  • Final report of the CCQM-K145 : Toxic and essential elements in bovine liver

    • Liver plays a major role in metabolism and acts as a source of energy for the body by storing glycogen. With the growing interest and investigation in the biological effects in recent years, it is important and necessary to develop accurate and comparable analytical methods for elements in bio-samples. It has, however, been 10 years since the tissue sample (bovine liver) of CCQM-K49 key comparison. The purpose of CCQM-K145 is to ensure the comparable and traceable measurement results for essential and toxic elements such as P, S, Zn, Mn, Ni, Mo, Sr, Cr, Co, Pb, As and Hg in bovine liver among NMIs and other designated measurement bodies worldwide. The comparison was agreed by IAWG as 6th IAWG Benchmarking Exercise with Zn and Ni as exemplary elements at the meeting in Korea in the early October 2016. The results of CCQM-K145 are expected to cover the measurement capability and support CMCs claiming for inorganic elements in the similar biological tissue materials and food samples. 30 NMIs and DIs registered in CCQM-K145. With respect to the methodology, a variety of techniques such as IDMS, ICP-OES, ICP-MS(non-ID), AAS and NAA were adopted by the participants. For Zn, Ni, Sr, Pb and Hg measurements, most participants chose ID-ICP-MS method, which showed the better performance in terms of consistency and reliability of the measurement results. In aspect of the traceability for the measurement results in CCQM-K145, most participants used their own (in house) CRMs or other NMI's CRMs to guarantee trace to SI unit. Most participants used similar matrix CRMs for quality control or method validation. Base on different statistic way to calculate the reference mass fraction values and associated uncertainties for each measurand, removal of the suspected extreme values, and discussion at the IAWG meetings, the median values are proposed as the KCRV for Zn, Ni, Mn, Mo, Cr, Pb and Hg; the arithmetic mean values are proposed as the KCRV for P, S, Sr, Co and As. In general, the performances of the majority of CCQM-K145 participants are very good, illustrating their measurement capabilities for Zn, Ni, P, S, Mn, Mo, Sr, Cr, As, Co, Pb and Hg in a complex biological tissue matrix. Bovine liver contains many kinds of nutrients and microelements, it can be regarded as a typical representative material of biological tissue and food. In CCQM-K145, the analytes involved alkali metals and transition elements, metalloids/semi-metals and non metals with a range of mass fraction from mg/g to μg/kg. CCQM-K145 also tested the ability of NMIs/DIs to determine elements that were easy to be lost and polluted, and interfered significantly. The chemical pretreatment methods of samples used in the comparison is suitable for general food and biological matrix samples. A variety of measurement methods used in the comparison represent the main instrumental technology for elemental analysis. Therefore, for supporting CMC claim, CCQM-K145 is readily applicable to measurement of more elements in a wide range of biological materials (including liquids and solids) and meat products.   • Alkaline pH Is a Signal for Optimal Production and Secretion of the Heat Labile Toxin, LT in Enterotoxigenic Escherichia Coli (ETEC)

    • Enterotoxigenic Escherichia coli (ETEC) cause secretory diarrhea in children and travelers to endemic areas. ETEC spreads through the fecal-oral route. After ingestion, ETEC passes through the stomach and duodenum before it colonizes the lower part of the small intestine, exposing bacteria to a wide range of pH and environmental conditions. This study aimed to determine the impact of external pH and activity of the Cyclic AMP receptor protein (CRP) on the regulation of production and secretion of heat labile (LT) enterotoxin. ETEC strain E2863wt and its isogenic mutant E2863ΔCRP were grown in LBK media buffered to pH 5, 7 and 9. GM1 ELISA, cDNA and cAMP analyses were carried out on bacterial pellet and supernatant samples derived from 3 and 5 hours growth and from overnight cultures. We confirm that CRP is a repressor of LT transcription and production as has been shown before but we show for the first time that CRP is a positive regulator of LT secretion both in vitro and in vivo. LT secretion increased at neutral to alkaline pH compared to acidic pH 5 where secretion was completely inhibited. At pH 9 secretion of LT was optimal resulting in 600 percent increase of secreted LT compared to unbuffered LBK media. This effect was not due to membrane leakage since the bacteria were viable at pH 9. The results indicate that the transition to the alkaline duodenum and/or exposure to high pH close to the epithelium as well as activation of the global transcription factor CRP are signals that induce secretion of the LT toxin in ETEC.

  • Tungsten based divertor development for Wendelstein 7-X

    • Wendelstein 7-X, the world’s largest superconducting stellarator in Greifswald (Germany), started plasma experiments with a water-cooled plasma-facing wall in 2022, allowing for long pulse operation. In parallel, a project was launched in 2021 to develop a W based divertor, replacing the current CFC divertor, to demonstrate plasma performance of a stellarator with a reactor relevant plasma facing materials with low tritium retention. The project consists of two tasks: Based on experience from the previous experimental campaigns and improved physics modelling, the geometry of the plasma-facing surface of the divertor and baffles is optimized to prevent overloads and to improve exhaust. In parallel, the manufacturing technology for a W based target module is qualified. This paper gives a status update of project. It focusses on the conceptual design of a W based target module, the manufacturing technology and its qualification, which is conducted in the framework of the EUROfusion funded WPDIV program. A flat tile design in which a target module is made of a single target element is pursued. The technology must allow for moderate curvatures of the plasma-facing surface to follow the magnetic field lines. The target element is designed for steady state heat loads of 10 MW/m2 (as for the CFC divertor). Target modules of a similar size and weight as for the CFC divertor are assumed (approx. < 0.25 m2 and < 60 kg) using the existing water cooling infrastructure providing 5 l/s and roughly maximum 15 bar pressure drop per module. The main technology under qualification is based on a CuCrZr heat sink made either by additive manufacturing using laser powder bed fusion (LPBF) or by uniaxial diffusion welding of pre-machined forged CuCrZr plates. After heat treatment, the plasma-facing side of the heat sink is covered by W or if feasible by the more ductile WNiFe, preferably by coating or alternatively by hot isostatic pressing W based tiles with a soft OFE-Cu interlayer. Last step is a final machining of the plasma-exposed surface and the interfaces to the water supply lines and supports to correct manufacturing deformations.

  • Hydropower Production Benefits More From 1.5 °C than 2 °C Climate Scenario

    • Hydropower plays an important role as renewable and clean energy in the world's overall energy supply. Electricity generation from hydropower represented approximately 16.6% of the world's total electricity and 70% of all renewable electricity in 2015. Determining the different effects of 1.5 and 2 °C of global warming has become a hot spot in water resources research. However, there are still few studies on the impacts of different global warming levels on gross hydropower potential. This study used a coupled hydrological and techno-economic model framework to assess hydropower production under global warming levels of 1.5 and 2 °C, while also considering gross hydropower potential, power consumption, and economic factors. The results show that both global warming levels will have a positive impact on the hydropower production of a tropical island (Sumatra) relative to the historical period; however, the ratio of hydropower production versus power demand provided by 1.5 °C of global warming is 40% higher than that provided by 2 °C of global warming under RCP6.0. The power generation by hydropower plants shows incongruous changing trends with hydropower potential under the same global warming levels. This inconformity occurs because the optimal sites for hydropower plants were chosen by considering not only hydropower potential but also economic factors. In addition, the reduction in CO2 emissions under global warming of 1.5 °C (39.06 × 106 t) is greater than that under global warming of 2 °C (10.20 × 106 t), which reveals that global warming decreases the benefits necessary to relieve global warming levels. However, the hydropower generation and the reduction in CO2 emissions will be far less than the energy demand when protected areas are excluded as potential sites for hydropower plants, with a sharp decrease of 40–80%. Thus, government policy-makers should consider the trade-off between hydropower generation and forest coverage area in nationally determined contributions. © 2020 The Authors.

    • In situ surface plasmon resonance (SPR) and quartz crystal microbalance (QCM) measurements have been employed to measure the adsorption kinetics and absolute adsorbed amount of the poly(N-isopropyl acrylamide) (pNIPAM) from bulk aqueous solution onto a hydrophobized gold substrate. The adsorption was carried out at 31 degreesC, which is just below the lower critical solution temperature of pNIPAM in water. We find that the shift in the coupling angle of the surface plasmon (proportional to the optical thickness) and the shift in the resonance frequency of the quartz crystal (proportional to the acoustic thickness) increase in parallel for most of the adsorption. Also, the change of dissipation is proportional to the change in frequency. These observations suggest that the buildup of the polymer layer proceeds via growth in thickness rather than by densification of a layer with constant thickness. We interpret this finding in the sense that the dense high-temperature phase wets the hydrophobic gold surface. The wetting layer has a fixed density and grows in thickness. In addition, the QCM has been used to study the temperature-induced conformational change for pNIPAM around the critical temperature. It was found that the technique was able to monitor additional adsorption that occurs when crossing the critical point, which was due to bulk phase separation. Desorption was also noted when crossing the critical point from the opposite direction, and for the given system the process was entirely reversible.

  • Why Do Bamboo Parenchyma Cells Show Higher Nanofibrillation Efficiency than Fibers : An Investigation on Their Hierarchical Cell Wall Structure

    • The cell walls of parenchyma cells and fibers in bamboo are both highly lignified with secondary thickening. However, the former were found to have much higher nanofibrillation efficiency than fibers via both protocols of ultrasonication and high pressure homogenization. To elucidate the inherent mechanism, detailed comparisons of chemical composition, cell morphology, cell wall density, pore structures, and structural organization of cell wall polymers were performed on native and pretreated cell walls of both parenchyma cells and fibers. Chemical compositional analysis showed that fibers have much higher cellulose (49.8% to 35.5%) but lower xylan content (21.1% to 36.2%) than parenchyma, while their lignin contents were similar (24.9% vs 22.9%). Polarized FTIR further revealed clear differences in the structural organization of polymers between the two types of cells, with all the polymers of fibers being more orderly assembled than those of parenchyma cells. The compact arrangement of polymers in the fibers was also supported by the much higher cell wall density (1.52 vs 1.28 g/cm3) and lower porosity (0.007 vs 0.013 cc/g after chemical pretreatments), as compared to the parenchyma cells. The study provides evidence that the anatomical characteristics of huge cavity-wall ratio, higher cell wall porosity, and less ordered arrangement of cell wall matrix polymers (mainly lignin) in parenchyma cells contribute to their higher nanofibrillation efficiency compared to fibers.

This paper outlines the main challenges, which the 5G transport networks are facing and discusses in more detail data plane, control architectures, and the tradeoff between different network abstraction models.
