  • Guidelines for the use and interpretation of assays for monitoring autophagy
  • 2012
  • Ingår i: Autophagy. - : Informa UK Limited. - 1554-8635 .- 1554-8627. ; 8:4, s. 445-544
  • Forskningsöversikt (refereegranskat)abstract
    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Formation of dense partonic matter in relativistic nucleus-nucleus collisions at RHIC: Experimental evaluation by the PHENIX Collaboration
  • 2005
  • Ingår i: Nuclear Physics, Section A. - : Elsevier BV. - 0375-9474. ; 757:1-2, s. 184-283
  • Forskningsöversikt (refereegranskat)abstract
    • Extensive experimental data from high-energy nucleus-nucleus collisions were recorded using the PHENIX detector at the Relativistic Heavy Ion Collider (RHIC). The comprehensive set of measurements from the first three years of RHIC operation includes charged particle multiplicities, transverse energy, yield ratios and spectra of identified hadrons in a wide range of transverse momenta (PT), elliptic flow, two-particle correlations, nonstatistical fluctuations, and suppression of particle production at high PT. The results are examined with an emphasis on implications for the formation of a new state of dense matter. We find that the state of matter created at RHIC cannot be described in terms of ordinary color neutral hadrons.

  • Minimum Information about a Biosynthetic Gene cluster
  • 2015
  • Ingår i: Nature Chemical Biology. - : Springer Science and Business Media LLC. - 1552-4450 .- 1552-4469. ; 11:9, s. 625-631
  • Forskningsöversikt (refereegranskat)abstract
    • A wide variety of enzymatic pathways that produce specialized metabolites in bacteria, fungi and plants are known to be encoded in biosynthetic gene clusters. Information about these clusters, pathways and metabolites is currently dispersed throughout the literature, making it difficult to exploit. To facilitate consistent and systematic deposition and retrieval of data on biosynthetic gene clusters, we propose the Minimum Information about a Biosynthetic Gene cluster (MIBiG) data standard.

  • Inland Water Greenhouse Gas Budgets for RECCAP2: 1. State-Of-The-Art of Global Scale Assessments
  • 2023
  • Ingår i: Global Biogeochemical Cycles. - : AMER GEOPHYSICAL UNION. - 0886-6236 .- 1944-9224. ; 37:5
  • Forskningsöversikt (refereegranskat)abstract
    • Inland waters are important emitters of the greenhouse gasses (GHGs) carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O) to the atmosphere. In the framework of the 2nd phase of the REgional Carbon Cycle Assessment and Processes (RECCAP-2) initiative, we review the state of the art in estimating inland water GHG budgets at global scale, which has substantially advanced since the first phase of RECCAP nearly 10 years ago. The development of increasingly sophisticated upscaling techniques, including statistical prediction and process-based models, allows for spatially explicit estimates that are needed for regionalized assessments of continental GHG budgets such as those established for RECCAP. A few recent estimates also resolve the seasonal and/or interannual variability in inland water GHG emissions. Nonetheless, the global-scale assessment of inland water emissions remains challenging because of limited spatial and temporal coverage of observations and persisting uncertainties in the abundance and distribution of inland water surface areas. To decrease these uncertainties, more empirical work on the contributions of hot-spots and hot-moments to overall inland water GHG emissions is particularly needed.

  • A review of femtosecond laser-induced emission techniques for combustion and flow field diagnostics
  • 2019
  • Ingår i: Applied Sciences (Switzerland). - : MDPI AG. - 2076-3417. ; 9:9
  • Forskningsöversikt (refereegranskat)abstract
    • The applications of femtosecond lasers to the diagnostics of combustion and flow field have recently attracted increasing interest. Many novel spectroscopic methods have been developed in obtaining non-intrusive measurements of temperature, velocity, and species concentrations with unprecedented possibilities. In this paper, several applications of femtosecond-laser-based incoherent techniques in the field of combustion diagnostics were reviewed, including two-photon femtosecond laser-induced fluorescence (fs-TPLIF), femtosecond laser-induced breakdown spectroscopy (fs-LIBS), filament-induced nonlinear spectroscopy (FINS), femtosecond laser-induced plasma spectroscopy (FLIPS), femtosecond laser electronic excitation tagging velocimetry (FLEET), femtosecond laser-induced cyano chemiluminescence (FLICC), and filamentary anemometry using femtosecond laser-extended electric discharge (FALED). Furthermore, prospects of the femtosecond-laser-based combustion diagnostic techniques in the future were analyzed and discussed to provide a reference for the relevant researchers.

  • A Review of the Distribution of Antibiotics in Water in Different Regions of China and Current Antibiotic Degradation Pathways
  • 2021
  • Ingår i: Frontiers in Environmental Science. - : Frontiers Media SA. - 2296-665X. ; 9
  • Forskningsöversikt (refereegranskat)abstract
    • Antibiotic pollution is becoming an increasingly serious threat in different regions of China. The distribution of antibiotics in water sources varies significantly in time and space, corresponding to the amount of antibiotics used locally. The main source of this contamination in the aquatic environment is wastewater from antibiotic manufacturers, large scale animal farming, and hospitals. In response to the excessive antibiotic contamination in the water environment globally, environmentally friendly alternatives to antibiotics are being developed to reduce their use. Furthermore, researchers have developed various antibiotic treatment techniques for the degradation of antibiotics, such as physical adsorption, chemical oxidation, photodegradation, and biodegradation. Among them, biodegradation is receiving increasing attention because of its low cost, ease of operation, and lack of secondary pollution. Antibiotic degradation by enzymes could become the key strategy of management of antibiotics pollution in the environment in future. This review summarizes research on the distribution of antibiotics in China's aquatic environments and different techniques for the degradation of antibiotics. Special attention is paid to their degradation by various enzymes. The adverse effects of the pollutants and need for more effective monitoring and mitigating pollution are also highlighted.

  • Ionic liquids as a new cornerstone to support hydrogen energy
  • 2023
  • Ingår i: Green Chemistry. - : Royal Society of Chemistry. - 1463-9262 .- 1463-9270. ; 25:13, s. 4981-4994
  • Forskningsöversikt (refereegranskat)abstract
    • As a fuel or energy carrier, hydrogen has been identified as a key way to decarbonize electricity, industry, transportation, and heating sectors. Hydrogen can be produced by a variety of methods, among which water electrolysis driven by renewable energy is sustainable and nearly carbon-free. To use hydrogen widely, storage and transportation over long distances are another key issue. Apart from storage at high pressure and low temperature, hydrogen can be stored in organic compounds via chemical bonding under relatively mild conditions. Efficient utilization of hydrogen includes hydrogen fuel cells as an alternative to internal combustion engines. From the above scenarios, catalysis and reaction media are the key factors for realizing hydrogen energy implementation. Ionic liquids (ILs) offer new opportunities due to their tunable functional groups, low vapor pressure, and stable structures as additives, solvents, and charge transfer materials. ILs are known to produce solid catalysts with controllable properties, decorate solid catalysts with modified electrons and geometric structures, and serve as electrolytes and hydrogen storage media. This review summarizes and recaps the recent progress in how ILs act as a cornerstone to support the production, storage, and utilization of hydrogen. Furthermore, critical challenges and future research directions of ILs in hydrogen energy applications are also outlined.

  • Preparation, characterization, evaluation and mechanistic study of organic polymer nano-photocatalysts for solar fuel production
  • 2022
  • Ingår i: Chemical Society Reviews. - : ROYAL SOC CHEMISTRY. - 0306-0012 .- 1460-4744. ; 51:16, s. 6909-6935
  • Forskningsöversikt (refereegranskat)abstract
    • Production of renewable fuels from solar energy and abundant resourses, such as water and carbon dioxide, via photocatalytic reactions is seen as a promising strategy to adequately address the climate challenge. Photocatalytic systems based on organic polymer nanoparticles (PNPs) are seen as one avenue to transform solar energy into hydrogen and other solar fuels. Semiconducting PNPs are light-harvesting materials with exceptional optical properties, photostability, low cost and low cytotoxity, whose performance surpasses conventional organic dyes and inorganic semiconductors. This review introduces the optimization strategies for the preparation methods of PNP via cocatalyst loading and morphology tuning. We present an analysis on how the preparative methods will impact the physico-chemical properties of these materials, and thus the catalytic activity. A list of experimental techniques is presented for characterization of the physico-chemical properties (optical, morphological, electrochemical and catalytic properties) of PNPs. We provide detailed analysis of PNP photochemistry during photocatalysis with focus on the mechanistic understanding of processes of internal charge generation and transport to the catalyst. This tutorial review provides the reader with the guidelines on current strategies used to optimize PNP performance highlighting the future directions of polymer nano-photocatalysts development.

  • Development and applicability analyses of ADS-4 entrainment model in large advanced PWR
  • 2020
  • Ingår i: Nuclear Engineering and Design. - : ELSEVIER SCIENCE SA. - 0029-5493 .- 1872-759X. ; 356
  • Forskningsöversikt (refereegranskat)abstract
    • Two new models of liquid entrainment through a vertical tee branch are proposed based on theoretical analyses and experimental data, which can well predict the test results of the prototype facility of CAP1400 and the scaled down facility of AP1000. Moreover, in order to verify the applicability of the new entrainment models, these models are implemented to RELAP5 through the safety analyses with a 2 in. cold leg SBLOCA in AP1000 comparing with the original RELAP5. The contrastive results show that the RELAP5 with proposed entrainment models has more reasonable results. In addition, the modified RELAP5 can calculate less primary coolant inventory which is more conservative. Besides, the less coolant inventory is accord with the beyond design basis accident test of APEX facility (Advanced Plant Experiment). Therefore, the proposed entrainment models are more appropriate for the ADS-4 entrainment safety analyses in large advanced PWR.
