  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • The Visual Object Tracking challenge VOT2016 aims at comparing short-term single-object visual trackers that do not apply pre-learned models of object appearance. Results of 70 trackers are presented, with a large number of trackers being published at major computer vision conferences and journals in the recent years. The number of tested state-of-the-art trackers makes the VOT 2016 the largest and most challenging benchmark on short-term tracking to date. For each participating tracker, a short description is provided in the Appendix. The VOT2016 goes beyond its predecessors by (i) introducing a new semi-automatic ground truth bounding box annotation methodology and (ii) extending the evaluation system with the no-reset experiment.

  • Improved efficiency of organic solar cell using MoS2 doped poly (3,4-ethylenedioxythiophene)(PEDOT) as hole transport layer

    • We report an efficient hole transporting layer (HTL) for organic solar cell (OSC) based on solution-processed organic-inorganic hybrid composed of ultrasonic-exfoliated MoS2 nanosheets and dopamine-copolymerized poly(3,4-ethylenedioxythiophene) (PEDOT) derivative (DA-P). The OSCs based on this new hybrid HTL show a marked performance improvement over those with single-component HTLs, and they retain up to 80% of their original power conversion efficiency after 35 days. Our investigations reveal that the boost in performance is due to a synergistic effect that improves both hole transport and extraction ability. This effect is mainly due to the doping of exfoliated-MoS2 nanosheets on DA-P. We employ a comprehensive range of spectroscopies to uncover that the dopant is derived from the oxidation products of MoS2 nanosheets during the ultrasonic exfoliation. Our work demonstrates an efficient hybrid HTL and offers new insights into the interaction of exfoliated-MoS2 nanosheets and the PEDOT derivatives.

  • Functional mechanisms underlying pleiotropic risk alleles at the 19p13.1 breast-ovarian cancer susceptibility locus

    • A locus at 19p13 is associated with breast cancer (BC) and ovarian cancer (OC) risk. Here we analyse 438 SNPs in this region in 46,451 BC and 15,438 OC cases, 15,252 BRCA1 mutation carriers and 73,444 controls and identify 13 candidate causal SNPs associated with serous OC (P=9.2 × 10-20), ER-negative BC (P=1.1 × 10-13), BRCA1-associated BC (P=7.7 × 10-16) and triple negative BC (P-diff=2 × 10-5). Genotype-gene expression associations are identified for candidate target genes ANKLE1 (P=2 × 10-3) and ABHD8 (P<2 × 10-3). Chromosome conformation capture identifies interactions between four candidate SNPs and ABHD8, and luciferase assays indicate six risk alleles increased transactivation of the ADHD8 promoter. Targeted deletion of a region containing risk SNP rs56069439 in a putative enhancer induces ANKLE1 downregulation; and mRNA stability assays indicate functional effects for an ANKLE1 3′-UTR SNP. Altogether, these data suggest that multiple SNPs at 19p13 regulate ABHD8 and perhaps ANKLE1 expression, and indicate common mechanisms underlying breast and ovarian cancer risk.

  • An n-n Heterojunction Configuration for Efficient Electron Transport in Organic Photovoltaic Devices

    • Selective electron transport and extraction are essential to the operation of photovoltaic devices. Electron transport layer (ETL) is therefore critical to organic photovoltaics (OPV). Herein, an ETL configuration is presented comprising a solution-processed n-n organic heterojunction to enhance electron transport and hole blocking, and boost power conversion efficiency (PCE) in OPV. Specifically, the n-n heterojunction is constructed by stacking a narrow-band n-type conjugated polymer layer (PNDIT-F3N) and a wide-band n-type conjugated molecule layer (Phen-NaDPO). Based on the ultraviolet photoelectron spectroscopy measurement and numerical simulation of current density-voltage characteristics, the formation of the built-in potential is investigated. In three OPVs with different active layers, substantial improvements are observed in performance following the introduction of this ETL configuration. The performance enhancement arises from the combination of selective carrier transport properties and reduced recombination. Another contributing factor is the good film-forming quality of the new ETL configuration, where the surface energies of the related materials are well-matched. The n-n organic heterojunction represents a viable and promising ETL construction strategy for efficient OPV devices.

  • Single-Atom-Thick Active Layers Realized in Nanolaminated Ti-3(AlxCu1-x)C-2 and Its Artificial Enzyme Behavior

    • A Ti-3(AlxCu1-x)C-2 phase with Cu atoms with a degree of ordering in the A plane is synthesized through the A site replacement reaction in CuCl2 molten salt. The weakly bonded single -atom -thick Cu layers in a Ti-3(AlxCu1-x)C-2 MAX phase provide actives sites for catalysis chemistry. As -synthesized Ti-3(AlxCu1-x)C-2 presents unusual peroxidase-like catalytic activity similar to that of natural enzymes. A fabricated Ti-3(AlxCu1-x)C-2/chitosan/glassy carbon electrode biosensor prototype also exhibits a low detection limit in the electrochemical sensing of H2O2. These results have broad implications for property tailoring in a nanolaminated MAX phase by replacing the A site with late transition elements.

  • River morphology redistributes potentially toxic elements in acid mine drainage-impacted river sediments: Evidence, causes, and implications

    • River morphology plays a vital role in the transport of substance within them. However, our understanding of how natural and artificial morphologies redistribute different potentially toxic elements in acid mine drainage (AMD)-contaminated rivers remains poor. In this study, we linked morphological river features and physicochemical sediment characteristics to trace the redistribution of various potentially toxic elements and elucidate their implications for remediating rivers prone to AMD pollution. A dense network of sediment/soil samples was collected from different river morphological units, such as channels, dam reservoirs, pools, floodplain sandbars and wetlands in an AMD-impacted river. The analyses showed that the contaminant levels in channel generally decreased downstream from the headwater mine site, however, local fluctuations in certain areas were observed due to the trapping effect of various dams along the river. The As and Pb concentrations were higher at floodplain sandbars, while river channels exhibited higher Cd and Zn contamination. The concentrations and geochemical fractions of As, Cd, Cu, Pb and Zn in sediment/soil cores from sandbar and river channel also varied. Additionally, structural equation modeling analysis indicated that spatial variations in contaminant distributions were directly affected by physicochemical properties (such as the soil/sediment Fe, Zn, and S concentrations, and pH), which are indirectly affected by river morphology. The diverse morphology of the river redistributed AMDderived contaminants and could be used to identify contamination hotspots. Our analyses suggested that the feasibility and efficiency of previously proposed countermeasures varied for contaminants in different geomorphological units. In river channels, As uptake from sediments by aquatic plants may be less efficient than Cd, Cu, and Zn uptake due to its lower bioavailability. Moreover, vegetation prevented contaminant enriched soil particle erosion more than it aided in the phytoremediation of As- and Pb-contaminated sandbars. Thus the finding of this study provide a theoretical foundation for further studies on the transport and storage of AMDderived contaminants in similar rivers, along with the development of targeted remediation methods.
