  • Epigenome-wide association study of body mass index, and the adverse outcomes of adiposity

    • Approximately 1.5 billion people worldwide are overweight or affected by obesity, and are at risk of developing type (2) diabetes, cardiovascular disease and related metabolic and inflammatory disturbances(1,2). Although the mechanisms linking adiposity to associated clinical conditions are poorly understood, recent studies suggest that adiposity may influence DNA methylation(3-6), a key regulator of gene expression and molecular phenotype(7). Here we use epigenome-wide association to show that body mass index (BMI; a key measure of adiposity) is associated with widespread changes in DNA methylation (187 genetic loci with P < 1 x 10(-7), range P = 9.2 x 10(-8) to 6.0 x 10(-46); n = 10,261 samples). Genetic association analyses demonstrate that the alterations in DNA methylation are predominantly the consequence of adiposity, rather than the cause. We find that methylation loci are enriched for functional genomic features in multiple tissues (P < 0.05), and show that sentinel methylation markers identify gene expression signatures at 38 loci (P < 9.0 x 10(-6), range P = 5.5 x 10(-6) to 6.1 x 10(-35), n = 1,785 samples). The methylation loci identify genes involved in lipid and lipoprotein metabolism, substrate transport and inflammatory pathways. Finally, we show that the disturbances in DNA methylation predict future development of type 2 diabetes (relative risk per 1 standard deviation increase in methylation risk score: 2.3 (2.07-2.56); P = 1.1 x 10(-54)). Our results provide new insights into the biologic pathways influenced by adiposity, and may enable development of new strategies for prediction and prevention of type 2 diabetes and other adverse clinical consequences of obesity.

    • The genus Monascus was described by van Tieghem (1884) to accommodate M. ruber and M. mucoroides, two species with non-ostiolate ascomata. Species delimitation in the genus is still mainly based on phenotypic characters, and taxonomic studies that include sequence data are limited. The genus is of economic importance. Species are used in fermented Asian foods as food colourants (e.g. 'red rice' (ang-kak, angka)) and found as spoilage organisms, and recently Monascus was found to be essential in the lifecycle of stingless bees. In this study, a polyphasic approach was applied combining morphological characters, ITS, LSU, beta-tubulin, calmodulin and RNA polymerase II second largest subunit sequences and extrolite data, to delimit species and to study phylogenetic relationships in Monascus. Furthermore, 30 Monascus isolates from honey, pollen and nests of stingless bees in Brazil were included. Based on this polyphasic approach, the genus Monascus is resolved in nine species, including three new species associated with stingless bees (M. flavipigmentosus sp. nov., M. mellicola sp. nov., M. recifensis sp. nov., M. argentinensis, M. floridanus, M. lunisporas, M. pallens, M. purpureus, M. ruber), and split in two new sections (section Floridani sect. nov., section Rubri sect. nov.). Phylogenetic analysis showed that the xerophile Monascus eremophilus does not belong in Monascus and monophyly in Monascus is restored with the transfer of M. eremophilus to Penicillium (P. eremophilum comb. nov.). A list of accepted and excluded Monascus and Basipetospora species is given, together with information on (ex-)types cultures and barcode sequence data.

    • Investigating the chemical homogeneity of stars born from the same molecular cloud at virtually the same time is very important for our understanding of the chemical enrichment of the interstellar medium and with it the chemical evolution of the Galaxy. One major cause of inhomogeneities in the abundances of open clusters is stellar evolution of the cluster members. In this work, we investigate variations in the surface chemical composition of member stars of the old open cluster M67 as a possible consequence of atomic diffusion effects taking place during the main-sequence phase. The abundances used are obtained from high-resolution UVES/FLAMES spectra within the framework of the Gaia-ESO Survey. We find that the surface abundances of stars on the main sequence decrease with increasing mass reaching a minimum at the turn-off. After deepening of the convective envelope in subgiant branch stars, the initial surface abundances are restored. We found the measured abundances to be consistent with the predictions of stellar evolutionary models for a cluster with the age and metallicity of M67. Our findings indicate that atomic diffusion poses a non-negligible constraint on the achievable precision of chemical tagging methods.

  • Classical analogy for the deflection of flux avalanches by a metallic layer

    • Sudden avalanches of magnetic flux bursting into a superconducting sample undergo deflections of their trajectories when encountering a conductive layer deposited on top of the superconductor. Remarkably, in some cases the flux is totally excluded from the area covered by the conductive layer. We present a simple classical model that accounts for this behaviour and considers a magnetic monopole approaching a semi-infinite conductive plane. This model suggests that magnetic braking is an important mechanism responsible for avalanche deflection.

  • Flux penetration in a superconducting film partially capped with a conducting layer

    • The influence of a conducting layer on the magnetic flux penetration in a superconducting Nb film is studied by magneto-optical imaging. The metallic layer partially covering the superconductor provides an additional velocity-dependent damping mechanism for the flux motion that helps to protect the superconducting state when thermomagnetic instabilities develop. If the flux advances with a velocity slower than omega = 2/mu(0)sigma t, where sigma is the cap layer conductivity and t is its thickness, the flux penetration remains unaffected, whereas for incoming flux moving faster than w, the metallic layer becomes an active screening shield. When the metallic layer is replaced by a perfect conductor, it is expected that the flux braking effect will occur for all flux velocities. We investigate this effect by studying Nb samples with a thickness step. Some of the observed features, namely the deflection of the flux trajectories at the border of the thick center, as well as the favored flux penetration at the indentation, are reproduced by time-dependent Ginzburg-Landau simulations.

  • Hybrid plasmonic-semiconducting fractal metamaterials for superior sensing of volatile compounds

    • Localized surface plasmon resonance (LSPR) is a subwavelength optical phenomenon that has found widespread use in bio-and chemical-sensing applications thanks to the possibility to efficiently transduce refractive index changes into wavelength shifts. However, is it very hard to transpose the successes demonstrated in liquid and physiological environment toward the detection of gasous molecules. In fact, the latter typically adsorb in an unspecific manner and induce very minute refractive index changes tipicaly below the sensor sensitivity. Here, we show first insights on the aerosol large-scale self-Assembly of metasurfaces made of monocrystalline Au nanoislands with uniform disorder over large scale. Notably, these architectures show tuneable disorder levels and demonstrate high-quality LSPR, enabling the fabrication of highly performing optical gas sensors detecting down to 10-5 variations in refractive index. Next, we use our aerosol synthesis method to integrate tailored fractals of dielectric TiO2 nanoparticles onto resonant plasmonic metasurfaces. We show how this integration strongly enhances the interaction between the plasmonic field and volatile organic molecules and provides a means for their selective detection. Interesting, the improved performance is the result of a synergetic behavior between the dielectric fractals and the plasmonic metasurface: in fact, upon this integration, the enhancement of plasmonic field is drastically extended, all the way up to a maximum thickness of 1.8 μm. Optimal dielectric-plasmonic structures allow measurements of changes in the refractive index of the gas mixture down to <8x10-6at room temperature and selective identification of three exemplary volatile organic compounds (VOCs). These findings provide a basis for the development of a novel family of hybrid dielectric-plasmonic materials with application extending from light harvesting and photo-catalysts to contactless sensors for non-invasive medical diagnostics.

    • Thirteen novel polymorphic microsatellite loci were isolated and characterized from the Amazonian Black Manakin, Xenopipo atronitens. The loci were screened using 30 samples from Brazil. A total of 13 loci were variable with number of alleles per locus ranging from 2 to 17, whilst the observed and expected heterozygosities varied from 0.067-0.900 and 0.310-0.910, respectively. Five loci deviated significantly from the Hardy-Weinberg equilibrium, whereas one pair of loci deviated significantly from linkage disequilibrium. The set of markers will be a useful tool for future population genetic and kinship studies of X. atronitens, an Amazonian white-sand vegetation specialist.

    • Polyoxometalates (POMs) represent a class of nanomaterials, which hold enormous promise for a range of energy-related applications. Their promise is owing to their "special"structure that gives POMs a truly unique ability to control redox reactions in energy conversion and storage. One such amazing capability is their large number of redox active sites that arises from the complex three-dimensional cluster of metal-oxide ions linked together by oxygen atoms. Here, a critical review on how POMs emerged from being molecular clusters for fundamental studies, to next-generation materials for energy applications is provided. We highlight how exploiting the versatility and activity of these molecules can lead to improved performance in energy devices such as supercapacitors and batteries, and in energy catalyst applications. The potential of POMs across numerous fields is systematically outlined by investigating structure-property-performance relationships and the determinant factors for energy systems. Finally, the challenges and opportunities for this class of materials with respect to addressing our pressing energy-related concerns are identified. This journal is © The Royal Society of Chemistry.
