  • Prenatal Bisphenol A Exposure is Linked to Epigenetic Changes in Glutamate Receptor Subunit Gene Grin2b in Female Rats and Humans

    • Bisphenol A (BPA) exposure has been linked to neurodevelopmental disorders and to effects on epigenetic regulation, such as DNA methylation, at genes involved in brain function. High doses of BPA have been shown to change expression and regulation of one such gene, Grin2b, in mice. Yet, if such changes occur at relevant doses in animals and humans has not been addressed. We investigated if low-dose developmental BPA exposure affects DNA methylation and expression of Grin2b in brains of adult rats. Furthermore, we assessed associations between prenatal BPA exposure and Grin2b methylation in 7-year old children. We found that Grin2b mRNA expression was increased and DNA methylation decreased in female, but not in male rats. In humans, prenatal BPA exposure was associated with increased methylation levels in girls. Additionally, Iow APGAR scores, a predictor for increased risk for neurodevelopmental diseases, were associated with higher Grin2b methylation levels in girls. Thus, we could link developmental BPA exposure and Iow APGAR scores to changes in the epigenetic regulation of Grin2b, a gene important for neuronal function, in a sexual dimorphic fashion. Discrepancies in exact locations and directions of the DNA methylation change might reflect differences between species, analysed tissues, exposure level and/or timing.

  • Inkjet printing of super yellow : Ink formulation, film optimization, OLEDs fabrication, and transient electroluminescence

    • Inkjet printing technique allows manufacturing low cost organic light emitting diodes (OLEDs) in ambient conditions. The above approach enables upscaling of the OLEDs fabrication process which, as a result, would become faster than conventionally used vacuum based processing techniques. In this work, we use the inkjet printing technique to investigate the formation of thin active layers of well-known light emitting polymer material: Super Yellow (poly(para-phenylene vinylene) copolymer). We develop the formulation of Super Yellow ink, containing non-chlorinated solvents and allowing stable jetting. Optimization of ink composition and printing resolution were performed, until good quality films suitable for OLEDs were obtained. Fabricated OLEDs have shown a remarkable characteristics of performance, similar to the OLEDs fabricated by means of spin coating technique. We checked that, the values of mobility of the charge carriers in the printed films, measured by transient electroluminescence, are similar to the values of mobility measured in spin coated films. Our contribution provides a complete framework for inkjet printing of high quality Super Yellow films for OLEDs. The description of this method can be used to obtain efficient printed OLEDs both in academic and in industrial settings.

  • Widespread episodic thiamine deficiency in Northern Hemisphere wildlife

    • Many wildlife populations are declining at rates higher than can be explained by known threats to biodiversity. Recently, thiamine (vitamin B-1) deficiency has emerged as a possible contributing cause. Here, thiamine status was systematically investigated in three animal classes: bivalves, ray-finned fishes, and birds. Thiamine diphosphate is required as a cofactor in at least five life-sustaining enzymes that are required for basic cellular metabolism. Analysis of different phosphorylated forms of thiamine, as well as of activities and amount of holoenzyme and apoenzyme forms of thiaminedependent enzymes, revealed episodically occurring thiamine deficiency in all three animal classes. These biochemical effects were also linked to secondary effects on growth, condition, liver size, blood chemistry and composition, histopathology, swimming behaviour and endurance, parasite infestation, and reproduction. It is unlikely that the thiamine deficiency is caused by impaired phosphorylation within the cells. Rather, the results point towards insufficient amounts of thiamine in the food. By investigating a large geographic area, by extending the focus from lethal to sublethal thiamine deficiency, and by linking biochemical alterations to secondary effects, we demonstrate that the problem of thiamine deficiency is considerably more widespread and severe than previously reported.

  • A computationally efficient hybrid 2D–3D subwoofer model

    • A subwoofer generates the lowest frequency range in loudspeaker systems. Subwoofers are used in audio systems for live concerts, movie theatres, home theatres, gaming consoles, cars, etc. During the last decades, numerical simulations have emerged as a cost- and time-efficient complement to traditional experiments in the design process of different products. The aim of this study is to reduce the computational time of simulating the average response for a given subwoofer design. To this end, we propose a hybrid 2D–3D model that reduces the computational time significantly compared to a full 3D model. The hybrid model describes the interaction between different subwoofer components as interacting modules whose acoustic properties can partly be pre-computed. This allows us to efficiently compute the performance of different subwoofer design layouts. The results of the hybrid model are validated against both a lumped element model and a full 3D model over a frequency band of interest. The hybrid model is found to be both accurate and computationally efficient.

  • Loudspeaker cabinet design by topology optimization

    • Using material distribution-based topology optimization, we optimize the bandpass design of a loudspeaker cabinet targeting low frequencies. The objective is to maximize the loudspeaker’s output power for a single frequency as well as a range of frequencies. To model the loudspeaker’s performance, we combine a linear electromechanical transducer model with a computationally efficient hybrid 2D–3D model for sound propagation. The adjoint variable approach computes the gradients of the objective function with respect to the design variables, and the Method of Moving Asymptotes (MMA) solves the topology optimization problem. To manage intermediate values of the material indicator function, a quadratic penalty is added to the objective function, and a non-linear filter is used to obtain a mesh independent design. By carefully selecting the target frequency range, we can guide the optimization algorithm to successfully generate a loudspeaker design with the required bandpass character. To the best of our knowledge, this study constitutes the first successful attempt to design the interior structure of a loudspeaker cabinet using topology optimization.

  • Multidimensional metrics of niche space for use with diverse analytical techniques

    • Multidimensional data are integral to many community-ecological studies and come in various forms, such as stable isotopes, compound specific analyses (e.g., amino acids and fatty acids), and both biodiversity and life history traits. Scientists employing such data often lack standardized metrics to evaluate communities in niche space where more than 2 dimensions are involved. To alleviate this problem, we developed a graphing and analytical approach for use with more than two variables, based on previously established stable isotope bi-plot metrics. We introduce here our community metrics as R scripts. By extending the original metrics to multiple dimensions, we created n-dimensional plots and metrics to characterize any set of quantitative measurements of a community. We demonstrate the utility of these metrics using stable isotope data; however, the approaches are applicable to many types of data. The resulting metrics provide more and better information compared to traditional analytic frameworks. The approach can be applied in many branches of community ecology, and it offers accessible metrics to quantitatively analyze the structure of communities across ecosystems and through time.

    • Initiation of celiac disease is triggered in the gastrointestinal tract by transglutaminase 2 (TG2) assisted deamidation of gluten peptides. Deamidation is a side-reaction to transamidation and occurs if primary amines are absent. In contrast to deamidation, transamidation does not trigger an immune response. The aim of the study was to identify a suitable food additive that interacts with TG2 binding motives in gluten-derived peptides to prevent deamidation/transamidation. Homology modelling of alpha 2-gliadin and computational screening of compounds for their binding affinity to a common TG2 binding motive (P) QLP were done by using computational approaches followed by experimental testing of TG2 activity. A database containing 1174 potential food grade ligands was screened against the model of alpha 2-gliadin (27 out of 33 aa). Out of the five best ligands, ascorbyl palmitate, was observed to decrease TG2 transamidation of gliadin by 82% +/- 2%. To completely silence the transamidation, we added zinc chloride (ZnCl2), and thereby reached a 99% +/- 1% inhibition of TG2 activity. In addition, we conducted a pilot experiment in which ascorbyl palmitate was observed to decrease TG2 deamidation of gliadin completely. We propose ascorbyl palmitate in combination with ZnCl2 with the future perspective to become an additive in celiac-safe foods.

  • Effects of liquid surface tension on gas capillaries and capillary forces at superamphiphobic surfaces

    • The formation of a bridging gas capillary between superhydrophobic surfaces in water gives rise to strongly attractive interactions ranging up to several micrometers on separation. However, most liquids used in materials research are oil-based or contain surfactants. Superamphiphobic surfaces repel both water and low-surface-tension liquids. To control the interactions between a superamphiphobic surface and a particle, it needs to be resolved whether and how gas capillaries form in non-polar and low-surface-tension liquids. Such insight will aid advanced functional materials development. Here, we combine laser scanning confocal imaging and colloidal probe atomic force microscopy to elucidate the interaction between a superamphiphobic surface and a hydrophobic microparticle in three liquids with different surface tensions: water (73 mN m−1), ethylene glycol (48 mN m−1) and hexadecane (27 mN m−1). We show that bridging gas capillaries are formed in all three liquids. Force-distance curves between the superamphiphobic surface and the particle reveal strong attractive interactions, where the range and magnitude decrease with liquid surface tension. Comparison of free energy calculations based on the capillary menisci shapes and the force measurements suggest that under our dynamic measurements the gas pressure in the capillary is slightly below ambient.

  • Topology Optimisation of Wideband Coaxial-to-Waveguide Transitions

    • To maximize the matching between a coaxial cable and rectangular waveguides, we present a computational topology optimisation approach that decides for each point in a given domain whether to hold a good conductor or a good dielectric. The conductivity is determined by a gradient-based optimisation method that relies on finite-difference time-domain solutions to the 3D Maxwell’s equations. Unlike previously reported results in the literature for this kind of problems, our design algorithm can efficiently handle tens of thousands of design variables that can allow novel conceptual waveguide designs. We demonstrate the effectiveness of the approach by presenting optimised transitions with reflection coefficients lower than −15dB over more than a 60% bandwidth, both for right-angle and end-launcher configurations. The performance of the proposed transitions is crossverified with a commercial software, and one design case is validated experimentally.

  • A predation cost to bold fish in the wild

    • Studies of predator-mediated selection on behaviour are critical for our understanding of the evolution and maintenance of behavioural diversity in natural populations. Consistent individual differences in prey behaviour, especially in the propensity to take risks (“boldness”), are widespread in the animal kingdom. Theory predicts that individual behavioural types differ in a cost-benefit trade-off where bolder individuals benefit from greater access to resources while paying higher predation-risk costs. However, explicitly linking predation events to individual behaviour under natural conditions is challenging and there is currently little data from the wild. We assayed individual behaviour and electronically tagged hundreds of fish (roach, Rutilus rutilus) before releasing them into their lake of origin, thereby exposing them to predation risk from avian apex predators (cormorants, Phalacrocorax carbo). Scanning for regurgitated tags at the cormorant roosting site provided data on individual predation events. We found that fish with higher boldness have a greater susceptibility to cormorant predation compared to relatively shy, risk-averse individuals. Our findings hereby provide unique and direct evidence of behavioural type-dependent predation vulnerability in the wild, i.e. that there is a predation cost to boldness, which is critical for our understanding of the evolution and maintenance of behavioural diversity in natural populations.
