  • Validated and longitudinally stable asthma phenotypes based on cluster analysis of the ADEPT study

    • Background: Asthma is a disease of varying severity and differing disease mechanisms. To date, studies aimed at stratifying asthma into clinically useful phenotypes have produced a number of phenotypes that have yet to be assessed for stability and to be validated in independent cohorts. The aim of this study was to define and validate, for the first time ever, clinically driven asthma phenotypes using two independent, severe asthma cohorts: ADEPT and U-BIOPRED. Methods: Fuzzy partition-around-medoid clustering was performed on pre-specified data from the ADEPT participants (n = 156) and independently on data from a subset of U-BIOPRED asthma participants (n = 82) for whom the same variables were available. Models for cluster classification probabilities were derived and applied to the 12-month longitudinal ADEPT data and to a larger subset of the U-BIOPRED asthma dataset (n = 397). High and low type-2 inflammation phenotypes were defined as high or low Th2 activity, indicated by endobronchial biopsies gene expression changes downstream of IL-4 or IL-13. Results: Four phenotypes were identified in the ADEPT (training) cohort, with distinct clinical and biomarker profiles. Phenotype 1 was "mild, good lung function, early onset", with a low-inflammatory, predominantly Type-2, phenotype. Phenotype 2 had a "moderate, hyper-responsive, eosinophilic" phenotype, with moderate asthma control, mild airflow obstruction and predominant Type-2 inflammation. Phenotype 3 had a "mixed severity, predominantly fixed obstructive, non-eosinophilic and neutrophilic" phenotype, with moderate asthma control and low Type-2 inflammation. Phenotype 4 had a "severe uncontrolled, severe reversible obstruction, mixed granulocytic" phenotype, with moderate Type-2 inflammation. These phenotypes had good longitudinal stability in the ADEPT cohort. They were reproduced and demonstrated high classification probability in two subsets of the U-BIOPRED asthma cohort. Conclusions: Focusing on the biology of the four clinical independently-validated easy-to-assess ADEPT asthma phenotypes will help understanding the unmet need and will aid in developing tailored therapies. As such, it has the potential for a particle physics program with a unique reach and which is complementary to that available at other facilities. This paper describes proposed particle physics activities for the ESS. These encompass the exploitation of both the neutrons and neutrinos produced at the ESS for high precision (sensitivity) measurements (searches).

  • Nuclear astrophysics with radioactive ions at FAIR

    • The nucleosynthesis of elements beyond iron is dominated by neutron captures in the s and r processes. However, 32 stable, proton-rich isotopes cannot be formed during those processes, because they are shielded from the s-process flow and r-process beta-decay chains. These nuclei are attributed to the p and rp process. For all those processes, current research in nuclear astrophysics addresses the need for more precise reaction data involving radioactive isotopes. Depending on the particular reaction, direct or inverse kinematics, forward or time-reversed direction are investigated to determine or at least to constrain the desired reaction cross sections. The Facility for Antiproton and Ion Research (FAIR) will offer unique, unprecedented opportunities to investigate many of the important reactions. The high yield of radioactive isotopes, even far away from the valley of stability, allows the investigation of isotopes involved in processes as exotic as the r or rp processes.

    • Molybdenum sulfide structures, particularly amorphous MoS3 nanoparticles, are promising materials in the search for cost-effective and scalable water-splitting catalysts. Ex situ observations show that the nanoparticles exhibit a composition change from MoS3 to defective MoS2 when subjected to hydrogen evolution reaction (HER) conditions, raising questions regarding the active surface sites taking part in the reaction. We tracked the in situ transformation of amorphous MoS3 nanoparticles under HER conditions through ambient pressure X-ray photoelectron spectroscopy and performed density functional theory studies of model MoSx systems. We demonstrate that, under operating conditions, surface sites are converted from MoS3 to MoS2 in a gradual manner and that the electrolytic current densities are proportional to the extent of the transformation. We also posit that it is the MoS2 edge-like sites that are active during HER, with the high activity of the catalyst being attributed to the increase in surface MoS2 edge-like sites after the reduction of MoS3 sites.

    • The concept of bonding and antibonding orbitals is fundamental in chemistry. The population of those orbitals and the energetic difference between the two reflect the strength of the bonding interaction. Weakening the bond is expected to reduce this energetic splitting, but the transient character of bond-activation has so far prohibited direct experimental access. Here we apply time-resolved soft X-ray spectroscopy at a free electron laser to directly observe the decreased bonding antibonding splitting following bond-activation using an ultrashort optical laser pulse.

    • A passive approximation problem is formulated where the target function is an arbitrary complex-valued continuous function defined on an approximation domain consisting of a finite union of closed and bounded intervals on the real axis. The norm used is a weighted L-p-norm where 1 <= p <= infinity. The approximating functions are Herglotz functions generated by a measure with Holder continuous density in an arbitrary neighborhood of the approximation domain. Hence, the imaginary and the real parts of the approximating functions are Holder continuous functions given by the density of the measure and its Hilbert transform, respectively. In practice, it is useful to employ finite B-spline expansions to represent the generating measure. The corresponding approximation problem can then be posed as a finite-dimensional convex optimization problem which is amenable for numerical solution. A constructive proof is given here showing that the convex cone of approximating functions generated by finite uniform B-spline expansions of fixed arbitrary order (linear, quadratic, cubic, etc.) is dense in the convex cone of Herglotz functions which are locally Holder continuous in a neighborhood of the approximation domain, as mentioned above. As an illustration, typical physical application examples are included regarding the passive approximation and optimization of a linear system having metamaterial characteristics, as well as passive realization of optimal absorption of a dielectric small sphere over a finite bandwidth.

  • Temperature-Independent Nuclear Quantum Effects on the Structure of Water

    • Nuclear quantum effects (NQEs) have a significant influence on the hydrogen bonds in water and aqueous solutions and have thus been the topic of extensive studies. However, the microscopic origin and the corresponding temperature dependence of NQEs have been elusive and still remain the subject of ongoing discussion. Previous x-ray scattering investigations indicate that NQEs on the structure of water exhibit significant temperature dependence [Phys. Rev. Lett. 94, 047801 (2005)]. Here, by performing wide-angle x-ray scattering of H2O and D2O droplets at temperatures from 275 K down to 240 K, we determine the temperature dependence of NQEs on the structure of water down to the deeply supercooled regime. The data reveal that the magnitude of NQEs on the structure of water is temperature independent, as the structure factor of D2O is similar to H2O if the temperature is shifted by a constant 5 K, valid from ambient conditions to the deeply supercooled regime. Analysis of the accelerated growth of tetrahedral structures in supercooled H2O and D2O also shows similar behavior with a clear 5 K shift. The results indicate a constant compensation between NQEs delocalizing the proton in the librational motion away from the bond and in the OH stretch vibrational modes along the bond. This is consistent with the fact that only the vibrational ground state is populated at ambient and supercooled conditions.

  • A trinity of sense : Using biomass in the transport sector for climate change mitigation

    • This thesis analyses two strategies for decreasing anthropogenic carbon dioxide (CO2) emissions: to capture and store CO2, and to increase the use of biomass. First, two concepts for CO2 capture with low capture penalties are evaluated. The concepts are an integrated gasification combined cycle where the oxygen is supplied by a membrane reactor, and a hybrid cycle where the CO2 is captured at elevated pressure. Although the cycles have comparatively high efficiencies and low penalties, they illustrate the inevitable fact that capturing CO2 will always induce significant efficiency penalties. Other strategies are also needed if CO2 emissions are to be forcefully decreased. An alternative is increased use of biomass, which partially could be used for production of motor fuels (biofuels). This work examines arguments for directing biomass to the transport sector, analyses how biofuels (and also some other means) may be used to reduce CO2 emissions and increase security of motor fuel supply. The thesis also explores the possibility of reducing CO2 emissions by comparatively easy and cost-efficient CO2 capture from concentrated CO2 streams available in some types of biofuel plants. Many conclusions of the thesis could be associated with either of three meanings of the word sense: First, there is reason in biofuel production – since it e.g. reduces oil dependence. From a climate change mitigation perspective, however, motor fuel production is often a CO2-inefficient use of biomass, but the thesis explores how biofuels’ climate change mitigation effects may be increased by introducing low-cost CO2 capture. Second, the Swedish promotion of biofuels appears to have been governed more by a feeling for attaining other goals than striving for curbing climate change. Third, it seems to have been the prevalent opinion among politicians that the advantages of biofuels – among them their climate change mitigation benefits – are far greater than the disadvantages and that they should be promoted. Another conclusion of the thesis is that biofuels alone are not enough to drastically decrease transport CO2 emissions; a variety of measures are needed such as fuels from renewable electricity and improvements of vehicle fuel economy.

  • Strain field inhomogeneities and stiffness changes in GMT containing voids

    • During compression moulding of glass mat thermoplastics (GMT), voids may form. However, it is not clear whether voids are as critical to mechanical performance in GMT as in thermoset composites. The present investigation also considers the general problem of damage mechanisms in GMT. Conventional tensile tests, acoustic emission, a stiffness degradation test and a speckle technique for strain field measurements are used as well as optical microscopy of polished cross-sections. The void content (up to 5%) does not significantly influence the strength or stiffness degradation process. The reason is the large inhomogeneity of the strain fields in GMT. Failure occurs in locally soft regions and void effects are of secondary importance. Details of the failure process are discussed, emphasising the large local strains in matrix-rich regions.
