    • Results of the EU ArcRisk project on human health impacts in the Arctic owing to climate-induced changes in contaminant cycling are summarized in the context of their policy application. The question on how will climate change affect the transport of selected persistent organic pollutants (POPS) and mercury, both to and within the Arctic has been addressed, as well as the issue of human health impacts of these pollutants in the Arctic in relation to exposed local populations. It was concluded that better characterization of primary and secondary sources of POPs and more accurate quantification of current and future releases of POPs from these sources are needed for better prediction of environmental exposure to these contaminants and interpretation of monitoring data. Further improvement of fate and transport modeling in the physical environment is necessary in order to consider in the models not only the relatively well studied direct effects of climate change (e.g., changes in temperature, ice and snow cover, precipitation, wind speed and ocean currents) on contaminants fate and behavior but also indirect effects, e.g., alterations in carbon cycling, catchment hydrology, land use, vegetation cover, etc. Long-term environmental monitoring of POPs (at multiple sampling stations within and outside the Arctic and at regular sampling intervals facilitates temporal trend analysis) and measurements of concentrations in human milk and blood plasma are needed. Finally, more information should be gathered on the human health effects of newly identified POPs, such as perfluorooctane-sulfonic acid (PFOS), polybrominated diphenyl ethers (PBDEs), hexabromocyclododecanes (HBCDs), and other substances with POP-like characteristics, particularly the effects on very young (including fetus) and elderly subgroups of the human population. The Arc Risk developed methodologies and tools that can be used in further studies to resolve various uncertainties already defined in the analysis of climate change impacts on POPs and mercury behavior and effects in the Arctic. The ArcRisk project has also developed very valuable databases that can be regarded as a starting point in further studies.

    • Case volume per 100 000 population and perioperative mortality rate (POMR) are key indicators to monitor and strengthen surgical services. However, comparisons of POMR have been restricted by absence of standardised approaches to when it is measured, the ideal denominator, need for risk adjustment, and whether data are available. We aimed to address these issues and recommend a minimum dataset by analysing four large mixed surgical datasets, two from well-resourced settings with sophisticated electronic patient information systems and two from resource-limited settings where clinicians maintain locally developed databases.

    • The present study investigated the compounds present in the low molecular mass fraction of Lentinus edodes mushroom (shiitake) extract and their anti-virulence activity against oral pathogens (reference and clinical Streptococcus mutans, Actinomyces naeslundii, and Prevotella intermedia strains). Oxalic, succinic, and quinic acids, and adenine, inosine, and uridine were identified by HPLC-DAD-ESI-MS/MS. Their anti-biofilm production and preformed biofilm disaggregation activities were studied using commercial standard compounds at different concentrations. As regards S. mutans, the highest activity was shown by adenine at 5 mg mL(-1) both in the biofilm inhibition (BI 50%) and biofilm disaggregation tests (BD 20%). Considering A. naeslundii, BI values close to 80% were registered for oxalic acid at 1 mg mL(-1) and 2 mg mL(-1) and BD 50% for quinic acid at 3 mg mL(-1). A weaker activity was found against P. intermedia. Furthermore, different mixtures of the commercial standards were tested showing that the activity of a compound can be strongly and sometimes negatively affected by the presence of the other compounds.

  • Four-dimensional distribution of the 2010 Eyjafjallajokull volcanic cloud over Europe observed by EARLINET

    • The eruption of the Icelandic volcano Eyjafjallajokull in April-May 2010 represents a natural experiment to study the impact of volcanic emissions on a continental scale. For the first time, quantitative data about the presence, altitude, and layering of the volcanic cloud, in conjunction with optical information, are available for most parts of Europe derived from the observations by the European Aerosol Research Lidar NETwork (EARLINET). Based on multi-wavelength Raman lidar systems, EARLINET is the only instrument worldwide that is able to provide dense time series of high-quality optical data to be used for aerosol typing and for the retrieval of particle microphysical properties as a function of altitude. In this work we show the four-dimensional (4-D) distribution of the Eyjafjallajokull volcanic cloud in the troposphere over Europe as observed by EARLINET during the entire volcanic event (15 April-26 May 2010). All optical properties directly measured (backscatter, extinction, and particle linear depolarization ratio) are stored in the EARLINET database available at www.earlinet.org. A specific relational database providing the volcanic mask over Europe, realized ad hoc for this specific event, has been developed and is available on request at www.earlinet.org. During the first days after the eruption, volcanic particles were detected over Central Europe within a wide range of altitudes, from the upper troposphere down to the local planetary boundary layer (PBL). After 19 April 2010, volcanic particles were detected over southern and south-eastern Europe. During the first half of May (5-15 May), material emitted by the Eyjafjallajokull volcano was detected over Spain and Portugal and then over the Mediterranean and the Balkans. The last observations of the event were recorded until 25 May in Central Europe and in the Eastern Mediterranean area. The 4-D distribution of volcanic aerosol layering and optical properties on European scale reported here provides an unprecedented data set for evaluating satellite data and aerosol dispersion models for this kind of volcanic events.

  • Association of maternal prenatal smoking GFI1-locus and cardiometabolic phenotypes in 18,212 adults

    • Background: DNA methylation at the GFI1-locus has been repeatedly associated with exposure to smoking from the foetal period onwards. We explored whether DNA methylation may be a mechanism that links exposure to maternal prenatal smoking with offspring's adult cardio-metabolic health. Methods: We meta-analysed the association between DNA methylation at GFI1-locus with maternal prenatal smoking, adult own smoking, and cardio-metabolic phenotypes in 22 population-based studies from Europe, Australia, and USA (n= 18,212). DNA methylation at the GFI1-locus was measured in whole-blood. Multivariable regression models were fitted to examine its association with exposure to prenatal and own adult smoking. DNA methylation levels were analysed in relation to body mass index (BMI), waist circumference (WC), fasting glucose (FG), high-density lipoprotein cholesterol (HDL-C), triglycerides (TG), diastolic, and systolic blood pressure (BP). Findings: Lower DNA methylation at three out of eight GFI1-CpGs was associated with exposure to maternal prenatal smoking, whereas, all eight CpGs were associated with adult own smoking. Lower DNA methylation at cg14179389, the strongest maternal prenatal smoking locus, was associated with increased WC and BP when adjusted for sex, age, and adult smoking with Bonferroni-corrected P < 0.012. In contrast, lower DNA methylation at cg09935388, the strongest adult own smoking locus, was associated with decreased BMI, WC, and BP (adjusted 1 x 10(-7) < P < 0.01). Similarly, lower DNA methylation at cg12876356, cg18316974, cg09662411, and cg18146737 was associated with decreased BMI and WC (5 x 10(-8) < P < 0.001). Lower DNA methylation at all the CpGs was consistently associated with higher TG levels. Interpretation: Epigenetic changes at the GFI1 were linked to smoking exposure in-utero/in-adulthood and robustly associated with cardio-metabolic risk factors.   • First evidence for smooth band termination in valence space in the mass 130 region : Spectroscopy of 127La

    • High spin states in 127La have been studied using the 32S(100Mo,p4n) reaction at a beam energy of 155 MeV. Gamma rays were detected using the EUROBALL III spectrometer. One of the side bands in 127La is observed to be populated to a spin of 83/2+. These data are compared with cranked Nilsson-Strutinski calculations which suggest that the structure is a smoothly terminating band, in valence space, based upon a π[(g7/2d5/2)5(h11/2)2]⊗ ν[(g7/2d5/2)12(h11/2)6(d3/2s1/2)2] configuration, which can carry a maximum spin of 47.5ℏ. This is the first identification of such a structure in this mass region.

    • Background Serum creatinine (SCR) is the most important biomarker for a quick and non-invasive assessment of kidney function in population-based surveys. A substantial proportion of the inter-individual variability in SCR level is explicable by genetic factors. Methods We performed a meta-analysis of genome-wide association studies of SCR undertaken in five population isolates ('discovery cohorts'), all of which are part of the European Special Population Network (EUROSPAN) project. Genes showing the strongest evidence for an association with SCR (candidate loci) were replicated in two additional population-based samples ('replication cohorts'). Results After the discovery meta-analysis, 29 loci were selected for replication. Association between SCR level and polymorphisms in the collagen type XXII alpha 1 (COL22A1) gene, on chromosome 8, and in the synaptotagmin-1 (SYT1) gene, on chromosome 12, were successfully replicated in the replication cohorts (p value = 1.0 × 10-6 and 1.7 × 10-4, respectively). Evidence of association was also found for polymorphisms in a locus including the gamma-aminobutyric acid receptor rho-2 (GABRR2) gene and the ubiquitin-conjugating enzyme E2-J1 (UBE2J1) gene (replication p value = 3.6 × 10-3). Previously reported findings, associating glomerular filtration rate with SNPs in the uromodulin (UMOD) gene and in the schroom family member 3 (SCHROOM3) gene were also replicated. Conclusions While confirming earlier results, our study provides new insights in the understanding of the genetic basis of serum creatinine regulatory processes. In particular, the association with the genes SYT1 and GABRR2 corroborate previous findings that highlighted a possible role of the neurotransmitters GABAA receptors in the regulation of the glomerular basement membrane and a possible interaction between GABAAreceptors and synaptotagmin-I at the podocyte level.
