    • The transcription factor Foxp3 dominantly controls regulatory T (Treg) cell function, and only its continuous expression guarantees the maintenance of full Treg cell-suppressive capacity. However, transcriptional regulators maintaining Foxp3 transcription are incompletely described. Here, we report that high E47 transcription factor activity in Treg cells resulted in unstable Foxp3 expression. Under homeostatic conditions, Treg cells expressed high levels of the E47 antagonist Id3, thus restricting E47 activity and maintaining Foxp3 expression. In contrast, stimulation of Id3-deficient or E47-overexpressing Treg cells resulted in the loss of Foxp3 expression in a subset of Treg cells in vivo and in vitro. Mechanistic analysis indicated that E47 activated expression of the transcription factor Spi-B and the suppressor of cytokine signaling 3 (SOCS3), which both downregulated Foxp3 expression. These findings demonstrate that the balance of Id3 and E47 controls the maintenance of Foxp3 expression in Treg cells and, thus, contributes to Treg cell plasticity.

    • Foxp3(+) regulatory T (Treg) cells are broadly divided into naive-like and activated Treg cells, however recent studies suggest further Treg cell heterogeneity. Treg cells contribute to impaired T cell responses in chronic infections, but the role of specific Treg cell subpopulations in viral infections is not well defined. Here, we report that activated Treg cells are separated into two transcriptionally distinct subpopulations characterized by low or high expression of the transcriptional regulator Id3. Id3(lo) Treg cells are a highly suppressive Treg cell subpopulation, expressing elevated levels of immunomodulatory molecules and are capable of broadly targeting T cell responses. Viral infection and interleukin-2 promote the differentiation of Id3(hi) into Id3(lo) Treg cells and during chronic infection Id3(lo) Treg cells are the predominant Treg cell population. Thus, our report provides a framework, in which different activated Treg cell subpopulations specifically affect immune responses, possibly contributing to T cell dysfunction in chronic infections.

    • South Africa and Russia are the world’s leading platinum group elements (PGE) producers with over 80 % of the global PGE output. Studies performed in the Bushveld Igneous Complex in South Africa and on the Kola Peninsula in Russia show that PGE mining and production activities are important regional PGE sources. Elevated PGE concentrations have been found in snow, soil, road dust, grass, moss and humus collected near PGE mining and production sites. The occurrence of elevated PGE concentrations near mining sites raises concern over environmental effects and exposure of the local population. Studies on the occurrence of PGE in remote environments also suggest that emissions from PGE production activities contribute to the global biogeochemical cycle of the PGE. The loss of PGE during metal production could be as much as 5 % of the global supply and represents a substantial economic loss. Studies on the impact of PGE mining and production activities are few. Further research is needed to better assess the environmental impact of PGE emissions from mining and production activities.

  • Laminin isoforms in atherosclerotic arteries from mice and man.

    • The properties of the arterial vasculature depend to a large extent on the activities of smooth muscle cells, which, in turn, are determined by their extracellular environment. During pathological conditions, such as atherosclerosis, this interaction is altered. In close proximity to medial smooth muscle cells are basement membrane components, such as different isoforms of laminin. These proteins can have great impact on cellular function via interaction with cell surface integrins. However, knowledge of laminins in smooth muscle cell basement membranes during normal and pathological conditions is scarce. Therefore, we have analyzed the presence of laminin isoforms in atherosclerotic lesions of apolipoprotein E (ApoE)-deficient mice. Our study revealed that the laminin chain isotype composition within atherosclerotic plaque tissue was different from the chain composition in the media. In addition, obvious differences in laminin chain composition could be observed in areas of the media, which were or were not associated with plaque tissue. Our major findings demonstrate that laminin gamma3 was exclusively present in media associated with plaque tissue. Laminin alpha2 was also enriched in these medial areas. Plaque tissue was predominantly enriched in laminin alpha5 chains. This general distribution applied to lesions both with and without a fibrous cap-like structure. The differential distribution of laminin chains were partially accompanied by changes in the presence of the integrin alpha subunits 7 and V. The distribution of laminin chains in human atherosclerotic arteries, with different size and morphology, grossly resembled their distribution in mouse arteries.

  • Microstructural study of the NbC to G-phase transformation in HP-Nb alloys

    • The microstructure of a centrifugally cast HP alloy was studied in its as-received state and after ageing at 900 °C. A multi-scale approach combining X Ray Diffraction (XRD), advanced electron microscopy modes (scanning and transmission electron microscopies (SEM, TEM), together with focused ion beam/SEM nanotomography (FIB-nt)) has been carried out to characterize the evolution of niobium carbides during ageing. After thermal treatment, the carbides exhibit a complex microstructure, consisting of a core of untransformed NbC, an intermediate layer of G-phase (Ni16Nb6Si7) with embedded nanometric titanium carbide precipitates, and an outer shell of alternating chromium carbides Cr23C6 and G-phase. A simple diffusion model was used to explain the faster external growth of G-phase compared to the internal NbC dissolution, and to determine a diffusion coefficient of niobium in the G-phase at 900 °C.
