    • Resin acids are constituents of natural and technical products of widespread use. Exposure is known to cause health effects in the airways and on the skin. Liquid chromatography/positive ion electrospray-mass spectrometry (HPLC/pos ESI-MS) was investigated for determination of 7-oxodehydroabietic (7-OXO), dehydroabietic (DHAA) and abietic acid (AA) in wood dust-containing air samples as a derivatisation-free alternative to the GC/FID HSE method 83/2, developed by the Health and Safety Executive UK. The resin acid 7-OXO was measured as a marker for oxidised resin acids, which are known to be the main contact allergens in colophonium. The found detection limits were 0.42 ng m(-3) for 7-OXO, 5.2 ng m(-3) for DHAA and 9.4 ng m(-3) for AA, respectively, which are considerably lower than with the GC/FID method (24, 115 and 89 ng m(-3)). The two methods correlated well, although consistently and significantly lower concentrations of 7-OXO were detected with LC/MS. The higher concentration of this compound with MDHS 83/2 is suggested to be an artefact from the derivatisation step in the presence of soluble wood dust remains.

  • Resin acids in commercial products and the work environment of Swedish wood pellets production : Analytical methodology, occurrence and exposure

    • The aims of the work this thesis is based upon were to develop convenient analytical procedures for determining resin acids in biological and environmental matrices, and apply them to enhance understanding of the occurrence, exposure to and uptake by exposed individuals of resin acids. Particular focus has been on the workplace environment of the Swedish wood pellets industry. Sample extraction procedures and high-performance liquid chromatography/electrospray ionisation-mass spectrometry (HPLC/ESI-MS) methodologies were developed for measuring resin acids in dust, skin and urine samples. Chromatographic separation of abietic (AA) and pimaric acid was achieved by using a polar-embedded C12 stationary phase. The HPLC/ESI-MS method avoids undesirable oxidation of AA, which was found to occur during the derivatisation step in the standard MDHS 83/2 gas chromatography/flame ionisation detection (GC/FID) methodology, leading to false observations of both AA and the oxidation product 7-oxodehydroabietic acid (7-OXO). Personal exposures to resin acids in the Swedish wood pellet production industry were found to be lower, on average, than the British Occupational Exposure Limit for rosin (50 µg/m3). The oxidised resin acid 7-OXO, was detected in both dust and skin samples indicating the presence of allergenic resin acids. A correlation between air and post-shift urinary concentrations of dehydroabietic acid (DHAA), and a trend towards an increase in urinary 7-OXO during work shifts, were also observed. Whether the increase in 7-OXO was due to direct uptake or metabolism of other resin acids cannot be concluded from the results. An efficient HPLC/UV methodology with diode-array detection was developed for screening commercial products for rosin that could be used in laboratories lacking mass spectrometers. Very high concentrations of free resin acids were detected in depilatory wax strips using the method.

    • Current approaches within affinity-based proteomics are driven both by the accessibility and availability of antigens and capture reagents, and by suitable multiplexed technologies onto which these are implemented. By combining planar microarrays and other multiparallel systems with sets of reagents, possibilities to discover new and unpredicted protein disease associations, either via directed hypothesis-driven or via undirected hypothesis-generating target selection, can be created. In the following stages, the discoveries made during these screening phases have to be verified for potential clinical relevance based on both technical and biological aspects. The use of affinity tools throughout discovery and verification has the potential to streamline the introduction of new markers, as transition into clinically required assay formats appears straightforward. In this article, we summarize some of the current building blocks within array-and affinity-based proteomic profiling with a focus on body fluids, by giving a perspective on how current and upcoming developments in this bioscience could enable a path of pursuit for biomarker discovery.

  • Type 2 diabetes candidate genes, including PAX5, cause impaired insulin secretion in human pancreatic islets

    • Type 2 diabetes (T2D) is caused by insufficient insulin secretion from pancreatic β-cells. To identify candidates contributing to T2D pathophysiology, we studied human pancreatic islets from ~300 individuals. We found 395 differentially expressed genes (DEGs) in islets from individuals with T2D, including, to our knowledge, novel (OPRD1, PAX5, TET1) and previously identified (CHL1, GLRA1, IAPP) candidates. A third of the identified islet expression changes may predispose to diabetes, as they associated with HbA1c in individuals not previously diagnosed with T2D. Most DEGs were expressed in human β-cells based on single-cell RNA-sequencing data. Additionally, DEGs displayed alterations in open chromatin and associated with T2D-SNPs. Mouse knock-out strains demonstrated that T2D-associated candidates regulate glucose homeostasis and body composition in vivo. Functional validation showed that mimicking T2D-associated changes for OPRD1, PAX5, and SLC2A2 impaired insulin secretion. Impairments in Pax5-overexpressing β-cells were due to severe mitochondrial dysfunction. Finally, we discovered PAX5 as a potential transcriptional regulator of many T2D-associated DEGs in human islets. Overall, we identified molecular alterations in human pancreatic islets contributing to β-cell dysfunction in T2D pathophysiology.

    • How can novels and short stories be used in historical research? There seems to be three different approaches to the relationship between fiction and society among Swedish historians today: Fiction as reflection of society, fiction as action that influences society and fiction as dialogue between text and society. The main advantage of using literary sources is that they can contribute to a deeper understanding of social phenomena. This is because novels and short stories are often biased, subjective and fictitious. Although the selection of appropriate novels and short stories for use in historical research raises questions of representativeness, similar methodological problems are part of all historical research. In general, it probably seems more proper to historians to choose realistic fiction that describes the contemporary society of the novelist, but this is not an absolute rule. Historians should not view the research methods employed to analyze fiction as particularly unique. Common historical approaches such as comparisons, argument analysis, discourse analysis of ideas, constructions and representations etc. can be applied to novels and short stories as well as to other types of written sources. In three readings of the same short story, 'Tvifvel' by Anne Charlotte Leffler, we provide three examples, based on different research interests, of how historians can analyze a fictional text. These examples also demonstrate different methodological approaches. One reading takes individual words and their meanings and connotations as a starting point. Another emphasizes dialogue in order to scrutinize the fictional characters' views and positions. A third reading focuses on key episodes in the story. As these readings show, fiction can be valuable in historical research by complicating, refining, and qualifying our understanding of the past.

  • Vad vill vi med historielärarutbildningen?

    • En central del av verksamheten vid nästan alla institutioner som bedriver undervisning och forskning i historia är att utbilda historielärare. Historiker av facket diskuterar emellertid sällan frågor om lärarutbildningens innehåll och uppläggning, utöver de som knyter an till specifika historievetenskapliga sakområden. Vad behöver framtida historielärare kunna, och hur skiljer det sig eventuellt från vad framtida historiker bör kunna? Hur förhåller sig ämnesstudier till ämnesdidaktik och utbildningsvetenskap? Svaren på den typen av frågor har stor betydelse inte bara för lärarutbildningen utan för universitetsämnet historia och dess utövare i stort. Hur ska vi som historiker förhålla oss till det? Som en utgångspunkt för en sådan diskussion har en grupp historiker verksamma inom lärarutbildningen vid Göteborgs och Örebro universitet genomfört en inventering av historiedidaktiken i ämneslärarutbildningen vid tio av totalt sjutton svenska lärosäten där sådan utbildning med inriktning mot historia inom gymnasieskolan bedrivs idag. Med hjälp av publikt tillgänglig information och en enkät via epost till de ansvariga för utbildningarna har gruppen kartlagt historiedidaktiska kursmål och kurslitteratur, examensarbetenas inriktning och vem som undervisar i historiedidaktik. Medan det finns vissa gemensamma drag i hur målen för utbildningarna formuleras, visar genomgången på betydande variationer mellan lärosätena avseende kurslitteratur, examensarbeten och bemanning. Mot den bakgrunden diskuteras i denna idé- och debattartikel vad likheter och skillnader kan bero på samt om, och i så fall hur, det är ett problem att historielärarutbildningarna är så olika.
