  • Phosphorus regulates ectomycorrhizal fungi biomass production in a Norway spruce forest

    • Ectomycorrhizal fungi (EMF) are important components of soil microbial communities, and EMF biomass can potentially increase carbon (C) stocks by accumulating in the soils as necromass and producing recalcitrant structures. EMF growth depends on the C allocated belowground by the host trees, and the nutrient limitation on tree growth is expected to influence this allocation. Therefore, studying EMF production and understanding the factors that regulates it in natural soils are important to understand C cycling in forests.Fungal mycelium collected from ingrowth mesh bags is commonly used to estimate EMF biomass, but these measurements might not reflect the total EMF production since turnover rates of the hyphae are not considered. Here we estimated EMF production and turnover in response to P fertilization (applied as superphosphate) in a Norway spruce forest where nitrogen (N) deposition has resulted in phosphorus (P) limitation of plant production by using a combination of mesh bags with different incubation periods and with Bayesian inferences. To test how localized patches of N and P influence EMF production and turnover we amended some bags with a nitrogen source (methylene urea) or P source (apatite). Additionally, the Bayesian model tested the effect of seasonality (time of mesh-bag harvesting) on EMF production and turnover.We found that turnover of EMF was not affected by P fertilization or mesh-bag amendment. P fertilization had a negative effect on EMF production in all the mesh-bag amendments, suggesting a reduced belowground C allocation to the EMF when P limitation is alleviated. Apatite amendment significantly increased EMF biomass production in comparison with the pure quartz bags in the control plots but not in the P-fertilized plots. This indicates that P-rich patches enhance EMF production in P-limited forests, but not when P is not limiting. Urea amendment had a generally positive effect on EMF production, but this was significantly reduced by P fertilization, suggesting that a decrease in EMF production due to the alleviated P limitation will affect N foraging. Seasonality had a significant effect on EMF production, and the differences registered between the treatments were higher during the warmer months and disappeared at the end of the growing season.Many studies highlight the importance of N for regulating belowground C allocation to EMF in northern coniferous forests, but here we show that the P status of the forest can be equally important for belowground carbon allocation to EMF production in areas with high N deposition.

Dir. 2018:18). I februari vartannat år ger Kärnavfallsrådet sin självständiga bedömning av det aktuella läget inom kärnavfallsområdet. Bedömningen presenteras i form av en kunskapslägesrapport. Syftet med rapporten är att upp-märksamma och beskriva frågor som Kärnavfallsrådet anser viktiga och att redogöra för rådets synpunkter i dessa. Kärnavfallsrådet överlämnar härmed till regeringen årets kunskapslägesrapport SOU 2022:7 Kunskapsläget på kärnavfallsområdet 2022 – Samhället, tekniken och etiken.Bakom denna rapport står samtliga ledamöter och sakkunniga i Kärnavfallsrådet. Kärnavfallsrådets vetenskapliga sekreterare Johanna Swedin har varit projektledare för arbetet med kunskapslägesrapporten.Rapporterna om kunskapsläget på kärnavfallsområdet åren 1998, 2001, 2004, 2007, 2010, 2011, 2012, 2013, 2014, 2015, 2016, 2017, 2018 och 2020 finns även tillgängliga i en engelsk version. Rådet kommer att publicera en engelsk översättning av årets rapport under våren 2022.

  • Divergent above- and below-ground responses of fungal functional groups to forest thinning

    • Forest disturbances have a strong effect on soil fungal communities and associated ecosystem processes. However, little is known about the response of mycelial biomass to disturbances, and how fungi reallocate carbon into different fungal structures under environmental stressors. We investigated above- and below-ground fungal biomass shifts in response to different intensities of forest management in Mediterranean Pinus pinaster forests. Soil fungal biomass was estimated by ergosterol quantification and production of sporocarps was estimated from repeated field samplings during 5 years in 26 experimental plots. Abundance of mycorrhizal and saprotrophic fungi belowground was determined using Pacific Biosciences sequencing of fungal ITS2 amplicons. Thinning had a prolonged negative effect belowground, inter- and intra-annually, on total fungal biomass and on the biomass of ectomycorrhizal fungi, but not on saprotrophic fungi. Total and ectomycorrhizal mushroom yields were negatively correlated with the total and the ectomycorrhizal mycelial biomass, respectively. Thinning also correlated positively with the aboveground/belowground ratio of both total and ectomycorrhizal fungal biomass. We show potential short-term shifts in resource allocation of fungi from below-to above-ground structures under disturbances such as forest thinning. Ectomycorrhizal fungi may respond to disturbances by increasing reproduction rather than colonizing the surrounding soil.

  • Benzylpenicillin-producing Trichophyton erinacei and methicillin resistant Staphylococcus aureus carrying the mecC gene on European hedgehogs : a pilot-study

    • BACKGROUND: A high carriage rate of methicillin-resistant Staphylococcus aureus with the mecC gene (mecC-MRSA) has been described among Wild European hedgehogs (Europeaus erineaus). Due to this frequent occurrence, it has been suggested that hedgehogs could be a natural reservoir for mecC-MRSA. However, the reason why hedgehogs carry mecC-MRSA remains unknown, but it has been hypothesized that mecC-MRSA could have evolved on the skin of hedgehogs due to the co-occurrence with antibiotic producing dermatophytes. The aim of this pilot-study was therefore to investigate if hedgehogs in Sweden carry Trichophyton spp. and to provide evidence that these dermatophytes are able to produce penicillin or similar substances. In addition, the study aimed to identify if dermatophytes co-occurred with mecC-MRSA.METHODS: Samples were collected from hedgehogs (Europeaus erineaus) that were euthanized or died of natural causes. All samples were screened for dermatophytes and mecC-MRSA using selective cultivation methods. Suspected isolates were characterized using PCR-based methods, genome sequencing and bioinformatic analyses. Identification of penicillin was performed by ultra-high-performance liquid chromatography-tandem mass spectrometry.RESULTS: In total 23 hedgehogs were investigated, and it was shown that two carried Trichophyton erinacei producing benzyl-penicillin, and that these hedgehogs also carried mecC-MRSA. The study also showed that 60% of the hedgehogs carried mecC-MRSA.CONCLUSION: The pilot-study demonstrated that Trichophyton erinacei, isolated from Swedish hedgehogs, can produce benzylpenicillin and that these benzylpenicillin-producing T. erinacei co-occurred with mecC-MRSA. The study also reconfirmed the high occurrence of mecC-MRSA among hedgehogs.

  • The NORMAN Association and the European Partnership for Chemicals Risk Assessment (PARC) : let’s cooperate!

    • The Partnership for Chemicals Risk Assessment (PARC) is currently under development as a joint research and innovation programme to strengthen the scientific basis for chemical risk assessment in the EU. The plan is to bring chemical risk assessors and managers together with scientists to accelerate method development and the production of necessary data and knowledge, and to facilitate the transition to next-generation evidence-based risk assessment, a non-toxic environment and the European Green Deal. The NORMAN Network is an independent, well-established and competent network of more than 80 organisations in the field of emerging substances and has enormous potential to contribute to the implementation of the PARC partnership. NORMAN stands ready to provide expert advice to PARC, drawing on its long experience in the development, harmonisation and testing of advanced tools in relation to chemicals of emerging concern and in support of a European Early Warning System to unravel the risks of contaminants of emerging concern (CECs) and close the gap between research and innovation and regulatory processes. In this commentary we highlight the tools developed by NORMAN that we consider most relevant to supporting the PARC initiative: (i) joint data space and cutting-edge research tools for risk assessment of contaminants of emerging concern; (ii) collaborative European framework to improve data quality and comparability; (iii) advanced data analysis tools for a European early warning system and (iv) support to national and European chemical risk assessment thanks to harnessing, combining and sharing evidence and expertise on CECs. By combining the extensive knowledge and experience of the NORMAN network with the financial and policy-related strengths of the PARC initiative, a large step towards the goal of a non-toxic environment can be taken.

    • Experimental studies suggest involvement of trimethylamine N-oxide (TMAO) in the aetiology of cardiometabolic diseases and chronic kidney disease (CKD), in part via metabolism of ingested food. Using a comparative biomimetic approach, we have investigated circulating levels of the gut metabolites betaine, choline, and TMAO in human CKD, across animal species as well as during hibernation in two animal species. Betaine, choline, and TMAO levels were associated with renal function in humans and differed significantly across animal species. Free-ranging brown bears showed a distinct regulation pattern with an increase in betaine (422%) and choline (18%) levels during hibernation, but exhibited undetectable levels of TMAO. Free-ranging brown bears had higher betaine, lower choline, and undetectable TMAO levels compared to captive brown bears. Endogenously produced betaine may protect bears and garden dormice during the vulnerable hibernating period. Carnivorous eating habits are linked to TMAO levels in the animal kingdom. Captivity may alter the microbiota and cause a subsequent increase of TMAO production. Since free-ranging bears seems to turn on a metabolic switch that shunts choline to generate betaine instead of TMAO, characterisation and understanding of such an adaptive switch could hold clues for novel treatment options in burden of lifestyle diseases, such as CKD.

    • BACKGROUND: Despite centuries of research, debate remains on the scaling of metabolic rate to mass especially for intraspecific cases. The high variation of body mass within brown bears presents a unique opportunity to study the intraspecific effects of body mass on physiological variables. The amplitude of metabolic rate reduction in hibernators is dependent on body mass of the species. Small hibernators have high metabolic rates when euthermic but experience a drastic decrease in body temperature during torpor, which is necessary to reach a very low metabolic rate. Conversely, large hibernators, such as the brown bear (Ursus arctos), show a moderate decrease in temperature during hibernation, thought to be related to the bear's large size. We studied body mass, abdominal body temperature, heart rate, and accelerometer-derived activity from 63 free-ranging brown bears (1-15 years old, 15-233 kg). We tested for relationships between body mass and body temperature, heart rate, and hibernation duration.RESULTS: The smallest individuals maintained lower body temperatures during hibernation, hibernated longer, and ended hibernation later than large bears. Unlike body temperature, winter heart rates were not associated with body mass. In summer, the opposite pattern was found, with smaller individuals having higher body temperature and daytime heart rates. Body mass was associated with body temperature in the winter hypometabolic state, even in a large hibernating mammal. Smaller bears, which are known to have higher thermal conductance, reached lower body temperatures during hibernation. During summer, smaller bears had higher body temperatures and daytime heart rates, a phenomenon not previously documented within a single mammalian species.CONCLUSION: We conclude that the smallest bears hibernated more deeply and longer than large bears, likely from a combined effect of basic thermodynamics, the higher need for energy savings, and a lower cost of warming up a smaller body.

  • Forage availability, supplementary feed and ungulate density : Associations with ungulate damage in pine production forests

    • Populations of large herbivores, including members of the deer family Cervidae, are expanding across and within many regions of the northern hemisphere. Because their browsing on trees can result in economic losses to forestry and strongly affect ecosystems, it is becoming increasingly important to understand how best to mitigate resultant damage. Previous research has highlighted the importance of regulating deer density and the availability of alternative forage to reduce browsing damage levels in conifer production stands. However, often only one or two proxies of forage availability have been used instead of applying a broad foodscape approach and more knowledge is needed to understand which types of alternative forage best mitigate damage. We conducted field inventories of damage that occurred during the previous fall/winter in 112 production stands in southern Sweden, while also measuring forage availability and cervid faecal pellets in the surrounding landscape (16 ha). Local landowners provided data on supplementary feeding. We found that variation in cervid (Alces alces, Capreolus capreolus, Cervus elaphus and Dama dama) browsing damage to top shoots or stems of young Scots pine trees (Pinus sylvestris, hereon pine), was better explained by the availability of alternative natural forage (using several indices and species of trees and shrubs) than by supplementary feeding. The proportion of damaged pine trees was higher in stands with a lower density of pine stems; in landscapes with a lower density of key broadleaf tree species (genera Sorbus, Salix, Populus and Quercus); and in landscapes with more open land (agricultural fields and paddocks). Damage was also higher in stands where relatively large amounts of moose faeces was found, while not related to the amount of faeces from other cervid species. The amount of supplementary feed (silage or other types such as root vegetables) did not explain variation in pine damage, but the result was possibly affected by relatively few study areas supplying sufficient data on supplementary feeding. The results from our inventory illustrate the efficacy of using naturally growing forage to mitigate browsing damage to young pine trees in managed landscapes. Creation of such forage is also recommended over supplementary feeding because of co-benefits to forest biodiversity and ecosystem services.

    • Brown bears conserve muscle and bone mass during six months of inactive hibernation. The molecular mechanisms underlying hibernation physiology may have translational relevance for human therapeutics. We hypothesize that protective mechanisms involve increased tissue availability of the insulin-like growth factors (IGFs). In subadult Scandinavian Brown Bears, we observed that mean plasma IGF-1 and IGF-2 during hibernation was reduced to 36±10% and 56±15%, respectively, compared to the active state (N=12). Western ligand blotting identified IGFBP-3 as the major IGF binding protein in the active state, while IGFBP-2 was co-dominant during hibernation. Acid labile subunit (ALS) levels in hibernation were 41±16% those of the active state (N=6). Analysis of available grizzly bear RNA sequencing data revealed unaltered liver mRNA IGF-1, IGFBP-2, and IGFBP-3 levels, whereas ALS was significantly reduced during hibernation (N=6). Reduced ALS synthesis and circulating levels during hibernation should prompt a shift from ternary IGF/IGFBP/ALS to smaller binary IGF/IGFBP complexes, thereby increasing IGF tissue availability. Indeed, Size Exclusion Chromatography of bear plasma, demonstrate a shift to lower molecular weight IGF-containing complexes in the hibernating versus the active state. Further, we note that the major IGF-2 mRNA isoform expressed in liver in both Scandinavian brown bears and grizzly bears was an alternative splice variant in which Ser29 was replaced with a tetrapeptide possessing a positively charged Arg residue. Homology modelling of the bear IGF-2/IGFBP-2 complex showed the tetrapeptide in proximity to the heparin binding domain involved in bone-specific targeting of this complex. In conclusion, this study provides data which suggest that increased IGF tissue availability combined with tissue-specific targeting contribute to tissue preservation in hibernating bears.

  • Differential Changes in Circulating Steroid Hormones in Hibernating Brown Bears : Preliminary Conclusions and Caveats

    • Brown bears are obese when they enter the den, and after 6 mo of hibernation and physical inactivity, bears show none of the adverse consequences of a sedentary lifestyle in humans, such as cardiovascular disease, type 2 diabetes, and kidney failure. The metabolic mechanisms that drive hibernation physiology in bears are poorly defined, but systemic endocrine regulators are likely involved. To investigate the potential role of steroid hormones, we quantified the total levels of 12 steroid hormones, the precursor cholesterol, sex hormone-binding globulin (SHBG), and corticosterone-binding globulin (CBG) in paired serum samples from subadult free-ranging Scandinavian brown bears during the active and hibernation states. During hibernation, androstenedione and testosterone were significantly decreased in subadult female bears (n=13), whereas they increased in all males but one (n=6) and therefore did not reach a significant difference. Despite this difference, SHBG increased more than 20-fold during hibernation for all bears. Compared with SHBG concentrations in humans, bear levels were very low in the active state, but during hibernation, levels equaled high levels in humans. The increased SHBG levels likely maintain a state of relative quiescence of the reproductive hormones in hibernating bears. Interestingly, the combination of SHBG and testosterone levels results in similar free bioavailable testosterone levels of 70-80 pM in both subadult and adult sexually active male bears, suggesting a role for SHBG in controlling androgen action during hibernation in males. Dehydroepiandrosterone sulfate, dihydrotestosterone, and estradiol levels were below the detection limit in all but one animal. The metabolically active glucocorticoids were significantly higher in both sexes during hibernation, whereas the inactive metabolite cortisone was reduced and CBG was low approaching the detection limit. A potential caveat is that the glucocorticoid levels might be affected by the ketamine applied in the anesthetic mixture for hibernating bears. However, increased hibernating cortisol levels have consistently been reported in both black bears and brown bears. Thus, we suggest that high glucocorticoid activity may support the hibernation state, likely serving to promote lipolysis and gluconeogenesis while limiting tissue glucose uptake to maintain a continuous glucose supply to the brain.
