    • The tribocorrosion behavior of titanium-based alloys is of significant interest as bio-implant materials. Bare alloys may not offer enough resistance to tribocorrosion, so coatings could be used to improve their performance. An important biomedical alloy, Ti-13Nb-13Zr, and a newly developed β titanium alloy called "Gum metal" (Ti-23%Ni-0.7%Ta-2%Zr-1 %O2) were used as substrates in the current work. Both were coated with conventional and nano-ceramic materials of Al2O3-TiO2. Bilayered coatings of ZrO2+Al2O3-13%TiO2 were also applied to the Ti-13Nb-13Zr using plasma spray. The coatings on Ti-13Nb-13Zr were applied using plasma spray, whereas that on the Gum metal was applied by a detonation gun (D-Gun). Surface morphology was characterized using a scanning electron microscope (SEM). Tribocorrosion experiments were performed in salt water using a linear reciprocating ball-on-plate tribometer with an aluminum ball as the slider. The nano particles are embedded in the fully melted splats and offered better crack propagation resistance. The high velocity of the D-Gun process resulted in a higher volume fraction of the embedded nano particles and produced substantial improvement in wear resistance relative to the air-plasma-sprayed coating. The conventional coating, with its higher porosity, exhibited a high corrosion rate compared to nano coating. The D-Gun coating, with its lower porosity, had a higher corrosion resistance than the plasma-sprayed coating, but bilayered plasma-sprayed coating showed even higher corrosion resistance, owing to its dense microstructure. Open-circuit potential measurements before and during tribocorrosion showed that the bilayered plasma-sprayed coating had better tribocorrosion resistance than the other coatings. Electrochemical impedance spectroscopy indicated stable impedance values for the bilayered plasma-sprayed coating before and after tribocorrosion.     • OBJECTIVE: To evaluate the relationship between A1C and glucose therapy intensification (GTI) in patients with diabetes mellitus (DM) hospitalized for acute myocardial infarction (AMI). RESEARCH DESIGN AND METHODS: A1C was measured as part of routine care (clinical A1C) or in the core laboratory (laboratory A1C, results unavailable to clinicians). GTI predictors were identified using hierarchical Poisson regression. RESULTS: Of 1,274 patients, 886 (70%) had clinical A1C and an additional 263 had laboratory A1C measured. Overall, A1C was <7% in 419 (37%), 7-9% in 415 (36%), and >9% in 315 patients (27%). GTI occurred in 31% of patients and was more frequent in those with clinical A1C both before (34 vs. 24%, P < 0.001) and after multivariable adjustment (relative risk 1.34 [95% CI 1.12-1.62] vs. no clinical A1C). CONCLUSIONS: Long-term glucose control is poor in most AMI patients with DM, but only a minority of patients undergo GTI at discharge. Inpatient A1C assessment is strongly associated with intensification of glucose-lowering therapy.

  • Differential Role of HAMP-like Linkers in Regulating the Functionality of the Group III Histidine Kinase DhNik1p

    • Nik1 orthologs are sensor kinases that function upstream of the high osmolarity glycerol/p38 MAPK pathway in fungi. They contain a poly-HAMP module at their N terminus, which plays a pivotal role in osmosensing as well as fungal death upon exposure to fludioxonil. DhNik1p is a typical member of this class that contains five HAMP domains and four HAMP-like linkers. We investigated the contribution of each of the HAMP-like linker regions to the functionality of DhNik1p and found that the HAMP4b linker was essential as its deletion resulted in the complete loss of activity. Replacement of this linker with flexible peptide sequences did not restore DhNik1p activity. Thus, the HAMP-like sequence and possibly structural features of this linker region are indispensable for the kinase activity of DhNik1p. To gain insight into the global shape of the poly-HAMP module in DhNik1p (HAMP1-5), multi-angle laser light and small angle x-ray scattering studies were carried out. Those data demonstrate that the maltose-binding protein-tagged HAMP1-5 protein exist as a dimer in solution with an elongated shape of maximum linear dimension similar to 365 angstrom. Placement of a sequence similarity based model of the HAMP1-5 protein inside experimental data-based models showed how two chains of HAMP1-5 are entwined on each other and the overall structure retained a periodicity. Normal mode analysis of the structural model is consistent with the H4b linker being a key to native-like collective motion in the protein. Overall, our shape-function studies reveal how different elements in the HAMP1-5 structure mediate its function.

  • Chymase Level Is a Predictive Biomarker of Dengue Hemorrhagic Fever in Pediatric and Adult Patients

    • Background. Most patients with dengue experience mild disease, dengue fever (DF), while few develop the life-threatening diseases dengue hemorrhagic fever (DHF) or dengue shock syndrome (DSS). No laboratory tests predict DHF or DSS. We evaluated whether the serum chymase level can predict DHF or DSS in adult and pediatric patients and the influence of preexisting conditions (PECs) on chymase levels. Methods. Serum chymase levels were measured in patients presenting with undifferentiated fever to hospitals in Colombo District, Sri Lanka. The value of serum the chymase concentration and clinical signs and symptoms as predictors of DHF and/or DSS was evaluated by multivariate analysis. We assessed the influence of age, PECs, and day after fever onset on the robustness of the chymase level as a biomarker for DHF and/or DSS. Results. An elevated chymase level in acute phase blood samples was highly indicative of later diagnosis of DHF or DSS for pediatric and adult patients with dengue. No recorded PECs prevented an increase in the chymase level during DHF. However, certain PECs (obesity and cardiac or lung-associated diseases) resulted in a concomitant increase in chymase levels among adult patients with DHF. Conclusions. These results show that patients with acute dengue who present with high levels of serum chymase consistently are at greater risk of DHF. The chymase level is a robust prognostic biomarker of severe dengue for adult and pediatric patients.

  • Edge Intelligence in Softwarized 6G : Deep Learning-enabled Network Traffic Predictions

    • The 6G vision is envisaged to enable agile network expansion and rapid deployment of new on-demand microservices (e.g., visibility services for data traffic management, mobile edge computing services) closer to the network's edge IoT devices. However, providing one of the critical features of network visibility services, i.e., data flow prediction in the network, is challenging at the edge devices within a dynamic cloud-native environment as the traffic flow characteristics are random and sporadic. To provide the AI-native services for the 6G vision, we propose a novel edge-native framework to provide an intelligent prognosis technique for data traffic management in this paper. The prognosis model uses long short-term memory (LSTM)-based encoder-decoder deep learning, which we train on real time-series multivariate data records collected from the edge μ-boxes of a selected testbed network. Our result accurately predicts the statistical characteristics of data traffic and verifies the trained model against the ground truth observations. Moreover, we validate our novel framework with two performance metrics for each feature of the multivariate data.

  • Fusion pores with low conductance are cation selective

    • Many neurotransmitters are organic ions that carry a net charge, and their release from secretory vesicles is therefore an electrodiffusion process. The selectivity of early exocytotic fusion pores is investigated by combining electrodiffusion theory, measurements of amperometric foot signals from chromaffin cells with anion substitution, and molecular dynamics simulation. The results reveal that very narrow fusion pores are cation selective, but more dilated fusion pores become anion permeable. The transition occurs around a fusion pore conductance of ∼300 pS. The cation selectivity of a narrow fusion pore accelerates the release of positively charged transmitters such as dopamine, noradrenaline, adrenaline, serotonin, and acetylcholine, while glutamate release may require a more dilated fusion pore.
