  • Effective Field Theory and Inelastic Dark Matter Results from XENON1T

    • In this work we expand on the XENON1T nuclear recoil searches and study the individual signals of Dark Matter interactions from operators up to dimension-eight in a Chiral Effective Field Theory (ChEFT) as well as a model of inelastic Dark Matter using data from the two science runs of the detector totalling 1 tonne*year exposure. For these analyses we extended the region of interest from [4.9, 40.9]keVnr to [4.9, 54.4]keVnr to enhance our sensitivity for signals that peak at nonzero energies. We show that the data is consistent with a background only hypothesis, with small excesses in the models which peak between 20 and 50keVnr, obtaining a maximum local discovery significance of 1.7 for the VVs ChEFT model for a WIMP mass of 70GeV/c2, and 1.8 for an iDM particle of 50GeV/c2 with a mass splitting of 100keV/c2. For each model we report 90% confidence level upper limits. We also report limits on three benchmark models of WIMP interaction using ChEFT for which we investigate the effect of isospin breaking interactions, reporting up to 6 orders of magnitude weaker limits with respect to the isospin conserving case driven by cancellations in the expected rate.

  • Megahertz-rate Ultrafast X-ray Scattering and Holographic Imaging at the European XFEL

    • The advent of X-ray free-electron lasers (XFELs) has revolutionized fundamental science, from atomic to condensed matter physics, from chemistry to biology, giving researchers access to X-rays with unprecedented brightness, coherence, and pulse duration. All XFEL facilities built until recently provided X-ray pulses at a relatively low repetition rate, with limited data statistics. Here, we present the results from the first megahertz repetition rate X-ray scattering experiments at the Spectroscopy and Coherent Scattering (SCS) instrument of the European XFEL. We illustrate the experimental capabilities that the SCS instrument offers, resulting from the operation at MHz repetition rates and the availability of the novel DSSC 2D imaging detector. Time-resolved magnetic X-ray scattering and holographic imaging experiments in solid state samples were chosen as representative examples, providing an ideal test-bed for operation at megahertz rates. Nevertheless, our results are relevant and applicable to any other non-destructive XFEL experiments in the soft X-ray range.

  • Hh signalling regulates odorant receptor cilia localization in Drosophila

    • Hedgehog (Hh) signaling is a key regulatory pathway during development. Here, we show that in adult OSNs the Hh pathway regulate ?dorant receptor transport to cilia and put forward a novel non-developmental function of the pathway as a neuromodulator. We demonstrate that the level of Hh signal modulate the OSNs response to odors. We show that knock down of Hh and Smoothened (Smo), a transmembrane protein that transduce the signal, are required for receptor transport. We further show that the coreceptor, Orco, has an Hh independent transport path and that knock down of Smo segregate OR and Orco to different vesicular compartments. Last, we show that the odor response to the second receptor type in Drosophila olfaction, the ionotropic receptors (IRs), also require Hh signalling. Thus, Hh signalling is a general regulator of the odorant response that fulfils the criteria of being a potential player in Drosophila odorant adaptation.

    • Only a small fraction of the world’s fungi is described, and the large number of unnamed fungal sequences from environmental DNA serves as a significant complication for taxonomic identification in metabarcoding studies.  There are several deeply diverged fungal lineages—some hypothesized to represent class and order ranks—currently known only by environmental DNA sequences that represent them.  Here, we describe a previously unidentified fungal culture as the first living representative of one of these lineages (previously referred to as ‘clade GS25’, hypothesized as an early diverging lineage sister to the Pucciniomycotina, Basidiomycota) and present its sequenced genome.  We used a phylogenomic approach to test the placement and taxonomic rank of this lineage.  We formally describe the first known species in Eludereomycetes, class nov. (Pucciniomycotina, Basidiomycota): Eluderea minerophilus, gen. et sp. nov.  This species was isolated from Pinus sylvestris roots collected from a Swedish pine forest, where previously published environmental sequencing data from the site indicate that it is almost exclusively found in the deeper mineral soil horizons.  Our analysis shows that the most closely related described species to E. minerophilus likely belongs to the Classiculomycetes, a class within the Pucciniomycotina that is morphologically, molecularly, and ecologically distinct from the Eludereomycetes.  Identification of a living representative for this lineage previously detected in environmental DNA, ‘clade GS25’, and describing it as a new species was a somewhat serendipitous discovery made possible in part due to characteristics of two previously published studies.  We call attention to some of these aspects and propose a set of practices that could be adopted by the research community to help facilitate more connections between living fungus and environmental DNA sequence.  The use of such practices would in turn help to alleviate some of the complications associated with unidentified fungal DNA sequences in reference databases and contribute towards a more complete understanding of fungal diversity.
