  • Genetic Imbalance Is Associated With Functional Outcome After Ischemic Stroke

    • Background and Purpose-We sought to explore the effect of genetic imbalance on functional outcome after ischemic stroke (IS). Methods-Copy number variation was identified in high-density single-nucleotide polymorphism microarray data of IS patients from the CADISP (Cervical Artery Dissection and Ischemic Stroke Patients) and SiGN (Stroke Genetics Network)/ GISCOME (Genetics of Ischaemic Stroke Functional Outcome) networks. Genetic imbalance, defined as total number of protein-coding genes affected by copy number variations in an individual, was compared between patients with favorable (modified Rankin Scale score of 0-2) and unfavorable (modified Rankin Scale score of = 3) outcome after 3 months. Subgroup analyses were confined to patients with imbalance affecting ohnologs-a class of dose-sensitive genes, or to those with imbalance not affecting ohnologs. The association of imbalance with outcome was analyzed by logistic regression analysis, adjusted for age, sex, stroke subtype, stroke severity, and ancestry. Results-The study sample comprised 816 CADISP patients (age 44.2 +/- 10.3 years) and 2498 SiGN/GISCOME patients (age 67.7 +/- 14.2 years). Outcome was unfavorable in 122 CADISP and 889 SiGN/GISCOME patients. Multivariate logistic regression analysis revealed that increased genetic imbalance was associated with less favorable outcome in both samples (CADISP: P=0.0007; odds ratio=0.89; 95% CI, 0.82-0.95 and SiGN/GISCOME: P=0.0036; odds ratio=0.94; 95% CI, 0.91-0.98). The association was independent of age, sex, stroke severity on admission, stroke subtype, and ancestry. On subgroup analysis, imbalance affecting ohnologs was associated with outcome (CADISP: odds ratio=0.88; 95% CI, 0.80-0.95 and SiGN/GISCOME: odds ratio=0.93; 95% CI, 0.89-0.98) whereas imbalance without ohnologs lacked such an association. Conclusions-Increased genetic imbalance was associated with poorer functional outcome after IS in both study populations. Subgroup analysis revealed that this association was driven by presence of ohnologs in the respective copy number variations, suggesting a causal role of the deleterious effects of genetic imbalance.

  • Software-based data acquisition and processing for neutron detectors at European Spallation Source-early experience from four detector designs

    • European Spallation Source (ESS) will deliver neutrons at high flux for use in diverse neutron scattering techniques. The neutron source facility and the scientific instruments will be located in Lund, and the Data Management and Software Centre (DMSC), in Copenhagen. A number of detector prototypes are being developed at ESS together with its European in-kind partners, for example: SoNDe, Multi-Grid, Multi-Blade and Gd-GEM. These are all position sensitive detectors but use different techniques for the detection of neutrons. Except for digitization of electronics readout, all neutron data is anticipated to be processed in software. This provides maximum flexibility and adaptability and allows deep inspection of the raw data for commissioning which will reduce the risk of starting up new detector technologies. But it also requires development of high performance software processing pipelines and optimized and scalable processing algorithms. This report provides a description of the ESS system architecture for the neutron data path. Special focus is on the interface between the detectors and DMSC which is based on UDP over Ethernet links. The report also describes the software architecture for detector data processing and the tools we have developed, which have proven very useful for efficient early experimentation, and can be run on a single laptop. Processing requirements for the SoNDe, Multi-Grid, Multi-Blade and Ge-GEM detectors are presented and compared to event processing rates archived so far.

    • Resistive bulk Micromegas chambers, produced at CERN, have been installed at the new CERN Gamma Irradiation Facility (GIF++) in order to study the effects of ageing and to evaluate the detector behaviour under high irradiation. The chambers have an active area of 10×10cm2, strip pitch of 400μm and an amplification gap of 128μm. We present the detector performance as a function of the background rate of up to 20MHz/cm2.

  • In-field monitoring of cleaning efficiency in waste water treatment plants using two phenol-sensitive biosensors

    • Two amperometric biosensors based on the enzymes cellobiose dehydrogenase (CDH) and quinoprotein-dependent glucose dehydrogenase (GDH), have been applied for monitoring the phenolic content in water samples, collected at different stages of a waste water treatment process, thus representing different cleaning levels of two waste water treatment plants (WWTPs). The biosensor measurements were performed in-field, compared with the results obtained by liquid chromatography-mass spectrometry and were further correlated with the cleaning efficiencies of the WWTPs. The effect of several potentially interfering compounds on the sensor response was also studied. The general purpose of the study was to evaluate the potential use of biosensors, not as quantitative tools for phenol analysis, but rather as screening tools indicating a certain trend, i.e. compounds present or not present, and potential correlation with sample toxicity. It was found that the biosensors and LC-MS results were not quantitatively comparable, however, both sensors could follow the decrease of the phenol content from the influent, primary treated and effluent waters. In addition, the correlation between biosensor inhibition and sample toxicity is discussed.

  • First measurements with new high-resolution gadolinium-GEM neutron detectors

    • European Spallation Source instruments like the macromolecular diffractometer (NMX) require an excellent neutron detection efficiency, high-rate capabilities, time resolution, and an unprecedented spatial resolution in the order of a few hundred micrometers over a wide angular range of the incoming neutrons. For these instruments solid converters in combination with Micro Pattern Gaseous Detectors (MPGDs) are a promising option. A GEM detector with gadolinium converter was tested on a cold neutron beam at the IFE research reactor in Norway. The mu TPC analysis, proven to improve the spatial resolution in the case of B-10 converters, is extended to gadolinium based detectors. For the first time, a Gd-GEM was successfully operated to detect neutrons with a measured efficiency of 11.8% at a wavelength of 2 angstrom and a position resolution better than 250 mu m.

  • Multi-GEM detectors in high particle fluxes

    • Gaseous Electron Multipliers (GEM) are well known for stable operation at high particle fluxes. We present a study of the intrinsic limits of GEM detectors when exposed to veiy high particle fluxes of the order of MHz/mm2. We give an interpretation to the variations of the effective gain, which, as a function of the particle flux, first increases and then decreases. Wre also discuss the reduction of the ion back-flow with increasing flux. We present measurements and simulations of a triple GEM detector, describing its behaviour in terms of accumulation of positive ions that results in changes of the transfer fields and the amplification fields. The behaviour is expected to be common to all multi-stage amplification devices where the efficiency of transferring the electrons from one stage to the next one is not 100%.
