    • The Cherenkov Telescope Array (CTA) is a new observatory for very high-energy (VHE) gamma rays. CTA has ambitions science goals, for which it is necessary to achieve full-sky coverage, to improve the sensitivity by about an order of magnitude, to span about four decades of energy, from a few tens of GeV to above 100 TeV with enhanced angular and energy resolutions over existing VHE gamma-ray observatories. An international collaboration has formed with more than 1000 members from 27 countries in Europe, Asia, Africa and North and South America. In 2010 the CTA Consortium completed a Design Study and started a three-year Preparatory Phase which leads to production readiness of CTA in 2014. In this paper we introduce the science goals and the concept of CTA, and provide an overview of the project. (C) 2013 Elsevier B.V. All rights reserved.

    • Attaining a sufficient level of safety from the point of view of stability is typically considered to be a matter of design. However, it is impossible to ensure safety only by design measures, and operational measures can then represent a complementary tool for efficiently and cost-effectively increasing the overall safety of the vessel. Time could therefore be coming for systematically considering operational measures as a recognised and regulated integral part of a holistic approach to ship safety from the point of view of stability. This paper therefore aims at capturing recent trends of research targeting operational safety measures, with specific attention to the intact ship condition. Open challenges and opportunities for research are identified, potential benefits and shortcomings of different options are discussed, and needs and possibilities for further developments in this area are explored. As an overall goal, this paper aims at providing food for thoughts as well as a ground for further proceeding towards the target of implementing a virtuous integrated approach to intact ship safety, from the point of view of stability, which gives due credit to effective and robust operational risk control options.

    • In Naval Architecture, the concept of "stability" has a very wide meaning, embracing ship stability fundamentals with ship dynamics and ultimately ship safety. As such, the subject is of paramount importance for its wide implications on design and operation of ships and floating units. Research in this field has, therefore, an important impact on the fleet safety, and the International Conferences on Stability of Ships and Ocean Vehicles (STAB Conferences) and the International Ship Stability Workshops (ISSW) are certainly the venues where expertise and contemporary developments in this specific field tend to be collected and thoroughly debated. With the aim of analysing the current status and possible future perspectives of research in the field of ship stability, dynamics and safety, this paper provides an extensive review of the findings presented at STAB Conferences and ISSW events in the period 2009-2014. The reviewed material is organised according to a set of identified macro-topics of research. On the basis of the reviewed material, consolidated research topics as well as emerging subjects are laid out, and ideas for possible future research are also brought forward. Discussion is also provided regarding the link between research and educational aspects.

    • Vehicle electrification is a strong trend that introduces new challenges, such as increased input speed of the transmission and increased power density. Alsothe noise emittance of the gearbox is of increasing importance, as the sound of the gearbox is no longer masked by the internal combustion engine. Pressed and sintered powder metallurgical steel could be an interesting alternative towrought steel; the internal porosity has a dampening effect on the noise, and gears can be made in a fast and efficient process. However, current manufacturing of powder metallurgical steel has significant performance limitations. The Nanotechnology Enhanced Sintered Steel Processing project aims to reduce the gap in performance between conventional steel and powder metallurgical steel. One of the potential benefits is that with the inclusion of nano-powder the density can be increased. To validate the new material, its performance needs to be compared to the performance of current generation powder metallurgical materials and also to wrought steel. It is therefor crucial to be able to test and evaluate different materials and gears. This thesis has developed methods for testing, comparing, and evaluating the performance of gears. Powder metallurgical steel has been tested and compared to wrought steel; the efficiency as well as pitting life have been investigated in an FZG test rig. Also the effects of different surface finishing operations have been evaluated. The gear flanks were measured in-situ in the gearbox using a stylus instrument; an optimisation routine was created to fit the measurements to the theoretical involute profile. This enabled an in-depth analysis of surface wear and presented an opportunity to investigate micropitting initiation. It was found that the damage mechanisms of wrought steel and powder metallurgical steel are similar and related to the surface finishing method. However, the powder metallurgical steel was also susceptible to sub-surface cracks. Superfinished gears can be negatively influenced by the lack of tip relief as cracks initiate in the surface layer of the root, rapidly destroying the tooth.

    • A 3D lattice Boltzmann (LB) model with twenty-seven discrete velocities is presented and used for the simulation of three-dimensional porous media flows. Its accuracy in combination with the half-way bounce back boundary condition is assessed. Characteristic properties of the gas diffusion layers that are used in polymer electrolyte fuel cells can be determined with this model. Simulation in samples that have been obtained via X-ray tomographic microscopy, allows to estimate the values of permeability and relative effective diffusivity. Furthermore, the computational LB results are compared with the results of other numerical tools, as well as with experimental values.

    • The development of an improved process for decentralised combined heat and power generation from biomass gasification was the main goal of this project. Based on gasification tests performed in a bench-scale pressurised fluidised gasifier a concept for an allothermal gasification unit was made. Air-blown as well as steam-blown tests were carried out considering three different kinds of biomass (birch, salix, and crushed pelletised straw) against a wide spectrum of operation conditions with different bed-materials (silica sands, magnesite and dolomite). The aim of the gas quality optimisation was the production of a fuel gas (syngas) with a high H2 and low tar content suitable for a gas engine. The tar content was measured with SPA method and a gravimetric method. The tests were performed at the Royal Institute of Technology, Stockholm, in close co-operation with the University of Essen. Jenbacher AG designed a new type of cylinder head for the direct feeding of the hot pressurised syngas into the gas engine in co-operation with the Technical University of Graz. Taking the results obtained into account a concept of decentralised combined heat and power generation (CHP) unit with an electrical power output of 1 MW was set up.

    • Operating Polymer Electrolyte Fuel Cells (PEFC) under high current density conditions, causes significant losses related to liquid water saturation in the gas diffusion layer (GDL). The blockage of pores inside the material has a strong influence on its effective gas transport properties. Here we report on the combination of in-situ X-ray tomographic microscopy (XTM) of PEFC and the numerical determination of gas transport properties using Lattice Boltzmann and finite difference methods. The GDL domains (Toray TGP-H-060) of two identical cells, each with 11 mm(2) active area, were analyzed in sections of about 0.3 to 0.8 mm(2) size. Saturation levels between 0.1 and 0.4 were found, with higher saturation under the ribs. The saturated and the non-saturated states of the GDL samples were compared in order to quantify the dependence of gas phase permeability and effective relative diffusivity on liquid water saturation. Both these relative measures were found to follow power relationships of (1-s)(lambda), where the exponent. was approximately 3 for all cases except for the in-plane diffusivity where it was closer to 2.

  • An integrated cell atlas of the lung in health and disease

    • Single-cell technologies have transformed our understanding of human tissues. Yet, studies typically capture only a limited number of donors and disagree on cell type definitions. Integrating many single-cell datasets can address these limitations of individual studies and capture the variability present in the population. Here we present the integrated Human Lung Cell Atlas (HLCA), combining 49 datasets of the human respiratory system into a single atlas spanning over 2.4 million cells from 486 individuals. The HLCA presents a consensus cell type re-annotation with matching marker genes, including annotations of rare and previously undescribed cell types. Leveraging the number and diversity of individuals in the HLCA, we identify gene modules that are associated with demographic covariates such as age, sex and body mass index, as well as gene modules changing expression along the proximal-to-distal axis of the bronchial tree. Mapping new data to the HLCA enables rapid data annotation and interpretation. Using the HLCA as a reference for the study of disease, we identify shared cell states across multiple lung diseases, including SPP1 + profibrotic monocyte-derived macrophages in COVID-19, pulmonary fibrosis and lung carcinoma. Overall, the HLCA serves as an example for the development and use of large-scale, cross-dataset organ atlases within the Human Cell Atlas.

    • We investigated SARS-CoV-2 potential tropism by surveying expression of viral entry-associated genes in single-cell RNA-sequencing data from multiple tissues from healthy human donors. We co-detected these transcripts in specific respiratory, corneal and intestinal epithelial cells, potentially explaining the high efficiency of SARS-CoV-2 transmission. These genes are co-expressed in nasal epithelial cells with genes involved in innate immunity, highlighting the cells’ potential role in initial viral infection, spread and clearance. The study offers a useful resource for further lines of inquiry with valuable clinical samples from COVID-19 patients and we provide our data in a comprehensive, open and user-friendly fashion at www.covid19cellatlas.org.
