  • Blockade of the CD93 pathway normalizes tumor vasculature to facilitate drug delivery and immunotherapy

    • The immature and dysfunctional vascular network within solid tumors poses a substantial obstacle to immunotherapy because it creates a hypoxic tumor microenvironment that actively limits immune cell infiltration. The molecular basis underpinning this vascular dysfunction is not fully understood. Using genome-scale receptor array technology, we showed here that insulin-like growth factor binding protein 7 (IGFBP7) interacts with its receptor CD93, and we subsequently demonstrated that this interaction contributes to abnormal tumor vasculature. Both CD93 and IGFBP7 were up-regulated in tumor-associated endothelial cells. IGFBP7 interacted with CD93 via a domain different from multimerin-2, the known ligand for CD93. In two mouse tumor models, blockade of the CD93/IGFBP7 interaction by monoclonal antibodies promoted vascular maturation to reduce leakage, leading to reduced tumor hypoxia and increased tumor perfusion. CD93 blockade in mice increased drug delivery, resulting in an improved antitumor response to gemcitabine or fluorouracil. Blockade of the CD93 pathway triggered a substantial increase in intratumoral effector T cells, thereby sensitizing mouse tumors to immune checkpoint therapy. Last, analysis of samples from patients with cancer under anti-programmed death 1/programmed death-ligand 1 treatment revealed that overexpression of the IGFBP7/CD93 pathway was associated with poor response to therapy. Thus, our study identified a molecular interaction involved in tumor vascular dysfunction and revealed an approach to promote a favorable tumor microenvironment for therapeutic intervention.

  • Evidence of a large cooling between 1690 and 1740 AD in southern Africa

    • A 350-year-long, well-dated delta O-18 stalagmite record from the summer rainfall region in South Africa is positively correlated with regional air surface temperatures at interannual time scales. The coldest period documented in this record occurred between 1690 and 1740, slightly lagging the Maunder Minimum (1645-1710). A temperature reconstruction, based on the correlation between regional surface temperatures and the stalagmite delta O-18 variations, indicates that parts of this period could have been as much as 1.4 degrees C colder than today. Significant cycles of 22, 11 and 4.8 years demonstrate that the solar magnetic and the El Nino-Southern Oscillation cycle could be important drivers of multidecadal to interannual climate variability in this region. The observation that the most important driver of stalagmite delta O-18 on interannual time scales from this subtropical region is regional surface temperature cautions against deterministic interpretations of delta O-18 variations in low-latitude stalagmites as mainly driven by the amount of precipitation.

    • High-performance polymer solar cells of an alternating polyfluorene copolymer, PFDTBT, and a fullerene derivative were studied. It was shown that PFDTBT is a promising candidate for obtaining high energy conversion efficiency when combined with a suitable electron acceptor. Results showed that high-performance solar cells with extended spectral coverage and increased open-circuit voltage could be prepared from composites prepared from electron acceptor.

  • Situation of urban mobility in Pakistan : Before, during, and after the COVID-19 lockdown with climatic risk perceptions

    • The coronavirus pandemic (COVID-19) has impacted the usual global movement patterns, atmospheric pollutants, and climatic parameters. The current study sought to assess the impact of the COVID-19 lockdown on urban mobility, atmospheric pollutants, and Pakistan’s climate. For the air pollution assessment, total column ozone (O3), sulphur dioxide (SO2), and tropospheric column nitrogen dioxide (NO2) data from the Ozone Monitoring Instrument (OMI), aerosol optical depth (AOD) data from the Multi-angle Imaging Spectroradiometer (MISR), and dust column mass density (PM2.5) data from the MERRA-2 satellite were used. Furthermore, these datasets are linked to climatic parameters (temperature, precipitation, wind speed). The Kruskal–Wallis H test (KWt) is used to compare medians among k groups (k > 2), and the Wilcoxon signed-rank sum test (WRST) is for analyzing the differences between the medians of two datasets. To make the analysis more effective, and to justify that the variations in air quality parameters are due to the COVID-19 pandemic, a Generalized Linear Model (GLM) was used. The findings revealed that the limitations on human mobility have lowered emissions, which has improved the air quality in Pakistan. The results of the study showed that the climatic parameters (precipitation, Tmax, Tmin, and Tmean) have a positive correlation and wind speed has a negative correlation with NO2 and AOD. This study found a significant decrease in air pollutants (NO2, SO2, O3, AOD) of 30–40% in Pakistan during the strict lockdown period. In this duration, the highest drop of about 28% in NO2 concentrations has been found in Karachi. Total column O3 did not show any reduction during the strict lockdown, but a minor decline was depicted as 0.38% in Lahore and 0.55% in Islamabad during the loosening lockdown. During strict lockdown, AOD was reduced up to 23% in Islamabad and 14.46% in Lahore. The results of KWt and WRST evident that all the mobility indices are significant (p < 0.05) in nature. The GLM justified that restraining human activities during the lockdown has decreased anthropogenic emissions and, as a result, improved air quality, particularly in metropolitan areas.

  • Induced morphological changes on vicinal MgO (100) subjected to high-temperature annealing: Step formation and surface stability

    • Highly regular step and terrace structures have been produced on surfaces of single crystalline MgO, miscut from the low-index (001) plane, upon annealing in air. Here, the evolution of the surface morphology of such surfaces is investigated. We demonstrate that the periodicity of these structures can be widely tuned in the submicron range by controlling the annealing conditions. Surface faceting resulted from annealing in the temperature range 1100-1580 °C. The surfaces were characterized by atomic force microscope, X-ray diffraction, transmission electron microscope, and X-ray photoelectron spectroscopy to assess the role of contamination, temperature, and miscut angle in the final morphology. The presence of Al contamination in the post-annealed samples was found to be essential for the formation of the step and terrace structure. The stability of the resultant structures when exposed to ambient conditions is discussed. The cause of the apparent destruction of the surface morphology upon long-term atmospheric exposure has been identified, and a method to recover the faceted morphology is proposed. Overall, the study further facilitates the growth of nanostructures on such faceted surfaces.

  • Intercomparison of 14C Analysis of Carbonaceous Aerosols : Exercise 2009

    • Radiocarbon analysis of the carbonaceous aerosol allows an apportionment of fossil and non-fossil sources of airborne particulate matter (PM). A chemical separation of total carbon (TC) into its subfractions organic carbon (OC) and elemental carbon (EC) refines this powerful technique, as OC and EC originate from different sources and undergo different processes in the atmosphere. Although C-14 analysis of TC, EC, and OC has recently gained increasing attention, interlaboratory quality assurance measures have largely been missing, especially for the isolation of EC and OC. In this work, we present results from an intercomparison of 9 laboratories for C-14 analysis of carbonaceous aerosol samples on quartz fiber filters. Two ambient PM samples and 1 reference material (RM 8785) were provided with representative filter blanks. All laboratories performed C-14 determinations of TC and a subset of isolated EC and OC for isotopic measurement. In general, C-14 measurements of TC and OC agreed acceptably well between the laboratories, i.e. for TC within 0.015-0.025 (FC)-C-14 for the ambient filters and within 0.041 (FC)-C-14 for RM 8785. Due to inhomogeneous filter loading, RM 8785 demonstrated only limited applicability as a reference material for C-14 analysis of carbonaceous aerosols. C-14 analysis of EC revealed a large deviation between the laboratories of 28-79% as a consequence of different separation techniques. This result indicates a need for further discussion on optimal methods of EC isolation for C-14 analysis and a second stage of this intercomparison.
