    • This is the second out of three white papers from the TERRANOVA project, The European Landscape Learning Initiative an Innovative Training Network consortium of the European Union’s Marie Skłodowska-Curie Actions. Between 2019 and 2023, fifteen Early Stage Researchers (ESRs - ie. PhD students) are trained within this project to envision ‘nature-based' landscape developments for Europe, relying on an integrated and multidisciplinary approach. TERRANOVA seeks to outline the necessity to promote good communication skills in research to transfer efficiently ‘landscape knowledge’ to policymakers and the public. This white paper presents TERRANOVA’s vision on landscape transformation and stakeholders’ engagement in knowledge production. First, we reflect on the relevance of engaging with stakeholders in research. Second, we present the outcomes of a workshop conducted with practitioners in nature conservation and reflect on the challenges they reported to integrate interdisciplinarity in their practice. We conclude that the inclusion of stakeholders and practitioners at every stage is key to ensure that research outcomes have a societal impact. Strengthening this approach will ensure that TERRANOVA ESRs’ research outputs are understandable and useful for land managers and decision-makers.HIGHLIGHTS: RECOMMENDATIONSEstablishing connections between TERRANOVA and stakeholders involved in landscape management is crucial to ensure the relevancy and usefulness of ESRs’ research.TERRANOVA researchers and stakeholders met to discuss how research can be tailored to stakeholders’ needs during a workshop on Friday 26th February 2021.TERRANOVA’s ESRs collected recommendations for their own research practices that reinforced their willingness to engage with stakeholders, connect with practitioners and rely on participatory methods for landscape management interventions.Stakeholders identified the engagement of different local stakeholders, the coexistence of diverging visions about conservation goals, the unexpected outcomes of top-down policy incentives, and the difficulty of setting conservation goals priority as main challenges in conservation practices.

    • Aims. Parameters for 55 nearby metal-poor stars are determined using high-resolution spectroscopy. Together with similar data takenfrom a recent analysis, they are used to show trends of their Galactic evolution with stellar [Fe/H] or [Mg/H] abundances. Theseparation of abundance ratios between disk and halo stars is used as a basic criterion for population membership.Methods. After careful selection of a clean subsample free of suspected or known binaries and peculiar stars, abundances of Mg, Na and Al are based on NLTE kinetic equilibrium calculations applied to spectrum synthesis methods.Results. The relation between [Na/Mg] and [Fe/H] is a continuous enrichment through all three Galactic populations spanning a rangeof values between a metal-poor plateau at [Na/Mg] = −0.7 and solar values. [Al/Mg] displays a step-like difference between starsof the Galactic halo with [Al/Mg] ∼ −0.45 and the two disk populations with [Al/Mg] ∼ +0.10. [Al/Mg] ratios, together with the[Mg/Fe] ratios, asymmetric drift velocities V, and stellar evolutionary ages, make possible the individual discrimination between stars of the thick disk and the halo. At present, this evidence is limited by the small number of stars, and by the theoretical and empirical uncertainties of stellar age determinations, but it achieves a high significance.Conclusions. While the stellar sample is not complete with respect to space volume, the resulting abundances indicate the necessityto revise current models of chemical evolution to allow for an adequate production of Al in early stellar generations.

    • LC/MS is an analytical technique that, due to its high sensitivity, has become increasingly popular for the generation of metabolic signatures in biological samples and for the building of metabolic data bases. However, to be able to create robust and interpretable ( transparent) multivariate models for the comparison of many samples, the data must fulfil certain specific criteria: (i) that each sample is characterized by the same number of variables, (ii) that each of these variables is represented across all observations, and (iii) that a variable in one sample has the same biological meaning or represents the same metabolite in all other samples. In addition, the obtained models must have the ability to make predictions of, e. g. related and independent samples characterized accordingly to the model samples. This method involves the construction of a representative data set, including automatic peak detection, alignment, setting of retention time windows, summing in the chromatographic dimension and data compression by means of alternating regression, where the relevant metabolic variation is retained for further modelling using multivariate analysis. This approach has the advantage of allowing the comparison of large numbers of samples based on their LC/MS metabolic profiles, but also of creating a means for the interpretation of the investigated biological system. This includes finding relevant systematic patterns among samples, identifying influential variables, verifying the findings in the raw data, and finally using the models for predictions. The presented strategy was here applied to a population study using urine samples from two cohorts, Shanxi (People's Republic of China) and Honolulu ( USA). The results showed that the evaluation of the extracted information data using partial least square discriminant analysis (PLS-DA) provided a robust, predictive and transparent model for the metabolic differences between the two populations. The presented findings suggest that this is a general approach for data handling, analysis, and evaluation of large metabolic LC/MS data sets.
