  • AIF Overexpression Aggravates Oxidative Stress in Neonatal Male Mice After Hypoxia-Ischemia Injury

    • There are sex differences in the severity, mechanisms, and outcomes of neonatal hypoxia-ischemia (HI) brain injury, and apoptosis-inducing factor (AIF) may play a critical role in this discrepancy. Based on previous findings that AIF over-expression aggravates neonatal HI brain injury, we further investigated potential sex differences in the severity and molecular mechanisms underlying the injury using mice that overexpress AIF from homozygous transgenes. We found that the male sex significantly aggravated AIF-driven brain damage, as indicated by the injury volume in the gray matter (2.25 times greater in males) and by the lost volume of subcortical white matter (1.71 greater in males) after HI. As compared to females, male mice exhibited more severe brain injury, correlating with reduced antioxidant capacities, more pronounced protein carbonylation and nitration, and increased neuronal cell death. Under physiological conditions (without HI), the doublecortin-positive area in the dentate gyrus of females was 1.15 times larger than in males, indicating that AIF upregulation effectively promoted neurogenesis in females in the long term. We also found that AIF stimulated carbohydrate metabolism in young males. Altogether, these findings corroborate earlier studies and further demonstrate that AIF is involved in oxidative stress, which contributes to the sex-specific differences observed in neonatal HI brain injury.

  • Coherent dynamics of multi-spin V-B(-) center in hexagonal boron nitride

    • Hexagonal boron nitride (hBN) has recently been demonstrated to contain optically polarized and detected electron spins that can be utilized for implementing qubits and quantum sensors in nanolayered-devices. Understanding the coherent dynamics ofmicrowave driven spins in hBN is of crucial importance for advancing these emerging new technologies. Here, we demonstrate and study the Rabi oscillation and related phenomena of a negatively charged boron vacancy (V-B(-)) spin ensemble in hBN. We report on different dynamics of the V-B(-) spins at weak and strong magnetic fields. In the former case the defect behaves like a single electron spin system, while in the latter case it behaves like a multi-spin system exhibiting multiple-frequency dynamical oscillation as beat in the Ramsey fringes. We also carry out theoretical simulations for the spin dynamics of V-B(-) and reveal that the nuclear spins can be driven via the strong electron nuclear coupling existing in V-B(-) center, which can be modulated by the magnetic field and microwave field.

  • Exploring the cellulolytic and hemicellulolytic activities of manganese peroxidase for lignocellulose deconstruction

    • BackgroundA cost-effective pretreatment and saccharification process is a necessary prerequisite for utilizing lignocellulosic biomass (LCB) in biofuel and biomaterials production. Utilizing a multifunctional enzyme with both pretreatment and saccharification functions in a single step for simultaneous biological pretreatment and saccharification process (SPS) will be a green method of low cost and high efficiency. Manganese peroxidase (MnP, EC 1.11.1.13), a well-known lignin-degrading peroxidase, is generally preferred for the biological pretreatment of biomass. However, exploring the role and performance of MnP in LCB conversion will promote the application of MnP for lignocellulose-based biorefineries.ResultsIn this study, we explored the ability of an MnP from Moniliophthora roreri, MrMnP, in LCB degradation. With Mn2+ and H2O2, MrMnP decomposed 5.0 g/L carboxymethyl cellulose to 0.14 mM of reducing sugar with a conversion yield of 5.0 mg/g, including 40 μM cellobiose, 70 μM cellotriose, 20 μM cellotetraose, and 10 μM cellohexaose, and degraded 1.0 g/L mannohexaose to 0.33 μM mannose, 4.08 μM mannotriose, and 4.35 μM mannopentaose. Meanwhile, MrMnP decomposed 5.0 g/L lichenan to 0.85 mM of reducing sugar with a conversion yield of 30.6 mg/g, including 10 μM cellotriose, 20 μM cellotetraose, and 80 μM cellohexose independently of Mn2+ and H2O2. Moreover, the versatility of MrMnP in LCB deconstruction was further verified by decomposing locust bean gum and wheat bran into reducing sugars with a conversion yield of 54.4 mg/g and 29.5 mg/g, respectively, including oligosaccharides such as di- and tri-saccharides. The catalytic mechanism underlying MrMnP degraded lignocellulose was proposed as that with H2O2, MrMnP oxidizes Mn2+ to Mn3+. Subsequently, it forms a complex with malonate, facilitating the degradation of CMC and mannohexaose into reducing sugars. Without H2O2, MrMnP directly oxidizes malonate to hydroperoxyl acetic acid radical to form compound I, which then attacks the glucosidic bond of lichenan.ConclusionThis study identified a new function of MrMnP in the hydrolysis of cellulose and hemicellulose, suggesting that MrMnP exhibits its versatility in the pretreatment and saccharification of LCB. The results will lead to an in-depth understanding of biocatalytic saccharification and contribute to forming new enzymatic systems for using lignocellulose resources to produce sustainable and economically viable products and the long-term development of biorefinery, thereby increasing the productivity of LCB as a green resource.

  • Tunable Magnetism in Atomically Thin Itinerant Antiferromagnet with Room-Temperature Ferromagnetic Order

    • Addressing the need for modulated spin configurations is crucial, as they serve as the foundational building blocks for next-generation spintronics, particularly in atomically thin structures and at room temperature. In this work, we realize intrinsic ferromagnetism in monolayer flakes and tunable ferro-/antiferromagnetism in (Fe0.56Co0.44)5GeTe2 antiferromagnets. Remarkably, the ferromagnetic ordering (≥1 L) and antiferromagnetic ordering (≥4 L) remain discernible up to room temperature. The TC (∼310 K) of the monolayer flakes sets a record high for known exfoliated monolayer van der Waals magnets. Within the framework of A-type antiferromagnetism, a notable odd-even layer-number effect at elevated temperatures (T = 150 K) is observed. Of particular interest is the strong ferromagnetic order in even-layer flakes at low temperatures. The intricate interplay among magnetic field strength, layer number, and temperature gives rise to a diverse array of phenomena, holding promise not only for new physics but also for practical applications.

    • © 2020 The Authors The interaction between apoptosis-inducing factor (AIF) and cyclophilin A (CypA) has been shown to contribute to caspase-independent apoptosis. Blocking the AIF/CypA interaction protects against glutamate-induced neuronal cell death in vitro, and the purpose of this study was to determine the in vivo effect of an AIF/CypA interaction blocking peptide (AIF(370-394)-TAT) on neonatal mouse brain injury after hypoxia-ischemia (HI). The pups were treated with AIF (370-394)-TAT peptide intranasally prior to HI. Brain injury was significantly reduced at 72 h after HI in the AIF(370-394)-TAT peptide treatment group compared to vehicle-only treatment for both the gray matter and the subcortical white matter, and the neuroprotection was more pronounced in males than in females. Neuronal cell death was evaluated in males at 8 h and 24 h post-HI, and it was decreased significantly in the CA1 region of the hippocampus and the nucleus habenularis region after AIF(370-394)-TAT treatment. Caspase-independent apoptosis was decreased in the cortex, striatum, and nucleus habenularis after AIF(370-394)-TAT treatment, but no significant change was found on caspase-dependent apoptosis as indicated by the number of active caspase-3-labeled cells. Further analysis showed that both AIF and CypA nuclear accumulation were decreased after treatment with the AIF(370-394)-TAT peptide. These results suggest that AIF(370-394)-TAT inhibited AIF/CypA translocation to the nucleus and reduced HI-induced caspase-independent apoptosis and brain injury in young male mice, suggesting that blocking AIF/CypA might be a potential therapeutic target for neonatal brain injury.

  • A Nitroxide Radical Conjugated Polymer as an Additive to Reduce Nonradiative Energy Loss in Organic Solar Cells

    • Nonfullerene-acceptor-based organic solar cells (NFA-OSCs) are now set off to the 20% power conversion efficiency milestone. To achieve this, minimizing all loss channels, including nonradiative photovoltage losses, seems a necessity. Nonradiative recombination, to a great extent, is known to be an inherent material property due to vibrationally induced decay of charge-transfer (CT) states or their back electron transfer to the triplet excitons. Herein, it is shown that the use of a new conjugated nitroxide radical polymer with 2,2,6,6-tetramethyl piperidine-1-oxyl side groups (GDTA) as an additive results in an improvement of the photovoltaic performance of NFA-OSCs based on different active layer materials. Upon the addition of GDTA, the open-circuit voltage (VOC), fill factor (FF), and short-circuit current density (JSC) improve simultaneously. This approach is applied to several material systems including state-of-the-art donor/acceptor pairs showing improvement from 15.8% to 17.6% (in the case of PM6:Y6) and from 17.5% to 18.3% (for PM6:BTP-eC9). Then, the possible reasons behind the observed improvements are discussed. The results point toward the suppression of the CT state to triplet excitons loss channel. This work presents a facile, promising, and generic approach to further improve the performance of NFA-OSCs.

    • Crop pests and diseases (CPDs) are emerging threats to global food security, but trends in the occurrence of pests and diseases remain largely unknown due to the lack of observations for major crop producers. Here, on the basis of a unique historical dataset with more than 5,500 statistical records, we found an increased occurrence of CPDs in every province of China, with the national average rate of CPD occurrence increasing by a factor of four (from 53% to 218%) during 1970–2016. Historical climate change is responsible for more than one-fifth of the observed increment of CPD occurrence (22% ± 17%), ranging from 2% to 79% in different provinces. Among the climatic factors considered, warmer nighttime temperatures contribute most to the increasing occurrence of CPDs (11% ± 9%). Projections of future CPDs show that at the end of this century, climate change will lead to an increase in CPD occurrence by 243% ± 110% under a low-emissions scenario (SSP126) and 460% ± 213% under a high-emissions scenario (SSP585), with the magnitude largely dependent on the impacts of warmer nighttime temperatures and decreasing frost days. This observation-based evidence highlights the urgent need to accurately account for the increasing risk of CPDs in mitigating the impacts of climate change on food production.

    • Feasibility of Ti3AlC2 phase as the protective coatings of accident tolerant fuels (ATFs) was investigated by means of ions irradiation, nanoindentation and transmission electron microscopy. Au ions irradiation was carried out on thin Ti3AlC2 film to simulate the high displacement damage induced by the energetic particles in the nuclear reactors. Nanoindentation on the Ti3AlC2 film was followed subsequently as a source of external stress to simulate the high pressure applied on the cladding in nuclear reactor cores of pressurized water reactors (PWRs). TEM was used to characterize the microstructural evolution of Ti3AlC2 film after irradiation and nanoindentation. TEM analysis shows that Ti3AlC2 film remains pristine layered structure and no amorphization was detected after irradiation to similar to 14 dpa. The combined nanoindentation and TEM show that no rupture and exfoliation of the Au-irradiated Ti3AlC2 film occur even the extern stress and total elongation induced by nanoindentation reach to 16.6 GPa and similar to 5%, respectively. The above results show good irradiation resistance and good ductility as well as excellent adhesion of the Ti3AlC2 coating on the substrate after high dose irradiation and under high external stress. This indicates the good feasibility of Ti3AlC2 thin films as the coatings of ATF claddings. (C) 2018 Elsevier B.V. All rights reserved.

  • Adsorption of phenanthrene on Al (oxy) hydroxides formed under the influence of tannic acid

    • Although there has been substantial research done on adsorption of metals/metalloids by Al (oxy)hydroxides, little is known regarding the adsorption of polyaromatic hydrocarbons (PAHs) on Al (oxy)hydroxides, especially those formed in the presence of organic acid. This paper investigated the adsorption of phenanthrene on Al (oxy)hydroxides formed with initial tannate/Al molar ratios (MRs) 0, 0.001, 0.01, and 0.1 (referred to MR0, MR0.001, MR0.01, and MR0.1, respectively) through batch adsorption experiments and FTIR study. The results showed that Al (oxy)hydroxides were important sorbents for phenanthrene. The adsorption kinetic data were fitted well with the pseudo-second-order equation. According to a modified Freundlich model, the adsorption capacities of Al (oxy)hydroxides followed a descending order of MR0.1 > MR0 ≥ MR0.01 > MR0.001, which was inconsistent with the organic carbon content in the Al (oxy)hydroxides. Adsorption capacity correlated with the specific surface area, micropore area, and micropore diameter of Al (oxy)hydroxides, yet the relationships were not statistically significant (P > 0.05). FTIR results showed that physical interaction was essential in phenanthrene adsorption onto the Al (oxy)hydroxides, which could be explained by an entropy-driven process. Surface hydrophobicity of Al (oxy)hydroxides played a key role in phenanthrene adsorption. Additional π–π electron donor–acceptor interaction of phenanthrene (acting as electron donor) with aromatic ring of tannic acid (electron acceptor) could be also important in phenanthrene adsorption by high MR Al (oxy)hydroxides, yet it needs further study. The findings obtained in the present study are of fundamental significance in understanding the mechanism of immobilization of PAHs in low organic matter but oxide-rich soils.
