    • Plastics debris in the marine environment, including resin pellets, fragments and microscopic plastic fragments, contain organic contaminants, including polychlorinated biphenyls ( PCBs), polycyclic aromatic hydrocarbons, petroleum hydrocarbons, organochlorine pesticides (2,2-bis(p-chlorophenyl)-1,1,1-trichloroethane, hexachlorinated hexanes), polybrominated diphenylethers, alkylphenols and bisphenol A, at concentrations from sub ng g(-1) to mg g(-1). Some of these compounds are added during plastics manufacture, while others adsorb from the surrounding seawater. Concentrations of hydrophobic contaminants adsorbed on plastics showed distinct spatial variations reflecting global pollution patterns. Model calculations and experimental observations consistently show that polyethylene accumulates more organic contaminants than other plastics such as polypropylene and polyvinyl chloride. Both a mathematical model using equilibrium partitioning and experimental data have demonstrated the transfer of contaminants from plastic to organisms. A feeding experiment indicated that PCBs could transfer from contaminated plastics to streaked shearwater chicks. Plasticizers, other plastics additives and constitutional monomers also present potential threats in terrestrial environments because they can leach from waste disposal sites into groundwater and/or surface waters. Leaching and degradation of plasticizers and polymers are complex phenomena dependent on environmental conditions in the landfill and the chemical properties of each additive. Bisphenol A concentrations in leachates from municipal waste disposal sites in tropical Asia ranged from sub mu g l(-1) to mg l(-1) and were correlated with the level of economic development.

  • The effect of altering spherical aberration on the static accommodative response.

    • PURPOSE: To investigate the effect of altering the spherical aberration (SA) of the eye on the static accommodative response.METHODS: Participants were fitted with nominally afocal contact lenses with controlled amounts of SA of either -0.2, -0.1, 0.0, +0.1 or +0.2 microm for a 5-mm pupil. Measurements of SA and root mean square (RMS) total aberration for the eye plus lens for each participant were determined with a Complete Ophthalmic Analysis System aberrometer. Accommodation was stimulated either by placing targets at different dioptric distances from the eye, or by using a fixed distance target and placing negative-powered lenses in front of the eye. Accommodation responses were determined with a Shin-Nippon autorefractor.RESULTS: For both stimuli situations, the slope of the accommodation stimulus-response function was lowest for the lenses with +0.2 microm SA, and increased as the amount of SA was reduced. There was a significant negative correlation between SA and slope. Lag of accommodation at 33 cm correlated well with added SA, but did not correlate with total RMS error. There was no significant difference between the responses at 30 min after lens wear started and the responses after 1 h.CONCLUSIONS: Adding negative SA to the eye generally improves the slope of the accommodation stimulus-response curve and decreases lag of accommodation, and positive added SA depresses the slope of the stimulus-response curve and increases lag. The effect seems to be specific to SA, as there was no relationship between lag and RMS error. Altering SA may be a viable way of changing accommodative functions in clinical situations.

  • DISSOCIATIVE RECOMBINATION OF VIBRATIONALLY COLD CH+3 AND INTERSTELLAR IMPLICATIONS

    • CH3+ is an important molecular ion in the astrochemistry of diffuse clouds, dense clouds, cometary comae, and planetary ionospheres. However, the rate of one of the major destruction mechanisms of CH3+, dissociative recombination (DR), has long been uncertain, hindering the use of CH3+ as an astrochemical probe. Here, we present the first absolute measurement of the DR of vibrationally cold CH3+, which has been made using the heavy storage ring CRYRING in Stockholm, Sweden. From our collision-energy-dependent cross sections, we infer a thermal rate constant of k(T) = 6.97(+/- 0.03) x 10(-7)(T/300)(-0.61(+/- 0.01)) cm(3) s(-1) over the region 10 K <= T <= 1000 K. At low collision energies, we have measured the branching fractions of the DR products to be CH3 (0.00(- 0.00)(+ 0.01)), CH2 + H (0.35(-0.01)(+ 0.01)), CH + 2H (0.20(-0.02)(+0.02)), CH + H-2 (0.10(-0.01)(+0.01)), and C + H-2 + H (0.35(-0.02)(+ 0.01)), indicating that two or more C-H bonds are broken in 65% of all collisions. We also present vibrational calculations which indicate that the CH3+ ions in the storage ring were relaxed to the vibrational ground state by spontaneous emission during the storage time. Finally, we discuss the implications of these new measurements for the observation of CH3+ in regions of the diffuse interstellar medium where CH+ is abundant.

  • Tipping elements and amplified polar warming during the Last Interglacial

    • Irreversible shifts of large-scale components of the Earth system (so-called ‘tipping elements’) on policy-relevant timescales are a major source of uncertainty for projecting the impacts of future climate change. The high latitudes are particularly vulnerable to positive feedbacks that amplify change through atmosphere-ocean-ice interactions. Unfortunately, the short instrumental record does not capture the full range of past or projected climate scenarios (a situation particularly acute in the high latitudes). Natural archives from past periods warmer than present day, however, can be used to explore drivers and responses to forcing, and provide data against which to test models, thereby offering insights into the future. The Last Interglacial (129–116,000 years before present) — the warmest interglacial of the last 800,000 years — was the most recent period during which global temperatures were comparable with low-end 21st Century projections (up to 2 °C warmer, with temperature increase amplified over polar latitudes), providing a potentially useful analogue for future change. Substantial environmental changes happened during this time. Here we synthesise the nature and timing of potential high-latitude tipping elements during the Last Interglacial, including sea ice, extent of the boreal forest, permafrost, ocean circulation, and ice sheets/sea level. We also review the thresholds and feedbacks that likely operated through this period. Notably, substantial ice mass loss from Greenland, the West Antarctic, and possibly sectors of the East Antarctic drove a 6–9 m rise in global sea level. This was accompanied by reduced summer sea-ice extent, poleward-extended boreal forest, and reduced areas of permafrost. Despite current chronological uncertainties, we find that tipping elements in the high latitudes all experienced rapid and abrupt change (within 1–2 millennia of each other) across both hemispheres, while recovery to prior conditions took place over multi-millennia. Our synthesis demonstrates important feedback loops between tipping elements, amplifying polar and global change during the Last Interglacial. The high sensitivity and tight interconnections between polar tipping elements suggests that they could exhibit similar thresholds of vulnerability in the future, particularly if the aspirations of the Paris Agreement are not met.

  • HARS3 for a Roboticized Isaac Newton Telescope

    • We present a description of a new instrument development, HARPS3, planned to be installed on an upgraded and roboticized Isaac Newton Telescope by end-2018. HARPS3 will be a high resolution (R similar or equal to 115,000) echelle spectrograph with a wavelength range from 380-690 nm. It is being built as part of the Terra Hunting Experiment {a future 10 year radial velocity measurement programme to discover Earth-like exoplanets. The instrument design is based on the successful HARPS spectrograph on the 3.6m ESO telescope and HARPS-N on the TNG telescope. The main changes to the design in HARPS3 will be: a customised fibre adapter at the Cassegrain focus providing a stabilised beam feed and on-sky fibre diameter approximate to 1.4 arcsec, the implementation of a new continuous flow cryostat to keep the CCD temperature very stable, detailed characterisation of the HARPS3 CCD to map the effective pixel positions and thus provide an improved accuracy wavelength solution, an optimised integrated polarimeter and the instrument integrated into a robotic operation. The robotic operation will optimise our programme which requires our target stars to be measured on a nightly basis. We present an overview of the entire project, including a description of our anticipated robotic operation.

  • Non-genotoxic carcinogen exposure induces defined changes in the 5-hydroxymethylome

    • BACKGROUND: Induction and promotion of liver cancer by exposure to non-genotoxic carcinogens coincides with epigenetic perturbations, including specific changes in DNA methylation. Here we investigate the genome-wide dynamics of 5-hydroxymethylcytosine (5hmC) as a likely intermediate of 5-methylcytosine (5mC) demethylation in a DNA methylation reprogramming pathway. We use a rodent model of non-genotoxic carcinogen exposure using the drug phenobarbital.RESULTS:Exposure to phenobarbital results in dynamic and reciprocal changes to the 5mC/5hmC patterns over the promoter regions of a cohort of genes that are transcriptionally upregulated. This reprogramming of 5mC/5hmC coincides with characteristic changes in the histone marks H3K4me2, H3K27me3 and H3K36me3. Quantitative analysis of phenobarbital-induced genes that are involved in xenobiotic metabolism reveals that both DNA modifications are lost at the transcription start site, while there is a reciprocal relationship between increasing levels of 5hmC and loss of 5mC at regions immediately adjacent to core promoters.CONCLUSIONS:Collectively, these experiments support the hypothesis that 5hmC is a potential intermediate in a demethylation pathway and reveal precise perturbations of the mouse liver DNA methylome and hydroxymethylome upon exposure to a rodent hepatocarcinogen.
