    • Denna rapport presenterar resultat för mängden av småbiotoper vid åkermark i det svenska landskapet. Analyserna görs på uppdrag av Jordbruksverket, som underlag för bl.a. utvärderingen av miljökvalitetsmålet Ett rikt odlingslandskap. Särskilda rutiner har tagits fram för att aggregera olika variabler för att välja ut de småbiotoper som uppfyller de krav som Jordbruksverket har ställt upp, ur den befintliga databasen. Urvalet av småbiotoper är anpassat för att överensstämma med det urval av objekt som ingår i det s.k. KULT-stödet (miljöersättning till lantbrukare för skötsel av värdefulla natur- och kulturmiljöer) inom Jordbruksverkets Landsbygdsprogram. Arbetet har utförts vid institutionen för skoglig resurshushållning, Sveriges lantbruks-universitet, Umeå. Resultaten baseras på data från flygbildsinventeringen inom det nationella miljöövervakningsprogrammet NILS (Nationell Inventering av Landskapet i Sverige) vilket följer tillstånd och förändringar i det svenska landskapet och hur dessa påverkar förutsättningarna för den biologiska mångfalden. NILS finansieras av Naturvårdsverket, och ingår där i programområde Landskap. Ett viktigt syfte med programmet är att följa upp de nationella miljökvalitetsmålen för olika naturtyper och fungera som underlag för att se om genomförda policybeslut och miljövårdsåtgärder leder till önskade förbättringar

  • Screen printed digital circuits based on vertical organicelectrochemical transistors

    • Vertical organic electrochemical transistors (OECTs) have been manufactured solely using screenprinting. The OECTs are based on PEDOT:PSS (poly(3,4-ethylenedioxythiophene) doped with poly(styrene sulfonic acid)), which defines the active material for both the transistor channel and the gateelectrode. The resulting vertical OECT devices and circuits exhibit low-voltage operation, relativelyfast switching, small footprint and high manufacturing yield; the last three parameters are explainedby the reliance of the transistor configuration on a robust structure in which the electrolyte verticallybridges the bottom channel and the top gate electrode. Two different architectures of the verticalOECT have been manufactured, characterized and evaluated in parallel throughout this report. Inaddition to the experimental work, SPICE models enabling simulations of standalone OECTs andOECT-based circuits have been developed. Our findings may pave the way for fully integrated, lowvoltageoperating and printed signal processing systems integrated with e.g. printed batteries, solarcells, sensors and communication interfaces. Such technology can then serve a low-cost basetechnology for the internet of things, smart packaging and home diagnostics applications.

  • SUPERSMART: ecology and evolution in the era of big data

    • Rapidly growing biological data volumes – including molecular sequences, species traits, geographic occurrences, specimen collections, and fossil records – hold an unprecedented, yet largely unexplored potential to reveal how ecological and evolutionary processes generate and maintain biodiversity. Most biodiversity studies integrating ecological data and evolutionary history use an idiosyncratic step-by-step approach for the reconstruction of time-calibrated phylogenies in light of ecological and evolutionary scenarios. Here we introduce a conceptual framework, termed SUPERSMART (Self-Updating Platform for Estimating Rates of Speciation and Migration, Ages, and Relationships of Taxa), and provide a proof of concept for dealing with the moving targets of biodiversity research. This framework reconstructs dated phylogenies based on the assembly of molecular datasets and collects pertinent data on ecology, distribution, and fossils of the focal clade. The data handled for each step are continuously updated as databases accumulate new records. We exemplify the practice of our method by presenting comprehensive phylogenetic and dating analyses for the orders Primates and the Gentianales. We believe that this emerging framework will provide an invaluable tool for a wide range of hypothesis-driven research questions in ecology and evolution.

  • Multiplexed protein profiling by sequential affinity capture

    • Antibody microarrays enable parallelized and miniaturized analysis of clinical samples, and have proven to provide novel insights for the analysis of different proteomes. However, there are concerns that the performance of such direct labeling and single antibody assays are prone to off-target binding due to the sample context. To improve selectivity and sensitivity while maintaining the possibility to conduct multiplexed protein profiling, we developed a multiplexed and semi-automated sequential capture assay. This novel bead-based procedure encompasses a first antigen capture, labeling of captured protein targets on magnetic particles, combinatorial target elution and a read-out by a secondary capture bead array. We demonstrate in a proof-of-concept setting that target detection via two sequential affinity interactions reduced off-target contribution, while lowered background and noise levels, improved correlation to clinical values compared to single binder assays. We also compared sensitivity levels with single binder and classical sandwich assays, explored the possibility for DNA-based signal amplification, and demonstrate the applicability of the dual capture bead-based antibody microarray for biomarker analysis. Hence, the described concept enhances the possibilities for antibody array assays to be utilized for protein profiling in body fluids and beyond.

    • The Swedish pulp and paper industry's ability to exert monopsony power is tested both with a conjectural variation model, which parameterizes the firms' expectations about other firms' behavior, and with a model that estimates country-specific effects and effects of currency fluctuations on import prices. We use industry data for the 1970-1993 period and find only weak evidence of market power, in spite of the apparent lack of competition.

    • The effects of wood ash application on soil microbial processes were investigated in three drained forested peatlands, which differed in nutrient status and time since application. Measured variables included concentrations of soil elements and phospholipid fatty acids (PLFAs), net nitrogen mineralisation, nitrification and denitrification enzyme activity, potential methane oxidation, methane production and microbial respiration kinetics. Wood ash application had a considerable influence on soil element concentrations. This mirrored a decrease in the majority of the microbial biomarkers by more than one-third in the two oligotrophic peatlands, although microbial community composition was not altered. The decreases in PLFAs coincided with reduced net ammonification and net nitrogen mineralisation. Other measured variables did not change systematically as a result of wood ash application. No significant changes in microbial biomass or processes were found in the mesotrophic peatland, possibly because too little time (1 year) had elapsed since the wood ash application. This study suggests that oligotrophic peatlands can be substantially affected by wood ash for a period of at least four years after application. However, within 25 years of the wood ash application, the microbial biomass seemed to have recovered or adapted to enhanced element concentrations in the soil.

    • Noise is an environmental problem that affects children both at school and home. This chapter summarizes the results on noise disturbances from the “Children’s environment and health survey” conducted in Stockholm County in year 2003. It was found that children were most disturbed by noise at school. Thirty per cent of all 8- and 12 year old children in Stockholm County were annoyed by noise at school, mainly noise from other children. Children in Stockholm County were exposed to more noise in their homes than children in other parts of Sweden. Almost 50 % of children in inner city Stockholm and 20 % in the rest of the County were living in apartments with windows facing a major road, railway or industry. Eighteen per cent of all 8- and 12-year olds were disturbed by noise in their homes. Slightly less than 6 % reports that noise disturbed school homework or speech communication, whereas 4 % reported difficulties in falling asleep due to noise in the home. Approximately three per cent of parents to all 4-, 8- and 12-year old children reported that their children have reduced hearing, and three per cent of 8- and 12-year old children reported that they have tinnitus. It is concluded that noise in dwellings and schools should be reduced in order to provide good and healthy sound environments for children. Furthermore, the sound environment should be considered already at the planning stage, in order to minimize indoor and outdoor noise exposure in future dwellings and schools.

    • Objective This study focuses on the acute effects of vibration and how vibrations influence the measures of the thermal perception thresholds during different vibration magnitudes, frequencies, and durations.Methods The fingers of ten healthy subjects, five males and five females, were exposed to vibration under 16 conditions with a combination of different frequency, intensity and exposure time. The vibration frequency was 31.5 and 125 Hz and exposure lasted between 2 and 16 min. The energy-equivalent frequency weighted acceleration, according to ISO 5349-1, for the experimental time of 16 min was 2.5 or 5.0 m/s(2) (r.m.s.), corresponding to a 8-h equivalent acceleration, A(8) of 0.46 and 0.92 m/s(2), respectively. A measure of the thermal perception of cold and warmth was conducted before the different exposures to vibration. Immediately after the vibration exposure the acute effect was measured continuously on the exposed index finger for the first 75 s, followed by 30 s of measures at every minute for a maximum of 10 min. If the subject's thermal thresholds had not recovered, the measures continued for a maximum of 30 min with measurements taken every 5 min.Results For all experimental conditions and 30 s after exposure, the mean changes of the thresholds compared with the pre-test were found to be 0.05 and -0.67C for the warmth and cold thresholds, respectively. The effect of the vibration exposure was only significant on the cold threshold and only for the first minute after exposure when the threshold was decreased. The warmth threshold was not significantly affected at all. The frequency and the exposure time of the vibration stimuli had no significant influence on the perception thresholds for the sensation of cold or warmth. Increased equivalent frequency weighted acceleration resulted in a significant decrease of the subjects' cold threshold, not the warmth. The thresholds were unaffected when changes in the vibration magnitude were expressed as the frequency weighted acceleration or the unweighted acceleration.Conclusion When testing for the thermotactile thresholds, exposure to vibration on the day of a test might influence the results. Until further knowledge is obtained the previous praxis of 2 h avoidance of vibration exposure before assessment is recommended.
