    • A mechanism to enhance hardness in multilayer coatings is proposed. Using the technologically important hard transition metal carbides as prototypes, although the principle is transferable also to other systems, we demonstrate, from first-principles calculations, that by suitable alloying the energy difference between several competing structures in the transition meta[ carbides is small or tunable. This creates multiphase/polytypic compounds with a random or controllable layer stacking sequence, systems in which the propagation of dislocations can be strongly suppressed by a large number of interfaces between structures with different glide systems, accordingly allowing the possibility of a greatly enhanced hardness. With modern thin-film technologies, it should therefore be possible to deposit such materials that will express multilayer characteristics with only minor changes in the chemical constitution of the material, which is in contrast to conventional superlattices.

    • Anthropogenic light is ubiquitous in areas where humans are present and is showing a progressive increase worldwide. This has far-reaching consequences for most species and their ecosystems. The effects of anthropogenic light on natural ecosystems are highly variable and complex. Many species suffer from adverse effects and often respond in a highly specific manner. Ostensibly surveyable effects such as attraction and deterrence become complicated because these can depend on the type of behavior and specific locations. Here, we considered how solutions and new technologies could reduce the adverse effects of anthropogenic light. A simple solution to reducing and mitigating the ecological effects of anthropogenic light seems unattainable, because frugal lighting practices and turning off lights may be necessary to eliminate them.

  • Characterization of a dynamic metabolon producing the defense compound dhurrin in sorghum

    • Metabolic highways may be orchestrated by the assembly of sequential enzymes into protein complexes, or metabolons, to facilitate efficient channeling of intermediates and to prevent undesired metabolic cross-talk while maintaining metabolic flexibility. Here we report the isolation of the dynamic metabolon that catalyzes the formation of the cyanogenic glucoside dhurrin, a defense compound produced in sorghum plants. The metabolon was reconstituted in liposomes, which demonstrated the importance of membrane surface charge and the presence of the glucosyltransferase for metabolic channeling. We used in planta fluorescence lifetime imaging microscopy and fluorescence correlation spectroscopy to study functional and structural characteristics of the metabolon. Understanding the regulation of biosynthetic metabolons offers opportunities to optimize synthetic biology approaches for efficient production of high-value products in heterologous hosts.
