  • Molecular profiling using tissue microarrays as a tool to identify predictive biomarkers in laryngeal cancer treated with radiotherapy

    • Aim: To explore the usefulness of the expression of five potential cancer biomarkers in predicting outcome in patients with laryngeal cancer. Materials and Methods: In the present study, the Swedish National Cancer Registry databases were used to identify patients with laryngeal cancer diagnosed during the years 1978-2004 in the Uppsala-Örebro region and treated with radiotherapy. The expression of Ki-67, MutS homolog 2, (MSH2), p53, B-cell CLL/lymphoma 2 (Bcl-2) and cyclin D1 in the cancer cells was assessed immunohistochemically using tissue microarrays (TMAs) and its predictve value on survival and relapse was analyzed using Cox regression models. Results: A total of 39 patients were included in the present study. Nuclear MSH2 staining was statistically significantly correlated to Ki-67 expression (p=0.022). However, univariate and multivariate Cox analyses showed no statistically significant association between the expression of the investigated biomarkers and overall survival or relapse. Conclusion: The present exploratory study does not show any significant predictive value of the biomarkers examined with respect to survival or relapse. However, with larger patient cohorts, we believe that protein profiling using TMAs and immunohistochemistry is a feasible strategy for prognostic and predictive biomarker screening in laryngeal cancer.

  • Observation of Ni-54 : Cross-conjugate symmetry in f(7/2) mirror energy differences

    • Gamma decays from excited states up to J(pi)=6(+) in the N=Z-2 nucleus Ni-54 have been identified for the first time. Level energies are compared with those of the isobars Co-54 and Fe-54 and of the cross-conjugate nuclei of mass A=42. The good but puzzling f(7/2) cross-conjugate symmetry in mirror and triplet energy differences is analyzed. Shell model calculations reproduce the new data but the necessary nuclear charge-dependent phenomenology is not fully explained by modern nucleon-nucleon potentials.

    • A search for charged-lepton-flavor violating Formula Presented (Formula Presented, Formula Presented) interactions is presented, considering both top-quark production and decay. The data analyzed correspond to Formula Presented of proton-proton collisions at a center-of-mass energy of Formula Presented recorded with the ATLAS detector at the Large Hadron Collider. The analysis targets events containing two muons with the same electric charge, a hadronically decaying Formula Presented-lepton and at least one jet, with exactly one Formula Presented-tagged jet, produced by a Formula Presented interaction. Agreement with the Standard Model expectation within 1.6 standard deviations is observed, and limits are set at the 95% confidence level (CL) on the charged-lepton-flavor violation branching ratio of Formula Presented. An effective field theory interpretation is performed yielding 95% CL limits on Wilson coefficients, dependent on the flavor of the associated light quark and the Lorentz structure of the coupling. These range from Formula Presented for Formula Presented to Formula Presented for Formula Presented. An additional interpretation is performed for scalar leptoquark production inducing charged-lepton-flavor violation, with fixed intergenerational couplings. Upper limits on leptoquark coupling strengths are set at the 95% CL, ranging from Formula Presented to Formula Presented for leptoquark masses between 0.5 and 2.0 TeV. © 2024 CERN, for the ATLAS Collaboration.

  • Search for Nearly Mass-Degenerate Higgsinos Using Low-Momentum Mildly Displaced Tracks in pp Collisions at sqrt[s]=13 TeV with the ATLAS Detector

    • Higgsinos with masses near the electroweak scale can solve the hierarchy problem and provide a dark matter candidate, while detecting them at the LHC remains challenging if their mass splitting is O(1 GeV). This Letter presents a novel search for nearly mass-degenerate Higgsinos in events with an energetic jet, missing transverse momentum, and a low-momentum track with a significant transverse impact parameter using 140 fb^{-1} of proton-proton collision data at sqrt[s]=13 TeV collected by the ATLAS experiment. For the first time since LEP, a range of mass splittings between the lightest charged and neutral Higgsinos from 0.3 to 0.9 GeV is excluded at 95% confidence level, with a maximum reach of approximately 170 GeV in the Higgsino mass.

    • Searches for exclusive decays of the Higgs boson into D⁎γ and of the Z boson into D0γ and Ks0γ can probe flavour-violating Higgs boson and Z boson couplings to light quarks. Searches for these decays are performed with a pp collision data sample corresponding to an integrated luminosity of 136.3 fb−1 collected at s=13TeV between 2016–2018 with the ATLAS detector at the CERN Large Hadron Collider. In the D⁎γ and D0γ channels, the observed (expected) 95% confidence-level upper limits on the respective branching fractions are B(H→D⁎γ)

    • Liquid biopsies based on extracellular vesicles (EVs) represent a promising tool for treatment monitoring of tumors, including non-small-cell lung cancers (NSCLC). In this study, we report on a multiplexed electrokinetic sensor for surface protein profiling of EVs from clinical samples. The method detects the difference in the streaming current generated by EV binding to the surface of a functionalized microcapillary, thereby estimating the expression level of a marker. Using multiple microchannels functionalized with different antibodies in a parallel fluidic connection, we first demonstrate the capacity for simultaneous detection of multiple surface markers in small EVs (sEVs) from NSCLC cells. To investigate the prospects of liquid biopsies based on EVs, we then apply the method to profile sEVs isolated from the pleural effusion (PE) fluids of five NSCLC patients with different genomic alterations (ALK, KRAS or EGFR) and applied treatments (chemotherapy, EGFR- or ALKtyrosine kinase inhibitors). The vesicles were targeted against CD9, as well as EGFR and PD-L1, two treatment targets in NSCLC. The electrokinetic signals show detection of these markers on sEVs, highlighting distinct interpatient differences, e.g., increased EGFR levels in sEVs from a patient with EGFR mutation as compared to an ALK-fusion one. The sensors also detect differences in PD-L1 expressions. The analysis of sEVs from a patient prior and post ALK-TKI crizotinib treatment reveals significant increases in the expressions of some markers (EGFR and PD-L1). These results hold promise for the application of the method for tumor treatment monitoring based on sEVs from patient liquid biopsies.

    • A generalized linear model based on Poisson regression has been used to assess the impact of environmental variables modulating tropical cyclone frequency in six main cyclone development areas: the East Pacific, West Pacific, North Atlantic, North Indian, South Indian, and South Pacific. The analysis covers the period 1980-2009 and focuses on widely used meteorological parameters including wind shear, sea surface temperature, and relative humidity from different reanalyses as well as aerosol optical depth for different compounds simulated by the Goddard Chemistry Aerosol Radiation and Transport model. Circulation indices are also included. Cyclone frequency is obtained from the International Best Track Archive for Climate Stewardship. A strong link is found between cyclone frequency and the relative sea surface temperature, Atlantic Meridional Mode, and wind shear with significant explained log likelihoods in the North Atlantic of 37%, 27%, and 28%, respectively. A significant impact of black carbon and organic aerosols on cyclone frequency is found over the North Indian Ocean, with explained log likelihoods of 27%. A weaker but still significant impact is found for observed dust aerosols in the North Atlantic with an explained log likelihood of 11%. Changes in lower stratospheric temperatures explain 28% of the log likelihood in the North Atlantic. Lower stratospheric temperatures from a subset of Coupled Model Intercomparison Project Phase 5 models properly simulate the warming and subsequent cooling of the lower stratosphere that follows a volcanic eruption but underestimates the cooling by about 0.5 degrees C.

    • The Maribot Vane is an autonomous sailing drone project that has been in the prototype testing phase for two years, developing a fully capable platform to aid in long-term ocean research projects. With longer voyages required for further development, a more reliable physical platform is a high priority of the next-generation design. This paper applies a multi-objective, Reliability-Based Design Optimization (RBDO) approach to minimize the system cost and the probability of failure under anticipated operational conditions. The results will inform designers of the new platform about their trade-offs between the cost and reliability objectives.

  • Statistical anomaly detection for train fleets

    • We have developed a method for statistical anomaly detection that has been deployed in a tool for condition monitoring of train fleets. The tool is currently used by several railway operators across the world to inspect and visualize the occurrence of "event messages" generated on the trains. The anomaly detection component helps the operators quickly to find significant deviations from normal behavior and to detect early indications for possible problems. The method used is based on Bayesian principal anomaly, which is a framework for parametric anomaly detection using Bayesian statistics. The savings in maintenance costs of using the tool comes mainly from avoiding costly breakdowns and have been estimated to be several million Euros per year for the tool. In the long run, it is expected that maintenance costs can be reduced by between 5 and 10 percent with the help of the tool.

    • Ultra-fast time-delayed techniques have been recently applied in a number of studies where exotic nuclei were identified using advanced selection techniques. These include large Compton-suppressed Ge arrays, in-flight separators or recoil separators. Some of the new results are discussed in this presentation. Besides the results for Mg-32 and Pd-96, they include the first determination of the half-life of the 8(+) state in Ge-80, T-1/2 = 2.95(6) ns, and significantly more precise results for Mn-51 (3680 keV level) and V-48 (421 keV level), T-1/2 = 1760(40) ps and T-1/2.
