  • High degree of consensus amongst an expert panel regarding focal resurfacing of chondral and osteochondral lesions of the femur with mini-implants

    • Introduction: The rationale for the use of mini-implants for partial resurfacing in the treatment of femoral chondral and osteochondral lesions is still under debate. The evidence supporting best practise guidelines is based on studies with low-level evidence. A consensus group of experts was convened to collaboratively advance towards consensus opinions regarding the best available evidence. The purpose of this article is to report the resulting consensus statements. Methods: Twenty-five experts participated in a process based on the Delphi method of achieving consensus. Questions and statements were drafted via an online survey of two rounds, for initial agreement and comments on the proposed statements. An in-person meeting between the panellists was organised during the 2022 ESSKA congress to further discuss and debate each of the statements. A final agreement was made via a final online survey a few days later. The strength of consensus was characterised as: consensus, 51–74% agreement; strong consensus, 75–99% agreement; unanimous, 100% agreement. Results: Statements were developed in the fields of patient assessment and indications, surgical considerations and postoperative care. Between the 25 statements that were discussed by this working group, 18 achieved unanimous, whilst 7 strong consensus. Conclusion: The consensus statements, derived from experts in the field, represent guidelines to assist clinicians in decision-making for the appropriate use of mini-implants for partial resurfacing in the treatment of femoral chondral and osteochondral lesions.   • The search for disks or planetary objects around directly imaged companions : a candidate around DH Tauri B

    • Context. In recent decades, thousands of substellar companions have been discovered with both indirect and direct methods of detection. While the majority of the sample is populated by objects discovered using radial velocity and transit techniques, an increasing number have been directly imaged. These planets and brown dwarfs are extraordinary sources of information that help in rounding out our understanding of planetary systems.Aims. In this paper, we focus our attention on substellar companions detected with the latter technique, with the primary goal of investigating their close surroundings and looking for additional companions and satellites, as well as disks and rings. Any such discovery would shed light on many unresolved questions, particularly with regard to their possible formation mechanisms.Methods. To reveal bound features of directly imaged companions, whether for point-like or extended sources, we need to suppress the contribution from the source itself. Therefore, we developed a method based on the negative fake companion technique that first estimates the position in the field of view (FoV) and the flux of the imaged companion with high precision, then subtracts a rescaled model point spread function (PSF) from the imaged companion, using either an image of the central star or another PSF in the FoV. Next it performs techniques, such as angular differential imaging, to further remove quasi-static patterns of the star (i.e., speckle contaminants) that affect the residuals of close-in companions.Results. After testing our tools on simulated companions and disks and on systems that were chosen ad hoc, we applied the method to the sample of substellar objects observed with SPHERE during the SHINE GTO survey. Among the 27 planets and brown dwarfs we analyzed, most objects did not show remarkable features, which was as expected, with the possible exception of a point source close to DH Tau B. This candidate companion was detected in four different SPHERE observations, with an estimated mass of ~1MJup, and a mass ratio with respect to the brown dwarf of 1∕10. This binary system, if confirmed, would be the first of its kind, opening up interesting questions for the formation mechanism, evolution, and frequency of such pairs. In order to address the latter, the residuals and contrasts reached for 25 companions in the sample of substellar objects observed with SPHERE were derived. If the DH Tau Bb companion is real, the binary fraction obtained is ~7%, which is in good agreement with the results obtained for field brown dwarfs.Conclusions. While there may currently be many limitations affecting the exploration of bound features to directly imaged exoplanets and brown dwarfs, next-generation instruments from the ground and space (i.e., JWST, ELT, and LUVOIR) will be able to image fainter objects and, thus, drive the application of this technique in upcoming searches for exo-moons and circumplanetary disks.

    • DNA methylation is a fundamental epigenetic mark that governs gene expression and chromatin organization, thus providing a window into cellular identity and developmental processes1. Current datasets typically include only a fraction of methylation sites and are often based either on cell lines that underwent massive changes in culture or on tissues containing unspecified mixtures of cells2–5. Here we describe a human methylome atlas, based on deep whole-genome bisulfite sequencing, allowing fragment-level analysis across thousands of unique markers for 39 cell types sorted from 205 healthy tissue samples. Replicates of the same cell type are more than 99.5% identical, demonstrating the robustness of cell identity programmes to environmental perturbation. Unsupervised clustering of the atlas recapitulates key elements of tissue ontogeny and identifies methylation patterns retained since embryonic development. Loci uniquely unmethylated in an individual cell type often reside in transcriptional enhancers and contain DNA binding sites for tissue-specific transcriptional regulators. Uniquely hypermethylated loci are rare and are enriched for CpG islands, Polycomb targets and CTCF binding sites, suggesting a new role in shaping cell-type-specific chromatin looping. The atlas provides an essential resource for study of gene regulation and disease-associated genetic variants, and a wealth of potential tissue-specific biomarkers for use in liquid biopsies.

    • Diverse and inclusive marine research is paramount to addressing ocean sustainability challenges in the 21st century, as envisioned by the UN Decade of Ocean Science for Sustainable Development. Despite increasing efforts to diversify ocean science, women continue to face barriers at various stages of their career, which inhibits their progression to leadership within academic institutions. In this perspective, we draw on the collective experiences of thirty-four global women leaders, bolstered by a narrative review, to identify practical strategies and actions that will help empower early career women researchers to become the leaders of tomorrow. We propose five strategies: (i) create a more inclusive culture, (ii) ensure early and equitable career development opportunities for women ECRs, (iii) ensure equitable access to funding for women ECRs, (iv) offer mentoring opportunities and, (v) create flexible, family-friendly environments. Transformational, meaningful, and lasting change will only be achieved through commitment and collaborative action across various scales and by multiple stakeholders.

  • The synthesis of matrices of embedded semiconducting nanowires

    • In this work we report how single crystal nanowires can be assembled into regular arrays using mesoporous thin films to define the architecture. Mesoporous thin films were prepared by a sol-gel method. These provide films of very regular structure and dimensions. The films produced in this way have almost single crystal like structures and can also exhibit strong epitaxy to the underlying silicon substrate. The films are subjected to a supercritical fluid (SCF) environment in which a precursor is decomposed to yield nanowires of metals, semiconductors or oxides. Using these SCF conditions, pore filling is complete and the products are nanowires which are single crystals and structurally aligned in one direction. The growth mechanism of the nanowires is described and size effects discussed.

    • The worldwide obesity epidemic(1) makes it important to understand how lipid turnover (the capacity to store and remove lipids) regulates adipose tissue mass. Cross-sectional studies have shown that excess body fat is associated with decreased adipose lipid removal rates(2,3). Whether lipid turnover is constant over the life span or changes during long-term weight increase or loss is unknown. We determined the turnover of fat cell lipids in adults followed for up to 16 years, by measuring the incorporation of nuclear bomb test-derived C-14 in adipose tissue triglycerides. Lipid removal rate decreases during aging, with a failure to reciprocally adjust the rate of lipid uptake resulting in weight gain. Substantial weight loss is not driven by changes in lipid removal but by the rate of lipid uptake in adipose tissue. Furthermore, individuals with a low baseline lipid removal rate are more likely to remain weight-stable after weight loss. Therefore, lipid turnover adaptation might be important for maintaining pronounced weight loss. Together these findings identify adipose lipid turnover as an important factor for the long-term development of overweight/obesity and weight loss maintenance in humans.
