  • Optimizing the LSST Observing Strategy for Dark Energy Science : DESC Recommendations for the Wide-Fast-Deep Survey

    • Cosmology is one of the four science pillars of LSST, which promises to be transformative for our understanding of dark energy and dark matter. The LSST Dark Energy Science Collaboration (DESC) has been tasked with deriving constraints on cosmological parameters from LSST data. Each of the cosmological probes for LSST is heavily impacted by the choice of observing strategy. This white paper is written by the LSST DESC Observing Strategy Task Force (OSTF), which represents the entire collaboration, and aims to make recommendations on observing strategy that will benefit all cosmological analyses with LSST. It is accompanied by the DESC DDF (Deep Drilling Fields) white paper (Scolnic et al.). We use a variety of metrics to understand the effects of the observing strategy on measurements of weak lensing, large-scale structure, clusters, photometric redshifts, supernovae, strong lensing and kilonovae. In order to reduce systematic uncertainties, we conclude that the current baseline observing strategy needs to be significantly modified to result in the best possible cosmological constraints. We provide some key recommendations: moving the WFD (Wide-Fast-Deep) footprint to avoid regions of high extinction, taking visit pairs in different filters, changing the 2x15s snaps to a single exposure to improve efficiency, focusing on strategies that reduce long gaps (>15 days) between observations, and prioritizing spatial uniformity at several intervals during the 10-year survey.

  • Calmodulin acts as a state-dependent switch to control a cardiac potassium channel opening

    • Calmodulin (CaM) and PIP2 are potent regulators of the voltage-gated potassium channel KCNQ1 (KV7.1), which conducts the IKs current important for repolarization of cardiac action potentials. Although cryo-EM structures revealed intricate interactions between the KCNQ1 voltage-sensing domain (VSD), CaM, and PIP2, the functional consequences of these interactions remain unknown. Here, we show that CaM-VSD interactions act as a state-dependent switch to control KCNQ1 pore opening. Combined electrophysiology and molecular dynamics network analysis suggest that VSD transition into the fully-activated state allows PIP2 to compete with CaM for binding to VSD, leading to the conformational change that alters the VSD-pore coupling. We identify a motif in the KCNQ1 cytosolic domain which works downstream of CaM-VSD interactions to facilitate the conformational change. Our findings suggest a gating mechanism that integrates PIP2 and CaM in KCNQ1 voltage-dependent activation, yielding insights into how KCNQ1 gains the phenotypes critical for its function in the heart.

  • Associations between common genetic variants and income provide insights about the socioeconomic health gradient

    • We conducted a genome-wide association study (GWAS) on income among individuals of European descent and leveraged the results to investigate the socio-economic health gradient (N=668,288). We found 162 genomic loci associated with a common genetic factor underlying various income measures, all with small effect sizes. Our GWAS-derived polygenic index captures 1 - 4% of income variance, with only one-fourth attributed to direct genetic effects. A phenome-wide association study using this polygenic index showed reduced risks for a broad spectrum of diseases, including hypertension, obesity, type 2 diabetes, coronary atherosclerosis, depression, asthma, and back pain. The income factor showed a substantial genetic correlation (0.92, s.e. = .006) with educational attainment (EA). Accounting for EA's genetic overlap with income revealed that the remaining genetic signal for higher income related to better mental health but reduced physical health benefits and increased participation in risky behaviours such as drinking and smoking.
