  • Natural hematite for next-generation solid oxide fuel cells

    • Natural hematite ore is used as a novel electrolyte material for advanced solid oxide fuel cells (SOFCs). This hematite-based system exhibits a maximum power density of 225 mW cm −2 at 600 °C and reaches 467 mW cm −2 whenthe hematite is mixed with perovskite-structured La 0.6Sr0.4Co0.2Fe0.8O3–δ.These results demonstrate that natural materials for next-generation SOFCs can infl uence the multiutilization of natural resources, thereby affecting the environment and energy sustainability.

    • Sulfur is an essential element of life. Thiol-containing metabolites in all organisms are involved in the regulation of diverse biological processes. Especially, the microbiome produces bioactive metabolites or biological intermediates of this compound class. The analysis of thiol-containing metabolites is challenging due to the lack of specific tools, making these compounds difficult to investigate selectively. We have now developed a new methodology comprising bicyclobutane for chemoselective and irreversible capturing of this metabolite class. We utilized this new chemical biology tool immobilized onto magnetic beads for the investigation of human plasma, fecal samples, and bacterial cultures. Our mass spectrometric investigation detected a broad range of human, dietary and bacterial thiol-containing metabolites and we even captured the reactive sulfur species cysteine persulfide in both fecal and bacterial samples. The described comprehensive methodology represents a new mass spectrometric strategy for the discovery of bioactive thiol-containing metabolites in humans and the microbiome.

  • Multi-platform metabolomics assays to study the responsiveness of the human plasma and lung lavage metabolome

    • Metabolomics as a field has been used to track changes and perturbations in the human body by investigating metabolite profiles indicating the change of metabolite levels over time and in response to different challenges. In this thesis work, the main focus was on applying multiplatform-metabolomics to study the human metabolome following exposure to perturbations, such as diet (in the form of a challenge meal) and exhaust emissions (air pollution exposure in a controlled setting). The cutting-edge analytical platforms used for this purpose were nuclear magnetic resonance (NMR), as well as gas chromatography (GC) and liquid chromatography (LC) coupled to mass spectrometry (MS). Each platform offered unique characterization features, allowing detection and identification of a specific range of metabolites. The use of multiplatform-metabolomics was found to enhance the metabolome coverage and to provide complementary findings that enabled a better understanding of the biochemical processes reflected by the metabolite profiles. Using non-targeted analysis, a wide range of unknown metabolites in plasma were identified during the postprandial stage after a well-defined challenge meal (in Paper I). In addition, a considerable number of metabolites were detected and identified in lung lavage fluid after biodiesel exhaust exposure compared to filtered air exposure (in Paper II). In parallel, using targeted analysis, both lung lavage and plasma fatty acid metabolites were detected and quantified in response to filtered air and biodiesel exhaust exposure (in Paper III and IV).Data processing of raw data followed by data analysis, using both univariate and multivariate methods, enabled changes occurring in metabolites levels to be screened and investigated. For the initial pilot postprandial study, the aim was to investigate the plasma metabolome response after a well-defined meal during the postprandial stage for two types of diet. It was found that independent of the background diet type, levels of metabolites returned to their baseline levels after three hours. This finding was taken into consideration for the biodiesel exhaust exposures studies, designed to limit the impact of dietary effects. Both targeted and non-targeted approaches resulted in important findings. For instance, different metabolite profiles were detected in bronchial wash (BW) compared to bronchoalveolar lavage (BAL) fluid with mainly NMR and LC-MS. Furthermore, biodiesel exhaust exposure resulted in different metabolite profiles as observed by GC-MS, especially in BAL. In addition, fatty acid metabolites in BW, BAL, and plasma were shown to be responsive to biodiesel exhaust exposure, as measured by a targeted LC-MS/MS protocol. In summary, the new analytical methods developed to investigate the responsiveness of the human plasma and lung lavage metabolome proved to be useful in an analytical perspective, and provided important biological findings. However, further studies are needed to validate these results.

  • Comparative genomics reveals high biological diversity and specific adaptations in the industrially and medically important fungal genus Aspergillus

    • BackgroundThe fungal genus Aspergillus is of critical importance to humankind. Species include those with industrial applications, important pathogens of humans, animals and crops, a source of potent carcinogenic contaminants of food, and an important genetic model. The genome sequences of eight aspergilli have already been explored to investigate aspects of fungal biology, raising questions about evolution and specialization within this genus.ResultsWe have generated genome sequences for ten novel, highly diverse Aspergillus species and compared these in detail to sister and more distant genera. Comparative studies of key aspects of fungal biology, including primary and secondary metabolism, stress response, biomass degradation, and signal transduction, revealed both conservation and diversity among the species. Observed genomic differences were validated with experimental studies. This revealed several highlights, such as the potential for sex in asexual species, organic acid production genes being a key feature of black aspergilli, alternative approaches for degrading plant biomass, and indications for the genetic basis of stress response. A genome-wide phylogenetic analysis demonstrated in detail the relationship of the newly genome sequenced species with other aspergilli.ConclusionsMany aspects of biological differences between fungal species cannot be explained by current knowledge obtained from genome sequences. The comparative genomics and experimental study, presented here, allows for the first time a genus-wide view of the biological diversity of the aspergilli and in many, but not all, cases linked genome differences to phenotype. Insights gained could be exploited for biotechnological and medical applications of fungi.

  • In Situ AFM and Electrochemical Study of a Waterborne Acrylic Composite Coating with CeO2 Nanoparticles for Corrosion Protection of Carbon Steel

    • The corrosion protection of a waterborne acrylic composite coating with 1 wt% ceria nanoparticles (CeNP) coated on carbon steel in 3 wt% NaCl solution was investigated by ex-situ and in situ as well as electrochemical atomic force microscopy (AFM) observations, combined with open circuit potential (OCP) and electrochemical impedance spectroscopy (EIS) measurements. The synthesized CeNP were stabilized by acetic acid. The transmission electron microscopy characterization showed fine nano-size of as-synthesized CeNP, the ex-situ AFM imaging revealed uniform dispersion of the CeNP in the composite coating and greatly reduced nano-sized pinholes in the coating. The in situ and electrochemical (EC) AFM investigations indicate release of some CeNP and aggregates from the coating surface and then precipitation of some particles and cerium-compounds during the exposure. The OCP and EIS results demonstrated that the addition of 1 wt% CeNP leads to a significantly improved long term barrier type corrosion protection of the waterborne acrylic composite coating for carbon steel in 3 wt% NaCl solution. The beneficial effect of the CeNP is attributed to the blocking of nano-sized defects and inhibition by the cerium compounds originated from the acetic acid stabilized CeNP.

    • The cover feature image represents the differences in the luminescence response upon fluoride vs cyanide binding to a 1,4,7-triazacyclonane-based Eu(III) complex equipped with a carbostyril light-harvesting antenna. The addition of excess of fluoride boosts the Eu(III) emission quantum yield by up to 6-fold for such compounds. Cyanide addition extinguishes Eu(III) luminescence despite partial anion binding to a lanthanide 3+ ion. These compounds showed opposite responses upon fluoride and cyanide binding, and the results could facilitate the design of responsive probes for multiple anion types.   • New Paradigms in GPCR Drug Discovery : Structure Prediction and Design of Ligands with Tailored Properties

    • G protein-coupled receptors (GPCRs) constitute a large superfamily of membrane proteins with key roles in cellular signaling. Upon activation by a ligand, GPCRs transduce signals from the extracellular to the intracellular environment. GPCRs are important drug targets and are associated with diseases such as central nervous system (CNS) disorders, cardiovascular diseases, cancer, and diabetes. Currently, 34% of FDA-approved drugs mediate their effects via modulation of GPCRs. Research during the past decades has resulted in a deeper understanding of GPCR structure and function. Moreover, recent breakthroughs in structural biology allowed the determination of several atomic resolution GPCR structures. New paradigms in GPCR pharmacology have also emerged that can lead to improved drugs. Together, these advances provide new avenues for structure-based drug discovery. The work in this thesis focused on how the large amount of structural data gathered over the last decades can be used to model GPCR targets for which no experimental structures are available, and the use of structure-based virtual screening (SBVS) campaigns to identify ligands with tailored pharmacological properties. In paper I, we investigated how template selection affects the virtual screening performance of homology models of the D2 dopamine receptor (D2R) and serotonin 5-HT2A receptor (5-HT2AR). In papers II and III, SBVS methods were used to identify dual inhibitors of the A2A adenosine receptor (A2AAR) and an enzyme, which could be relevant for treatment of Parkinson’s Disease, and functionally selective D2R ligands from a focused library. Finally, we also investigated how structural information can complement computational and biophysical methods to model and characterize the A2AAR-D2R heterodimer (paper IV).

    • Liquid–liquid extraction of U (VI) from concentrated phosphoric acid by using (2-ethyl hexyl) phosphonic acid, mono (2-ethyl hexyl) ester (PC88A) and di-butyl butyl phosphonate (DBBP) has been investigated. The effect of different factors affecting the extraction process (PC88A concentration, DBBP concentration, shaking time, aqueous/organic phase ratio, phosphoric acid concentration and effect of diluents) have been investigated. The obtained data of temperature on the extraction showed that the enthalpy change is −17.15 kJ mol−1. Uranium was extracted from the strip liquor by using di (2-ethylhexyl) phosphoric acid and tri-octyl phosphine oxide mixture and finally converted to a high purity UO3 product using precipitation with hydrogen peroxide and heat treatment at 365 °C.
