplantation was performed on 29 Fischer344 rats to induce IRI, including nine rats which underwent distal ligation of the left anterior coronary artery (LAD) to yield distal MI (IRI+ dMI). RPreC was applied by occluding the left renal artery 5 min prior to reperfusion in six rats with IRI (IRI+ RPreC) as well as in seven with distal MI (IRI+ dMI+ RPreC). Microdialysis, histology and qRT-PCR for inducible nitric oxide synthase (iNOS) and endothelial nitric oxide synthase (eNOS) were performed after graft harvesting.ResultsIn contrast to IRI + dMI + RPreC (39 ± 7 μmol), glutamate decreased in IRI + RPreC and IRI + dMI as compared with IRI (26 ± 3 and 31 ± 8 vs 91 ± 20, μmol respectively, p < 0.007). The relative number of vacuolated intramyocardial artery nuclei decreased in IRI + dMI as compared with IRI (0.02 ± 0.01, range 0-12 vs. 0.42 ± 0.31, range 0-3.25 PSU respectively, p < 0.04). iNOS expression decreased in IRI + RPreC as compared with IRI (p < 0.04), and eNOS expression decreased in IRI + dMI + RPreC as compared with IRI + dMI (p < 0.006) along with increased glycerol release.ConclusionsdMI after IRI has a potentially beneficial myocardial impact after cardiac arrest, which is hampered by RPreC.

  • Storage, patterns and influencing factors for soil organic carbon in coastal wetlands of China

    • Soil organic carbon (SOC) in coastal wetlands, also known as "blue C," is an essential component of the global C cycles. To gain a detailed insight into blue C storage and controlling factors, we studied 142 sites across ca. 5000 km of coastal wetlands, covering temperate, subtropical, and tropical climates in China. The wetlands represented six vegetation types (Phragmites australis, mixed of P. australis and Suaeda, single Suaeda, Spartina alterniflora, mangrove [Kandelia obovata and Avicennia marina], tidal flat) and three vegetation types invaded by S. alterniflora (P. australis, K. obovata, A. marina). Our results revealed large spatial heterogeneity in SOC density of the top 1-m ranging 40-200 Mg C ha(-1), with higher values in mid-latitude regions (25-30 degrees N) compared with those in both low- (20 degrees N) and high-latitude (38-40 degrees N) regions. Vegetation type influenced SOC density, with P. australis and S. alterniflora having the largest SOC density, followed by mangrove, mixed P. australis and Suaeda, single Suaeda and tidal flat. SOC density increased by 6.25 Mg ha(-1) following S. alterniflora invasion into P. australis community but decreased by 28.56 and 8.17 Mg ha(-1) following invasion into K. obovata and A. marina communities. Based on field measurements and published literature, we calculated a total inventory of 57 x 10(6) Mg C in the top 1-m soil across China's coastal wetlands. Edaphic variables controlled SOC content, with soil chemical properties explaining the largest variance in SOC content. Climate did not control SOC content but had a strong interactive effect with edaphic variables. Plant biomass and quality traits were a minor contributor in regulating SOC content, highlighting the importance of quantity and quality of OC inputs and the balance between production and degradation within the coastal wetlands. These findings provide new insights into blue C stabilization mechanisms and sequestration capacity in coastal wetlands.

    • Grassland ecosystems play an important role in the global terrestrial silicon (Si) cycle, and Si is a beneficial elementand structural constituent for the growth of grasses. In previous decades, grasslands have been degradedto different degrees because of the drying climate and intense human disturbance. However, the impact of grasslanddegradation on the distribution and bioavailability of soil Si is largely unknown. Here, we investigated vegetationand soil conditions of 30 sites to characterize different degrees of degradation for grasslands in the agropastoralecotone of northern China. We then explored the impact of grassland degradation on the distributionand bioavailability of soil Si, including total Si and four forms of noncrystalline Si in three horizons (0–10,10–20 and 20–40 cm) of different soil profiles. The concentrations of noncrystalline Si in soil profiles significantlydecreased with increasing degrees of degradation, being 7.35 ± 0.88 mg g−1, 5.36 ± 0.39 mg g−1, 3.81 ±0.37 mg g−1 and 3.60±0.26 mg g−1 in non-degraded, lightly degraded, moderately degraded and seriously degradedgrasslands, respectively. Moreover, the storage of noncrystalline Si decreased from higher than 40 t ha−1to lower than 23 t ha−1. The corresponding bioavailability of soil Si also generally decreased with grassland degradation.These processes may not only affect the Si pools and fluxes in soils but also influence the Si uptake in plants. We suggest that grassland degradation can significantly affect the global grassland Si cycle. Grasslandmanagement methods such as fertilizing and avoiding overgrazing can potentially double the content and storageof noncrystalline Si in soils, thereby enhancing the soil Si bioavailability by N17%.

    • A large number of variants identified through clinical genetic testing in disease susceptibility genes are of uncertain significance (VUS). Following the recommendations of the American College of Medical Genetics and Genomics (ACMG) and Association for Molecular Pathology (AMP), the frequency in case-control datasets (PS4 criterion) can inform their interpretation. We present a novel case-control likelihood ratio-based method that incorporates gene-specific age-related penetrance. We demonstrate the utility of this method in the analysis of simulated and real datasets. In the analysis of simulated data, the likelihood ratio method was more powerful compared to other methods. Likelihood ratios were calculated for a case-control dataset of BRCA1 and BRCA2 variants from the Breast Cancer Association Consortium (BCAC) and compared with logistic regression results. A larger number of variants reached evidence in favor of pathogenicity, and a substantial number of variants had evidence against pathogenicity-findings that would not have been reached using other case-control analysis methods. Our novel method provides greater power to classify rare variants compared with classical case-control methods. As an initiative from the ENIGMA Analytical Working Group, we provide user-friendly scripts and preformatted Excel calculators for implementation of the method for rare variants in BRCA1, BRCA2, and other high-risk genes with known penetrance.

    • The formation and evolution of the landscape of the Chinese Loess Plateau (CLP) is debated because of uncertainties regarding dust provenance. We present a quantitative estimation of dust source contributions to the CLP, based on more than 37,100 detrital zircon U-Pb ages, combined with mineral assemblages and isotope analyses. Our results reveal that the CLP was stepwise formed by similar to 8 million years (Ma) and is mainly composed of material from the Northeastern Qinghai-Tibetan Plateau, with stepwise shifts in relative contributions of different eolian silt sources occurring at similar to 2.6 Ma and 1.5 to 1.2 Ma. We infer that these changes were driven by stepwise global cooling, which induced aridification and enhanced silt production in glaciated-cold climate dust source regions, as well as dust ablation in the expanded and regions. We propose that global cooling, rather than regional tectonic deformation, was the main driver of the formation and evolution of the CLP during late Cenozoic.
