  • Variations of gastric corpus microbiota are associated with early esophageal squamous cell carcinoma and squamous dysplasia

    • Observational studies revealed a relationship between changes in gastric mucosa and risk of esophageal squamous cell carcinoma (ESCC) which suggested a possible role for gastric microbiota in ESCC carcinogenesis. In this study we aimed to compare pattern of gastric corpus microbiota in ESCC with normal esophagus. Cases were included subjects with early ESCC (stage I–II) and esophageal squamous dysplasia (ESD) as the cancer precursor. Control groups included age and sex-matched subjects with mid-esophagus esophagitis (diseased-control) and histologically normal esophagus (healthy-control). DNA was extracted from snap-frozen gastric corpus tissues and 16S rRNA was sequenced on GS-FLX Titanium. After noise removal, an average of 3004 reads per sample was obtained from 93 subjects. We applied principal coordinate analysis to ordinate distances from beta diversity data. Pattern of gastric microbiota using Unifrac (p = 0.004) and weighted Unifrac distances (p = 0.018) statistically varied between cases and healthy controls. Sequences were aligned to SILVA database and Clostridiales and Erysipelotrichales orders were more abundant among cases after controling for multiple testing (p = 0.011). No such difference was observed between mid-esophagitis and healthy controls. This study is the first to show that composition of gastric corpus mucosal microbiota differs in early ESCC and ESD from healthy esophagus.

  • Poultry litter ash characterisation and recovery

    • This paper reports a complete characterisation of poultry litter ash and its potential use as a heavy metal stabiliser. We propose a novel approach, in which the ashes deriving from municipal solid waste incineration (MSWI) are combined with poultry litter ash, rather than with coal combustion flue gas desulfurisation (FGD) residues. Heavy metals stabilisation was demonstrated by comparing the elemental concentrations in the leaching solutions of the starting raw and stabilised materials: leachable Pb and Zn showed a reduced solubility. The characterisation was conducted by total reflection X-ray fluorescence (TXRF), X-ray diffraction (XRD), micro-Raman spectroscopy and scanning electron microscopy combined with energy-dispersive X-ray spectrometry (SEM-EDX). The results showed that the poultry litter ash was Ca-, P-, K- and S-rich (>29 g/kg). It contained amorphous materials (i.e. fly ash economiser (FAECO) 73% and fly ash cyclone (FACYC) 61%) and soluble phases (e.g. arkanite and sylvite; up to 13% FAECO and 28% FACYC), as well as resilient crystalline (up to 2% of FAECO and FACYC) and amorphous phases (e.g. hydroxyapatite). After two months, the Pb and Zn concentrations in the leachate solutions were below the limit set by the European regulations for waste disposal (<0.2 mg/L and 1.5 mg/L, respectively). We propose a mechanism for the heavy metals stabilisation based on the carbonation process and high amounts of P, Ca and reactive amorphous phases. In conclusion, it is demonstrated that poultry litter ash can be an effective secondary source of heavy metals, allowing their immobilisation through P- and Ca-based reactive amorphous phases.

  • A novel method for implementation of corrosion-induced cracks in the finite element models of reinforced concrete structures

    • Currently, there is a clear need for reliable procedures for condition assessment and service-life evaluation of existing infrastructures. Advanced 3D Nonlinear Finite Element (3D NLFE) analysis has proven to be capable of describing the behavior of reinforced concrete provided that detailed and appropriate condition assessment data are available. The present study aims to review and compare different procedures for coupling 3D NLFE analysis with condition assessment data to model corrosion induced cracking, and, consequently to find a method with better accuracy, less computational cost, and improved robustness. This paper introduces a new method for adding cracked elements directly to the finite element model, called Re-FEM. The force-displacement response, ultimate crack pattern, and failure mode from this model are compared with four other methods for a pull out test case on specimens under an accelerated corrosion process. Using this method, the force displacement response for the corroded specimens was overestimated by about 70%. However, the trend was promising and the failure mode and crack pattern were correct. Moreover, the analysis continued after the peak point in the force-displacement curve, which makes it possible to monitor the behavior of the specimen in the softening regime.
