    • Background: Histone acetyltransferase enzymes (HATs) are implicated in egulation of transcription. HATs from different families may overlap in arget and substrate specificity. esults: We isolated the elp3(+) gene encoding the histone cetyltransferase subunit of the Elongator complex in fission yeast and haracterized the phenotype of an Delta elp3 mutant. We examined genetic nteractions between Delta elp3 and two other HAT mutants, Delta mst2 nd Delta gcn5 and used whole genome microarray analysis to analyze heir effects on gene expression. onclusions: Comparison of phenotypes and expression profiles in single, ouble and triple mutants indicate that these HAT enzymes have verlapping functions. Consistent with this, overlapping specificity in istone H3 acetylation is observed. However, there is no evidence for verlap with another HAT enzyme, encoded by the essential mst1(+) gene.

  • The bumpy light curve of Type IIn supernova iPTF13z over 3 years

    • A core-collapse (CC) supernova (SN) of Type IIn is dominated by the interaction of SN ejecta with the circumstellar medium (CSM). Some SNe IIn (e.g. SN 2006jd) have episodes of re-brightening (''bumps'') in their light curves. We present iPTF13z, a Type IIn SN discovered on 2013 February 1 by the intermediate Palomar Transient Factory (iPTF). This SN showed at least five bumps in its declining light curve between 130 and 750 days after discovery. We analyse this peculiar behaviour and try to infer the properties of the CSM, of the SN explosion, and the nature of the progenitor star. We obtained multi-band optical photometry for over 1000 days after discovery with the P48 and P60 telescopes at Palomar Observatory. We obtained low-resolution optical spectra during the same period. We did an archival search for progenitor outbursts. We analyse the photometry and the spectra, and compare iPTF13z to other SNe IIn. In particular we derive absolute magnitudes, colours, a pseudo-bolometric light curve, and the velocities of the different components of the spectral lines. A simple analytical model is used to estimate the properties of the CSM. iPTF13z had a light curve peaking at Mr <~ -18.3 mag. The five bumps during its decline phase had amplitudes ranging from 0.4 to 0.9 mag and durations between 20 and 120 days. The most prominent bumps appeared in all the different optical bands, when covered. The spectra of this SN showed typical SN IIn characteristics, with emission lines of Hα (with broad component FWHM ~ 103 - 104 km s-1 and narrow component FWHM ~ 102 km s-1) and He I, but also with Fe II, Ca II, Na I D and Hβ P Cygni profiles (with velocities of ~ 103 km  s-1). A pre-explosion outburst was identified lasting >~ 50 days, with Mr  -15 mag around 210 days before discovery. Large, variable progenitor mass-loss rates (>~ 0.01 MSun yr-1) and CSM densities (>~ 10-16 g cm-3) are derived. The SN was hosted by a metal-poor dwarf galaxy at redshift z = 0.0328. We suggest that the light curve bumps of iPTF13z arose from SN ejecta interacting with denser regions in the CSM, possibly produced by the eruptions of a luminous blue variable progenitor star.

    • SN 2010jl was a luminous Type Lin supernova (SN), detected in radio, optical, X-ray and hard X-rays. Here we report on its six-year R- and g-band light curves obtained using the Palomar Transient Factory. The light curve was generated using a pipeline based on the proper image-subtraction method and we discuss the algorithm performances. As noted before, the R-band light curve, up to about 300 days after maximum light is well described by a power-law decline with a power-law index of alpha approximate to -0.5. Between day 300 and day 2300 after maximum light, it is consistent with a power-law decline, with a power-law index of about alpha approximate to -3.4. The longevity of the light curve suggests that the massive circumstellar material around the progenitor was ejected on timescales of at least tens of years prior to the progenitor explosion.

    • The progenitors of some supernovae (SNe) exhibit outbursts with super-Eddington luminosities prior to their final explosions. This behavior is common among SNe IIn, but the driving mechanisms of these precursors are not yet well-understood. SNHunt 275 was announced as a possible new SN during 2015 May. Here we report on pre-explosion observations of the location of this event by the Palomar Transient Factory (PTF) and report the detection of a precursor about 500 days prior to the 2015 May activity (PTF 13efv). The observed velocities in the 2015 transient and its 2013 precursor absorption spectra are low (1000-2000 km s(-1)), so it is not clear yet if the recent activity indeed marks the final disruption of the progenitor. Regardless of the nature of this event, we use the PTF photometric and spectral observations, as well as Swift-UVOT observations, to constrain the efficiency of the radiated energy relative to the total kinetic energy of the precursor. We find that, using an order-of-magnitude estimate and under the assumption of spherical symmetry, the ratio of the radiated energy to the kinetic energy is in the range of 4 x 10(-2) to 3.4 x 10(3).

  • Near-infrared Supernova Ia Distances : Host Galaxy Extinction and Mass-step Corrections Revisited

    • We present optical and near-infrared (NIR, Y-, J-, H-band) observations of 42 Type Ia supernovae (SNe Ia) discovered by the untargeted intermediate Palomar Transient Factory survey. This new data set covers a broad range of redshifts and host galaxy stellar masses, compared to previous SN Ia efforts in the NIR. We construct a sample, using also literature data at optical and NIR wavelengths, to examine claimed correlations between the host stellar masses and the Hubble diagram residuals. The SN magnitudes are corrected for host galaxy extinction using either a global total-to-selective extinction ratio, R-V = 2.0, for all SNe, or a best-fit R-V for each SN individually. Unlike previous studies that were based on a narrower range in host stellar mass, we do not find evidence for a "mass step," between the color- and stretch-corrected peak J and H magnitudes for galaxies below and above log(M-*/M-circle dot) = 10. However, the mass step remains significant (3 sigma) at optical wavelengths (g, r, i) when using a global R-V, but vanishes when each SN is corrected using their individual best-fit R-V. Our study confirms the benefits of the NIR SN Ia distance estimates, as these are largely exempted from the empirical corrections dominating the systematic uncertainties in the optical.

  • TYPE II SUPERNOVA ENERGETICS AND COMPARISON OF LIGHT CURVES TO SHOCK-COOLING MODELS

    • During the first few days after explosion, Type II supernovae (SNe) are dominated by relatively simple physics. Theoretical predictions regarding early-time SN light curves in the ultraviolet (UV) and optical bands are thus quite robust. We present, for the first time, a sample of 57 R-band SN II light curves that are well-monitored during their rise, with > 5 detections during the first 10 days after discovery, and a well-constrained time of explosion to within 1-3 days. We show that the energy per unit mass (E/M) can be deduced to roughly a factor of five by comparing early-time optical data to the 2011 model of Rabinak & Waxman, while the progenitor radius cannot be determined based on R-band data alone. We find that SN II explosion energies span a range of E/M = (0.2-20) x 10(51) erg/(10 M-circle dot), and have a mean energy per unit mass of < E/M > = 0.85 x 10(51) erg/(10 M-circle dot), corrected for Malmquist bias. Assuming a small spread in progenitor masses, this indicates a large intrinsic diversity in explosion energy. Moreover, E/M is positively correlated with the amount of Ni-56 produced in the explosion, as predicted by some recent models of core-collapse SNe. We further present several empirical correlations. The peak magnitude is correlated with the decline rate (Delta m(15)), the decline rate is weakly correlated with the rise time, and the rise time is not significantly correlated with the peak magnitude. Faster declining SNe are more luminous and have longer rise times. This limits the possible power sources for such events.

    • Phytochromes (PHYs) are photoreceptor proteins first discovered in plants, where they control a variety of photomorphogenesis events. PHYs as photochromic proteins can reversibly switch between two distinct states: a red light (Pr) and a far-red light (Pfr) absorbing form. The discovery of Bacteriophytochromes (BphPs) in nonphotosynthetic bacteria has opened new frontiers in our understanding of the mechanisms by which these natural photoswitches can control single cell development, although the role of BphPs in vivo remains largely unknown. BphPs are dimeric proteins that consist of a photosensory core module (PCM) and an enzymatic domain, often a histidine kinase. The PCM is composed of three domains (PAS, GAF, and PHY). It holds a covalently bound open-chain tetrapyrrole (biliverdin, BV) chromophore. Upon absorption of light, the double bond between BV rings C and D isomerizes and reversibly switches the protein between Pr and Pfr states. We report crystal structures of the wild-type and mutant (His275Thr) forms of the canonical BphP from the nonphotosynthetic myxobacterium Stigmatella aurantiaca (SaBphP2) in the Pr state. Structures were determined at 1.65A degrees and 2.2A degrees (respectively), the highest resolution of any PCM construct to date. We also report the room temperature wild-type structure of the same protein determined at 2.1A degrees at the SPring-8 Angstrom Compact free electron LAser (SACLA), Japan. Our results not only highlight and confirm important amino acids near the chromophore that play a role in Pr-Pfr photoconversion but also describe the signal transduction into the PHY domain which moves across tens of angstroms after the light stimulus.     • BACKGROUND Chronic constipation is a prevalent disorder that affects patients' quality of life and consumes resources in healthcare systems worldwide. In clinical practice, it is still considered a challenge as clinicians frequently are unsure as to which treatments to use and when. Over a decade ago, a Neurogastroenterology & Motility journal supplement devoted to the investigation and management of constipation was published (2009; 21 (Suppl.2)). This included seven articles, disseminating all themes covered during a preceding 2-day meeting held in London, entitled "Current perspectives in chronic constipation: a scientific and clinical symposium." In October 2018, the 3rd London Masterclass, entitled "Contemporary management of constipation" was held, again over 2 days. All faculty members were invited to author two new review articles, which represent a collective synthesis of talks presented and discussions held during this meeting. PURPOSE This article represents the first of these reviews, addressing epidemiology, diagnosis, clinical associations, pathophysiology, and investigation. Clearly, not all aspects of the condition can be covered in adequate detail; hence, there is a focus on particular "hot topics" and themes that are of contemporary interest. The second review addresses management of chronic constipation, covering behavioral, conservative, medical, and surgical therapies.

  • Variable Sodium Absorption in a Low-extinction Type Ia Supernova

    • Recent observations have revealed that some Type Ia supernovae exhibit narrow, time-variable Na I D absorption features. The origin of the absorbing material is controversial, but it may suggest the presence of circumstellar gas in the progenitor system prior to the explosion, with significant implications for the nature of the supernova (SN) progenitors. We present the third detection of such variable absorption, based on six epochs of high-resolution spectroscopy of the Type Ia supernova SN 2007le from the Keck I Telescope and the Hobby-Eberly Telescope. The data span a time frame of approximately three months, from 5 days before maximum light to 90 days after maximum. We find that one component of the Na I D absorption lines strengthened significantly with time, indicating a total column density increase of ~2.5 × 1012 cm-2. The data limit the typical timescale for the variability to be more than 2 days but less than 10 days. The changes appear to be most prominent after maximum light rather than at earlier times when the ultraviolet flux from the SN peaks. As with SN 2006X, we detect no change in the Ca II H and K absorption lines over the same time period, rendering line-of-sight effects improbable and suggesting a circumstellar origin for the absorbing material. Unlike the previous two supernovae exhibiting variable absorption, SN 2007le is not highly reddened (E B - V = 0.27 mag), also pointing toward circumstellar rather than interstellar absorption. Photoionization calculations show that the data are consistent with a dense (107 cm-3) cloud or clouds of gas located ~0.1 pc (3 × 1017 cm) from the explosion. These results broadly support the single-degenerate scenario previously proposed to explain the variable absorption, with mass loss from a nondegenerate companion star responsible for providing the circumstellar gas. We also present possible evidence for narrow Hα emission associated with the SN, which will require deep imaging and spectroscopy at late times to confirm. Some of the data presented herein were obtained at the W. M. Keck Observatory, which is operated as a scientific partnership among the California Institute of Technology, the University of California, and NASA.   • SN 2018fif : The Explosion of a Large Red Supergiant Discovered in Its Infancy by the Zwicky Transient Facility

    • High-cadence transient surveys are able to capture supernovae closer to their first light than ever before. Applying analytical models to such early emission, we can constrain the progenitor stars' properties. In this paper, we present observations of SN 2018fif (ZTF 18abokyfk). The supernova was discovered close to first light and monitored by the Zwicky Transient Facility (ZTF) and the Neil Gehrels Swift Observatory. Early spectroscopic observations suggest that the progenitor of SN 2018fif was surrounded by relatively small amounts of circumstellar material compared to all previous cases. This particularity, coupled with the high-cadence multiple-band coverage, makes it a good candidate to investigate using shock-cooling models. We employ the SOPRANOS code, an implementation of the model by Sapir & Waxman and its extension to early times by Morag et al. Compared with previous implementations, SOPRANOS has the advantage of including a careful account of the limited temporal validity domain of the shock-cooling model as well as allowing usage of the entirety of the early UV data. We find that the progenitor of SN 2018fif was a large red supergiant with a radius of R = 744.0(-128.0)(+183.0) R-circle dot and an ejected mass of M-ej = 9.3(-5.8)(+0.4) M-circle dot. Our model also gives information on the explosion epoch, the progenitor's inner structure, the shock velocity, and the extinction. The distribution of radii is double-peaked, with smaller radii corresponding to lower values of the extinction, earlier recombination times, and a better match to the early UV data. If these correlations persist in future objects, denser spectroscopic monitoring constraining the time of recombination, as well as accurate UV observations (e.g., with ULTRASAT), will help break the extinction/radius degeneracy and independently determine both.
