  • Compact X-ray microscope for the water window based on a high brightness laser plasma source

    • We present a laser plasma based x-ray microscope for the water window employing a high-average power laser system for plasma generation. At 90 W laser power a brightness of 7.4 x 10(11) photons/(s x sr x mu m(2)) was measured for the nitrogen Ly alpha line emission at 2.478 nm. Using a multilayer condenser mirror with 0.3 % reflectivity 10(6) photons/(mu m(2) x s) were obtained in the object plane. Microscopy performed at a laser power of 60 W resolves 40 nm lines with an exposure time of 60 s. The exposure time can be further reduced to 20 s by the use of new multilayer condenser optics and operating the laser at its full power of 130 W.

  • Twenty-three unsolved problems in hydrology (UPH) - a community perspective

    • This paper is the outcome of a community initiative to identify major unsolved scientific problems in hydrology motivated by a need for stronger harmonisation of research efforts. The procedure involved a public consultation through online media, followed by two workshops through which a large number of potential science questions were collated, prioritised, and synthesised. In spite of the diversity of the participants (230 scientists in total), the process revealed much about community priorities and the state of our science: a preference for continuity in research questions rather than radical departures or redirections from past and current work. Questions remain focused on the process-based understanding of hydrological variability and causality at all space and time scales. Increased attention to environmental change drives a new emphasis on understanding how change propagates across interfaces within the hydrological system and across disciplinary boundaries. In particular, the expansion of the human footprint raises a new set of questions related to human interactions with nature and water cycle feedbacks in the context of complex water management problems. We hope that this reflection and synthesis of the 23 unsolved problems in hydrology will help guide research efforts for some years to come.

  • An Integrated Understanding of the Rapid Metabolic Benefits of a Carbohydrate-Restricted Diet on Hepatic Steatosis in Humans

    • A carbohydrate-restricted diet is a widely recommended intervention for non-alcoholic fatty liver disease (NAFLD), but a systematic perspective on the multiple benefits of this diet is lacking. Here, we performed a short-term intervention with an isocaloric low-carbohydrate diet with increased protein content in obese subjects with NAFLD and characterized the resulting alterations in metabolism and the gut microbiota using a multi-omics approach. We observed rapid and dramatic reductions of liver fat and other cardiometabolic risk factors paralleled by (1) marked decreases in hepatic de novo lipogenesis; (2) large increases in serum beta-hydroxybutyrate concentrations, reflecting increased mitochondrial beta-oxidation; and (3) rapid increases in folate-producing Streptococcus and serum folate concentrations. Liver transcriptomic analysis on biopsy samples from a second cohort revealed downregulation of the fatty acid synthesis pathway and upregulation of folate-mediated one-carbon metabolism and fatty acid oxidation pathways. Our results highlight the potential of exploring diet-microbiota interactions for treating NAFLD.

  • Personal model-assisted identification of NAD(+) and glutathione metabolism as intervention target in NAFLD

    • To elucidate the molecular mechanisms underlying non-alcoholic fatty liver disease (NAFLD), we recruited 86 subjects with varying degrees of hepatic steatosis (HS). We obtained experimental data on lipoprotein fluxes and used these individual measurements as personalized constraints of a hepatocyte genome-scale metabolic model to investigate metabolic differences in liver, taking into account its interactions with other tissues. Our systems level analysis predicted an altered demand for NAD(+) and glutathione (GSH) in subjects with high HS. Our analysis and metabolomic measurements showed that plasma levels of glycine, serine, and associated metabolites are negatively correlated with HS, suggesting that these GSH metabolism precursors might be limiting. Quantification of the hepatic expression levels of the associated enzymes further pointed to altered de novo GSH synthesis. To assess the effect of GSH and NAD(+) repletion on the development of NAFLD, we added precursors for GSH and NAD(+) biosynthesis to the Western diet and demonstrated that supplementation prevents HS in mice. In a proof-of-concept human study, we found improved liver function and decreased HS after supplementation with serine (a precursor to glycine) and hereby propose a strategy for NAFLD treatment.

  • Ion acceleration with ultrafast lasers

    • Hot-electron confinement can build up fields capable of accelerating ions up to MeV energies when an ultrashort 35-fs laser pulse at similar to2x10(18) W/cm(2) interacts with a small spherical target. Singly charged ions with different masses have similar energies. A simple phenomenological model describes how ultrashort and less-energy-consumptive pulses drive ions to MeV energies. The energetic and spatial-emission characteristics of protons, deuterons and oxygen ions released from water and heavy-water droplets of similar to15 mum in size was determined for this interaction scenario.

  • High average brightness water window source for short-exposure cryomicroscopy

    • Laboratory water window cryomicroscopy has recently demonstrated similar image quality as synchrotron-based microscopy but still with much longer exposure times, prohibiting the spread to a wider scientific community. Here we demonstrate high-resolution laboratory water window imaging of cryofrozen cells with 10 s range exposure times. The major improvement is the operation of a lambda = 2.48 nm, 2 kHz liquid nitrogen jet laser plasma source with high spatial and temporal stability at high average brightness >1.5 x 10(12) ph/(s x sr x mu m(2) x line), i.e., close to that of early synchrotrons. Thus, this source enables not only biological x-ray microscopy in the home laboratory but potentially other applications previously only accessible at synchrotron facilities.

  • Ultrashort 1-kHz laser plasma hard x-ray source

    • We achieved a continuous, stable, ultrashort pulse hard x-ray point source by focusing 1.8-W, 1-kHz, 50-fs laser pulses onto a novel, 30-mum-diameter, high-velocity, liquid-metal gallium jet. This target geometry avoids most of the debris problems of solid targets and provides nearly 4pi illumination. Photon fluxes of 5 X 10(8) photons/s are generated in a two-component spectrum consisting of a broad continuum from 4 to 14 keV and strong K-alpha and K-beta emission lines at 9.25 and 10.26 keV. This source will find wide use in time-resolved x-ray diffraction studies and other applications.

  • Influence of laser pulse width on absolute EUV-yield from Xe-clusters

    • Using 50 fs (similar to 2 x 10(18) W/cm(2)) and 2 ps (similar to 5 x 10(16) W/cm(2)) pulses from a Ti:Sa multi-TW laser at 800 nm wavelength large Xe-clusters (10(5)...10(6) atoms per cluster) have been excited. Absolute yield measurements of EUV-emission in a wavelength range between 10 nm and 15 nm in combination with cluster target variation were carried out. The ps-laser pulse has resulted in about 30% enhanced and spatially more uniform EUV-emission compared to fs-laser excitation. Circularly polarized laser light instead of linear polarization results in enhanced emission which is probably caused by electrons gaining higher energies by the polarization dependent optical field ionization process. An absolute emission efficiency at 13.4 nm of up to 0.8% in 2 pi sr and 2.2% bandwidth has been obtained.

  • Kilohertz sources of hard x rays and fast ions with femtosecond laser plasmas

    • We demonstrate a new, stable, kilohertz femtosecond laser plasma source of hard-x-ray continuum and K-alpha emission that uses a microscopic liquid jet target that is continuous and debris free. Plasmas produced by ultrashort (50-fs) intense laser pulses from a fine (10-30-mum diameter) liquid Ga jet emit bright 9.3- and 10.3-keV K-alpha and K-beta lines superimposed on a multikilovolt bremmstrahlung continuum. Kilohertz femtosecond x-ray sources will find many applications in time-resolved x-ray diffraction and microscopy studies. As high-intensity lasers become more compact and operate at increasingly high repetition-rates, they require a target configuration that is both repeatable from shot to shot and debris free. Our target provides a pristine, unperturbed filament surface at rates >100 kHz. A number of liquid metal targets are considered. We show the hard-x-ray spectrum described above. The source was generated by a 50-fs-duration, 1-kHz, 2-W, high-intensity Ti:sapphire laser. Using the same technology, we also generate forward-going sub-mega-electron-volt (sub-MeV) protons from a 10-mum liquid water target at 1-kHz repetition rates. Kilohertz sources of high-energy ions will find many applications in time-resolved particle interaction studies and will lead to efficient generation of short-lived isotopes for use in nuclear medicine and other applications. The protons were detected with CR-39 track detectors in both the forward and the backward directions up to energies of similar to500 keV. As the intensity of compact high-repetition-rate lasers sources increases, we can expect improvements in the energy, conversion efficiency, and directionality to occur. The effect of these developments is discussed. As compact, high-repetition-rate femtosecond laser technology reaches focused intensities of similar to10(19) W/cm(2), many new applications of high-repetition-rate hard-x-ray and MeV ion sources will become practical.

  • Damage investigation on tungsten and diamond diffractive optics at a hard x-ray free-electron laser

    • Focusing hard x-ray free-electron laser radiation with extremely high fluence sets stringent demands on the x-ray optics. Any material placed in an intense x-ray beam is at risk of being damaged. Therefore, it is crucial to find the damage thresholds for focusing optics. In this paper we report experimental results of exposing tungsten and diamond diffractive optics to a prefocused 8.2 keV free-electron laser beam in order to find damage threshold fluence levels. Tungsten nanostructures were damaged at fluence levels above 500 mJ/cm(2). The damage was of mechanical character, caused by thermal stress variations. Diamond nanostructures were affected at a fluence of 59 000 mJ/cm(2). For fluence levels above this, a significant graphitization process was initiated. Scanning Electron Microscopy (SEM) and mu-Raman analysis were used to analyze exposed nanostructures.
