  • Exceptional overproduction of a functional human membrane protein

    • Eukaryotic-especially human-membrane protein overproduction remains a major challenge in biochemistry. Heterologously overproduced and purified proteins provide a starting point for further biochemical, biophysical and structural studies, and the lack of sufficient quantities of functional membrane proteins is frequently a bottleneck hindering this. Here, we report exceptionally high production levels of a correctly folded and crystallisable recombinant human integral membrane protein in its active form; human aquaporin 1 (hAQP1) has been heterologously produced in the membranes of the methylotrophic yeast Pichia pastoris. After solubilisation and a two step purification procedure, at least 90 mg hAQP1 per liter of culture is obtained. Water channel activity of this purified hAQP was verified by reconstitution into proteoliposomes and performing stopped-flow vesicle shrinkage measurements. Mass spectrometry confirmed the identity of hAQPI in crude membrane preparations, and also from purified protein reconstituted into proteoliposomes. Furthermore, crystallisation screens yielded diffraction quality crystals of untagged recombinant hAQP1. This study illustrates the power of the yeast P. pastoris as a host to produce exceptionally high yields of a functionally active, human integral membrane protein for subsequent functional and structural characterization.   • Criteria and Decision Support for A Sustainable Choice of Alternative Marine Fuels

    • To reach the International Maritime Organization, IMO, vision of a 50% greenhouse gas (GHG) emission reduction by 2050, there is a need for action. Good decision support is needed for decisions on fuel and energy conversion systems due to the complexity. This paper aims to get an overview of the criteria types included in present assessments of future marine fuels, to evaluate these and to highlight the most important criteria. This is done using a literature review of selected scientific articles and reports and the authors’ own insights from assessing marine fuels. There are different views regarding the goal of fuel change, what fuel names to use as well as regarding the criteria to assess, which therefore vary in the literature. Quite a few articles and reports include a comparison of several alternative fuels. To promote a transition to fuels with significant GHG reduction potential, it is crucial to apply a life cycle perspective and to assess fuel options in a multicriteria perspective. The recommended minimum set of criteria to consider when evaluating future marine fuels differ somewhat between fuels that can be used in existing ships and fuels that can be used in new types of propulsion systems

  • Study for Improving Maternal Pregnancy And Child ouTcomes (IMPACT): a study protocol for a Swedish prospective multicentre cohort study

    • Introduction First-trimester pregnancy risk evaluation facilitates individualised antenatal care, as well as application of preventive strategies for pre-eclampsia or birth of a small for gestational age infant. A range of early intervention strategies in pregnancies identified as high risk at the end of the first trimester has been shown to decrease the risk of preterm pre-eclampsia (<37 gestational weeks). The aim of this project is to create the Improving Maternal Pregnancy And Child ouTcomes (IMPACT) database; a nationwide database with individual patient data, including predictors recorded at the end of the first trimester and later pregnancy outcomes, to identify women at high risk of pre-eclampsia. A second aim is to link the IMPACT database to a biobank with first-trimester blood samples. Methods and analysis This is a Swedish prospective multicentre cohort study. Women are included between the 11th and 14th weeks of pregnancy. At inclusion, pre-identified predictors are retrieved by interviews and medical examinations. Blood samples are collected and stored in a biobank. Additional predictors and pregnancy outcomes are retrieved from the Swedish Pregnancy Register. Inclusion in the study began in November 2018 with a targeted sample size of 45 000 pregnancies by end of 2021. Creation of a new risk prediction model will then be developed, validated and implemented. The database and biobank will enable future research on prediction of various pregnancy-related complications. Ethics and dissemination Confidentiality aspects such as data encryption and storage comply with the General Data Protection Regulation and with ethical committee requirements. This study has been granted national ethical approval by the Swedish Ethical Review Authority (Uppsala 2018-231) and national biobank approval at Uppsala Biobank (18237 2 2018 231). Results from the current as well as future studies using information from the IMPACT database will be published in peer-reviewed journals.

    • Globally, about 1.3 million people die every year in road traffic crashes and about 50 million are injured. For long, the death toll on roads was considered to be a necessary price that we have to pay for our mobility and development. But beginning in the late 1990s, an alternative approach to road safety has become more and more influential. The Vision Zero movement declares that every severe crash in road traffic is an avoidable failure and that no other goal is satisfactory than zero fatalities and serious injuries. This is by no means a new idea. Similar views have been expressed in many other areas of safety management. Safety work based on the idea that every accident is one too much can be found in workplace safety, fire safety, aviation, suicide prevention, patient safety, infection control, and many other areas. Zero goals are also gaining traction in environmental protection and sustainable development work. Terms such as zero waste, zero emissions, zero carbon, and zero poverty have become increasingly important in climate and environmental policies. Detractors claim that Vision Zero is too stringent and therefore also unrealistic. But practical experience has shown again and again that the Vision Zero approach can efficiently reduce the number of deaths and serious injuries. Of course, it is not enough to set up Vision Zero as a goal. Its effects materialize when it is systematically applied, and every serious accident is treated as a failure that must not be repeated. This handbook is the first comprehensive collection of knowledge and experience of Vision Zero. Its contributing authors are experts from all around the world, representing a wide range of academic disciplines and an equally wide range of specializations in practical safety management.

  • Safe Efficient Vehicle Solutions -On Driving Forces for Future Road Transportations

    • The primary objective of this paper is to present the most relevant factors and driving forces that influence future sustainable road transportations and exemplify how they may influence the development. The research methodology used is explorative scenarios where data collected from workshops, expert panels and surveys lay the foundations for the explanatory models [1]. Several driving forces are identified. However, two of them are found to be more important for the study as they have a strong influence on the development of the road transport system; yet it is uncertain how these driving forces will develop. The first of these driving forces is the ability of the authorities to take an active role when developing a sustainable transport system and the second how actively people will demand and support changes in the vehicles and the transport system. Four different future road transportation scenarios have been created to explore how changes in these two driving forces will influence the development of vehicles and road transport system; these scenarios are explained together with characteristics of future road transportation solutions. It is concluded that plans for technology development need to consider the uncertainties of these driving forces in order to enable creation of robust development roadmaps.

    • This handbook provides a comprehensive treatment of Vision Zero, an innovative policy on public road safety developed in Sweden. Covering all the major topics of the subject, the book starts out with a thorough examination of the philosophy, ideas and principles behind Vision Zero. It looks at conditions for the effectiveness of the policy, principles of safety and responsibility as well as critique on the policy. Next, the handbook focuses on how the Vision Zero ideas have been received and implemented in various legislations and countries worldwide. It takes into account the way Vision Zero is looked at in the context of international organizations such as the WHO, the UN, and the OECD. This allows for a comparison of systems, models and effects. The third part of the handbook discusses the management and leadership aspects, including ISO standards, equity issues, other goals for traffic and transportation, and opportunities for the car industry. Part four delves into tools, technologies and organizational measures that contribute to the implementation of Vision Zero in road traffic. Examples of specific elements discussed are urban and rural road designs, human factor designs, and avoiding drunk and distracted driving. The final part of the handbook offers perspectives on the transfer of Vision Zero policy to other areas, ranging from air traffic to suicide prevention and nuclear energy.

  • Biofuels and land use in Sweden: an overview of land-use change effects

    • Supported by policies, biofuel production has been continuously increasing worldwide during recent years. However, concerns have been raised that biofuels, often advocated as the future substitute for greenhouse gas (GHG) intensive fossil fuels, may cause negative effects on the climate and the environment. When assessing GHG emissions from biofuels, the production phase of the biofuel crop is essential since this is the phase in which most of the GHG emissions occur during the life cycle of the fuel, often linked to land use and land management. Changes in land use can result from a wide range of anthropogenic activities including agriculture and forestry management, livestock and biofuel production. The report first presents a review of the literature in the different scientific areas related to land use change (LUC) and biofuel production. Knowledge gaps related to LUC is compiled and, a synthesis is developed highlighting major challenges and key findings. Main findings are that (i) deforestation, forest management, and climate change deforestation is a major contributor to GHG emissions and can contribute to soil erosion and carbon stock changes, (ii) albedo changes and the timing of emissions need to be better understood, (iii) to avoid degradation of biodiversity great care must be taken to develop sustainable biofuel production (iv) nutrient leakage and removal of forest residues can influence the biomass growth potential (v) to avoid fertility losses in agricultural soils during biofuel production, crops with low fertilizer needs, high nutrient use efficiency and high yields should be given priority (vi) indirect effects on land use are extremely complex to quantify without great uncertainty (vii) biofuels contribution to rising food prices and poverty even more challenging (viii) biofuel production can create jobs but also interfere with traditional ways of life and recreational values, (ix) to avoid negative effects, biofuel production should be developed in collaboration with the stakeholders involved: farmers, land owners, tourists, and industry. The literature review and synthesis presented in this report shows that land use on this planet is already placing high stress on ecosystems, atmosphere, soils and human life. Because of increased biofuel production, land use change is therefore at risk of aggravating these problems. Conclusions drawn are that the LUC caused by increasing use of biofuels can be negative to various degrees but that drawbacks can be mitigated through policy measures or technology developments. Examples include the cultivation of high-yielding crops, cultivation on abandoned arable land, and effective use of by-products and waste. To explore the opportunities that exist for beneficial land use change, continued responsible and sensitive collaboration between industry, policy-makers, researchers and local communities is a prerequisite.

  • Ecosystem services in life cycle assessment: A synthesis of knowledge and recommendations for biofuels

    • There is an increasing trend in promoting the use of biofuels for transportation as a low-fossil carbon energy source, but little knowledge on their multidimensional environmental impacts. Whole-system approaches, such as life cycle assessment (LCA), have been extensively employed to analyze the environmental performance of different biofuels. However, it remains unclear to which extent biofuels impact ecosystems and the services they provide, in particular related to different management practices. To overcome this challenge, this paper draws recommendations to better holistically address ecosystem services (ES) in LCA, with a focus on biofuels. We first pinpoint some of the challenges in accounting for the concept of ES in decision-making and review some of the existing ES classification frameworks and the usefulness of the cascade model. Second, we discuss the implications of identified context-specific aspects on the modeling of biofuel production impacts on ES in LCA. Finally, we propose a conceptual framework to link ES classification systems, the cascade model and the LCA approach. Although some challenges still remain unsolved, due to the existing life cycle impact assessment structure, existing ES frameworks and the cascade model are helpful tools to better include ES into LCA of different biofuels.

    • Glioblastoma multiforme (GBM) is the most aggressive brain tumor in adults, and despite state-of-the-art treatment, survival remains poor and novel therapeutics are sorely needed. The aim of the present study was to identify new synergistic drug pairs for GBM. In addition, we aimed to explore differences in drug-drug interactions across multiple GBM-derived cell cultures and predict such differences by use of transcriptional biomarkers. We performed a screen in which we quantified drug-drug interactions for 465 drug pairs in each of the 5 GBM cell lines U87MG, U343MG, U373MG, A172, and T98G. Selected interactions were further tested using isobole-based analysis and validated in 5 glioma-initiating cell cultures. Furthermore, drug interactions were predicted using microarray-based transcriptional profiling in combination with statistical modeling. Of the 5 465 drug pairs, we could define a subset of drug pairs with strong interaction in both standard cell lines and glioma-initiating cell cultures. In particular, a subset of pairs involving the pharmaceutical compounds rimcazole, sertraline, pterostilbene, and gefitinib showed a strong interaction in a majority of the cell cultures tested. Statistical modeling of microarray and interaction data using sparse canonical correlation analysis revealed several predictive biomarkers, which we propose could be of importance in regulating drug pair responses. We identify novel candidate drug pairs for GBM and suggest possibilities to prospectively use transcriptional biomarkers to predict drug interactions in individual cases.

  • Electrofuels – a possibility for shipping in a low carbon future?

    • Continued growth of carbon dioxide (CO2) emissions from the shipping industry until 2050 and beyond is expected although of the recent decline. The global share of anthropogenic CO2 emissions from ships is only about 2 percent, but there is a risk that this share will increase substantially if no action is taken. What are the possibilities for decarbonisation of the shipping industry, then? Some of the measures discussed are energy efficiency, use of biofuels and use of hydrogen. In this paper a fourth option is scrutinised – use of electrofuels. Electrofuels is an umbrella term for carbon-based fuels, e.g. methane or methanol, which are produced using electricity as the primary source of energy. The carbon in the fuel comes from CO2 which can be captured from various industrial processes such as exhaust gases, the sea or the air. The production of electrofuels is still in its infancy, and many challenges need to be overcome before electrofuels are brought to market on a large scale. First, this paper gives an overview of the current status of electrofuels regarding technologies, efficiencies and costs. Second, as electrofuels production requires significant amounts of CO2 and electricity, the feasibility to produce enough electrofuels to supply all ships bunkering in Sweden, with regionally produced electricity and regionally emitted CO2, and the amount of CO2 that is required to supply all ships globally is evaluated in two case studies assessing supply potential.
