  • High expression of cyclin D1 is associated to high proliferation rate and increased risk of mortality in women with ER-positive but not in ER-negative breast cancers
  • 2017
  • Ingår i: Breast Cancer Research and Treatment. - : SPRINGER. - 0167-6806 .- 1573-7217. ; 164:3, s. 667-678
  • Tidskriftsartikel (refereegranskat)abstract
    • Cyclin D1 has a central role in cell cycle control and is an important component of estrogen regulation of cell cycle progression. We have previously shown that high cyclin D expression is related to aggressive features of ER-positive but not ER-negative breast cancer. The aims of the present study were to validate this differential ER-related effect and furthermore explore the relationship between cyclin D overexpression and CCND1 gene amplification status in a node-negative breast cancer case-control study. Immunohistochemical nuclear expression of cyclin D1 (n = 364) and amplification of the gene CCND1 by fluorescent in situ hybridization (n = 255) was performed on tissue microarray sections from patients with T1-2N0M0 breast cancer. Patients given adjuvant chemotherapy were excluded. The primary event was defined as breast cancer death. Breast cancer-specific survival was analyzed in univariate and multivariable models using conditional logistic regression. Expression of cyclin D1 above the median (61.7%) in ER breast cancer was associated with an increased risk for breast cancer death (OR 3.2 95% CI 1.5-6.8) also when adjusted for tumor size and grade (OR 3.1). No significant prognostic impact of cyclin D1 expression was found among ER-negative cases. Cyclin D1 overexpression was significantly associated to high expression of the proliferation markers cyclins A (rho 0.19, p = 0.006) and B (rho 0.18, p = 0.003) in ER-positive tumors, but not in ER-negative cases. There was a significant association between CCND1 amplification and cyclin D1 expression (p = 0.003), but CCND1 amplification was not statistically significantly prognostic (HR 1.4, 95% CI 0.4-4.4). We confirmed our previous observation that high cyclin D1 expression is associated to high proliferation and a threefold higher risk of death from breast cancer in ER-positive breast cancer.

  • Using Fourier transform infrared spectroscopy to determine toxic gases in fires with lithium-ion batteries
  • 2016
  • Ingår i: Fire and Materials. - : Wiley. - 0308-0501 .- 1099-1018. ; 40:8, s. 999-1015
  • Tidskriftsartikel (refereegranskat)abstract
    • Batteries, in particular lithium-ion (Li-ion) batteries, are seen as an alternative to fossil fuels in the automotive sector. Li-ion batteries, however, have some safety issues including possible emissions of toxic fluorine-containing compounds during fire and other abuse situations. This paper demonstrates the possibilities to use the Fourier transform infrared technique to assess some of the most important compounds, including hydrogen fluoride and the far less often measured POF3 and PF5. The study is conducted in the cone calorimeter with different solvents used in Li-ion batteries. The measurements show that, in addition to hydrogen fluoride, with a known high toxicity, POF3 is emitted and can be quantified using Fourier transform infrared.

  • Flerspråkighet eller språkförbistring? : Finska segment i svenska medeltidsbrev 1350–1526
  • 2017
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • This thesis examines fragmentary Finnish in Late Old Swedish charters (c. 1350–1526) issued in the Finnish part of the Swedish realm, the diocese of Åbo. Consisting mostly of proper names, albeit occasionally displaying Finnish inflectional and derivational morphology, these fragments have previously not generally been regarded as representing actual written Finnish, but rather as onomastic loans or transcriptions of oral language by more or less monolingual Swedish scribes. This thesis attempts a description and analysis of the Swedish–Finnish language mixture, to see to what extent the embedding of Finnish segments in these Swedish-language charters can be said to reflect scribal proficiency in Finnish or a lack thereof. The thesis relies on theoretical and empirical findings in the fields of code-switching and historical sociolinguistics. To provide a socio-historical context for the linguistic analysis, sociolinguistic conditions in medieval Finland and the textual genre of medieval charters are outlined. The bilingual segments in the data are then described and compared with models of code-switching from modern studies, to see whether their form corresponds to patterns that could be expected of more or less balanced bilinguals. The choice between Swedish and Finnish linguistic variants is also considered in the light of textual and sociolinguistic factors, and a study is made of Finnish grammatical transfer in the scribal Swedish of medieval Finland.Although the scarcity of the medieval data does not allow definite conclusions, the tentative results reveal a language mixture that is mainly well formed, though limited in scope and with some instances of scribal errors that could be due to a lack of proficiency in Finnish. On the other hand, the insertion of Finnish segments shows a stylistic patterning that suggests a linguistic awareness on the part of the scribes, and the choice of Swedish prepositions in certain constructions differs quantitatively from the norm in non-Finnish parts of medieval Sweden, in a way that can partly be attributed to the influence of Finnish locative case semantics. While it is apparent that proficiency levels in Finnish must have varied somewhat among medieval scribes in Finland, the results point to a more or less bilingual proficiency, or at least extensive passive knowledge of Finnish.

    • In this paper I will examine the mixing of Swedish and Finnishin medieval charters from Finland from the 14th to the 16th century. In thesecharters, where Swedish from the mid-14th century onwards functions asthe de jure framing language, one can find embedded Finnish-languagelinguistic segments, mainly in complex personal and topographic designations.Apart from Finnish proper nouns, flectional and derivational endingsabound, and there are even sporadic occurrences of common nouns. With evidence from research in other mixed-language texts from medievalEurope, I present the following four main conditioning factors formixed language text formation as identified by previous scholars: 1) scriballinguistic deficiency, 2) traces of oral language, 3) pragmatic benefits of theuse of vernacular for achieving intelligibility, and 4) scribal multilingualism. In earlier research, the aforementioned language-mixing has hardlybeen commented on, and if it has been it has mostly been considered to betranscribed oral language rendered in a more or less obscured manner bynon-Finnish-speaking Swedish scribes. I argue that all of the four abovementionedconditioning factors for written language-mixing must be consideredin studying fragmentary vernacular language in Finnish charters. What might seem to be linguistic confusion at first glance can instead beinterpreted as multilingual proficiency and an intentional use of multilingualresources for communicative ends.

  • The prognostic role of HER2 expression in ductal breast carcinoma in situ (DCIS); a population-based cohort study
  • 2015
  • Ingår i: BMC Cancer. - : BioMed Central (BMC). - 1471-2407 .- 1471-2407. ; 15
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: HER2 is a well-established prognostic and predictive factor in invasive breast cancer. The role of HER2 in ductal breast carcinoma in situ (DCIS) is debated and recent data have suggested that HER2 is mainly related to in situ recurrences. Our aim was to study HER2 as a prognostic factor in a large population based cohort of DCIS with long-term follow-up. Methods: All 458 patients diagnosed with a primary DCIS 1986-2004 in two Swedish counties were included. Silver-enhanced in situ hybridisation (SISH) was used for detection of HER2 gene amplification and protein expression was assessed by immunohistochemistry (IHC) in tissue microarrays. HER2 positivity was defined as amplified HER2 gene and/or HER2 3+ by IHC. HER2 status in relation to new ipsilateral events (IBE) and Invasive Breast Cancer Recurrences, local or distant (IBCR) was assessed by Kaplan-Meier survival analyses and Cox proportional hazards regression models. Results: Primary DCIS was screening-detected in 75.5 % of cases. Breast conserving surgery (BCS) was performed in 78.6 % of whom 44.0 % received postoperative radiotherapy. No patients received adjuvant endocrine-or chemotherapy. The majority of DCIS could be HER2 classified (N = 420 (91.7 %)); 132 HER2 positive (31 %) and 288 HER2 negative (69 %)). HER2 positivity was related to large tumor size (P = 0.002), high grade (P < 0.001) and ER-and PR negativity (P < 0.001 for both). During follow-up (mean 184 months), 106 IBCRs and 105 IBEs were identified among all 458 cases corresponding to 54 in situ and 51 invasive recurrences. Eighteen women died from breast cancer and another 114 had died from other causes. The risk of IBCR was statistically significantly lower subsequent to a HER2 positive DCIS compared to a HER2 negative DCIS, (Log-Rank P = 0.03, (HR) 0.60 (95 % CI 0.38-0.94)). Remarkably, the curves did not separate until after 10 years. In ER-stratified analyses, HER2 positive DCIS was associated with lower risk of IBCR among women with ER negative DCIS (Log-Rank P = 0.003), but not for women with ER positive DCIS. Conclusions: Improved prognostic tools for DCIS patients are warranted to tailor adjuvant therapy. Here, we demonstrate that HER2 positive disease in the primary DCIS is associated with lower risk of recurrent invasive breast cancer.
