  • The human secretome

    • The proteins secreted by human cells (collectively referred to as the secretome) are important not only for the basic understanding of human biology but also for the identification of potential targets for future diagnostics and therapies. Here, we present a comprehensive analysis of proteins predicted to be secreted in human cells, which provides information about their final localization in the human body, including the proteins actively secreted to peripheral blood. The analysis suggests that a large number of the proteins of the secretome are not secreted out of the cell, but instead are retained intracellularly, whereas another large group of proteins were identified that are predicted to be retained locally at the tissue of expression and not secreted into the blood. Proteins detected in the human blood by mass spectrometry-based proteomics and antibody-based immuno-assays are also presented with estimates of their concentrations in the blood. The results are presented in an updated version 19 of the Human Protein Atlas in which each gene encoding a secretome protein is annotated to provide an open-access knowledge resource of the human secretome, including body-wide expression data, spatial localization data down to the single-cell and subcellular levels, and data about the presence of proteins that are detectable in the blood.

    • Global classification of the human proteins with regards to spatial expression patterns across organs and tissues is important for studies of human biology and disease. Here, we used a quantitative transcriptomics analysis (RNA-Seq) to classify the tissue-specific expression of genes across a representative set of all major human organs and tissues and combined this analysis with antibody- based profiling of the same tissues. To present the data, we launch a new version of the Human Protein Atlas that integrates RNA and protein expression data corresponding to 80% of the human protein-coding genes with access to the primary data for both the RNA and the protein analysis on an individual gene level. We present a classification of all human protein-coding genes with regards to tissue-specificity and spatial expression pattern. The integrative human expression map can be used as a starting point to explore the molecular constituents of the human body.

    • The investigated alumina forming FeNiCrAl model alloy shows protective oxidation behavior in dry and humid environment at 900 A degrees C. Hence, this type of alloy may replace conventional chromia forming austenitic alloys in aggressive oxidizing/reducing environments. A detailed investigation of the oxide scale development reveals a complex initial scale development. Firstly, at alloy grain boundaries, a thin Al rich oxide forms which is replaced by transient alumina platelets in dry and equiaxed alpha-Al2O3 crystallites in humid atmosphere. The scale at alloy grain centers develops via a layered scale of external chromia:Fe/Ni metal inclusions:internal alumina to a layered external spinel:internal alumina scale in dry atmosphere. In humid condition an additional oxide feature appears on the center of large alloy grains i.e. thick oxide protrusions. Despite the initially different phase compositions a continuous protective alpha-Al2O3 scale forms both atmospheres.

  • A pathology atlas of the human cancer transcriptome

    • Cancer is one of the leading causes of death, and there is great interest in understanding the underlying molecular mechanisms involved in the pathogenesis and progression of individual tumors. We used systems-level approaches to analyze the genome-wide transcriptome of the protein-coding genes of 17 major cancer types with respect to clinical outcome. A general pattern emerged: Shorter patient survival was associated with up-regulation of genes involved in cell growth and with down-regulation of genes involved in cellular differentiation. Using genome-scale metabolic models, we show that cancer patients have widespread metabolic heterogeneity, highlighting the need for precise and personalized medicine for cancer treatment. All data are presented in an interactive open-access database (www.proteinatlas.org/pathology) to allow genome-wide exploration of the impact of individual proteins on clinical outcomes.

    • Resolving the molecular details of proteome variation in the different tissues and organs of the human body will greatly increase our knowledge of human biology and disease. Here, we present a map of the human tissue proteome based on an integrated omics approach that involves quantitative transcriptomics at the tissue and organ level, combined with tissue microarray-based immunohistochemistry, to achieve spatial localization of proteins down to the single-cell level. Our tissue-based analysis detected more than 90% of the putative protein-coding genes. We used this approach to explore the human secretome, the membrane proteome, the druggable proteome, the cancer proteome, and the metabolic functions in 32 different tissues and organs. All the data are integrated in an interactive Web-based database that allows exploration of individual proteins, as well as navigation of global expression patterns, in all major tissues and organs in the human body.

    • Renewable energy integration, electric vehicles, grid modernization, and energy security are becoming increasingly important for societies worldwide. Well-educated electric power system engineers are essential to make this happen. This comparison involves two master's programs in two different regions: the Global South and Global North. Both programs are two-year master's programs based on a series of courses in electric power systems and supporting subjects. Individual and contextual strengths and weaknesses for each program are highlighted, compared, and discussed, including the importance of adapting the programs to local contexts while addressing global challenges in power systems. However, considering the global labor market and the uncertain future, and recognizing that engineers should be professionals for several decades, the programs should not be overly tied to the local context. In conclusion, recommendations for improvement are provided for both programs based on the strengths of the other program and their shared weaknesses. For example, the Global South program should enhance specialization and collaborate more with the industry, while the Global North program should promote interaction with other programs, include more aspects related to societal impact, and provide better guidance for course selection.

    • Separation of trivalent actinoid (An(III)) and lanthanoid (Ln(III)) ions is extremely challenging due to their similar ionic radii and chemical properties. Poly-aromatic nitrogen compounds acting as tetradentate chelating ligands to the metal ions in the extraction, have the ability to sufficiently separate An(III) from Ln(III). One of these compounds, 6,6'-bis(5,5,8,8-tetramethyl-5,6,7,8-tetrahydro-benzol[1,2,4]triazin-3-y l)-[2,2]bipyridine, CyMe4-BTBP, has proven to be resistant towards acidic environments and strong radiation from radioactive decomposition. EXAFS studies of the dicomplexes of CyMe4-BTBP with americium(III) and europium(III) in nitrobenzene, cyclohexanone, 1-hexanol, 1-octanol and malonamide (DMDOHEMA) in 1-octanol have been carried out to get a deeper understanding of the parameters responsible for the separation. The predominating complexes independent of solvent used are [Am(CyMe4-BTBP)(2)(NO3)](2+) and [Eu(CyMe4-BTBP)(2)](3+), respectively, which are present as outer-sphere ion-pairs with nitrate ions in the studied solvents with low relative permittivity. The presence of a nitrate ion in the first coordination sphere of the americium(III) complex compensates the charge density of the complex considerably in comparison when only outer-sphere ion-pairs are formed as for the [Eu(CyMe4-BTBP)(2)](3+) complex. The stability and solubility of a complex in a solvent with low relative permittivity increase with decreasing charge density. The [Am(CyMe4-BTBP)(2)(NO3)](2+) complex will therefore be increasingly soluble and stabilized over the [Eu(CyMe4-BTBP)(2)](3+) complex in solvents with decreasing relative permittivity of the solvent. The separation of americium(III) from europium(III) with CyMe4-BTBP as extraction agent will increase with decreasing relative permittivity of the solvent, and thereby also with decreasing solubility of CyMe4-BTBP. The choice of solvent is therefore a balance of a high separation factor and sufficient solubility of the CyMe4-BTBP ligand.

    • The NaCoSOFC project is focused on the development of nano coatings for SOFC interconnects. The project is sponsored by the Nordic Top Level Research Initiative and has four project partners: Sandvik Materials Technology which is producing coated interconnects, Chalmers University of Technology and the University of Oslo that characterize samples with respect to e.g. corrosion, Cr evaporation and ASR as well as Topsoe Fuel Cell that is testing the developed interconnects in its stacks. The developed coatings are based on a combination of Co with RE elements and exhibit high corrosion resistance, 10 fold decrease in Cr evaporation and ASR values that are approximately 50% of the uncoated material.
