    • Here we report the generation of a multimodal cell census and atlas of the mammalian primary motor cortex as the initial product of the BRAIN Initiative Cell Census Network (BICCN). This was achieved by coordinated large-scale analyses of single-cell transcriptomes, chromatin accessibility, DNA methylomes, spatially resolved single-cell transcriptomes, morphological and electrophysiological properties and cellular resolution input–output mapping, integrated through cross-modal computational analysis. Our results advance the collective knowledge and understanding of brain cell-type organization1–5. First, our study reveals a unified molecular genetic landscape of cortical cell types that integrates their transcriptome, open chromatin and DNA methylation maps. Second, cross-species analysis achieves a consensus taxonomy of transcriptomic types and their hierarchical organization that is conserved from mouse to marmoset and human. Third, in situ single-cell transcriptomics provides a spatially resolved cell-type atlas of the motor cortex. Fourth, cross-modal analysis provides compelling evidence for the transcriptomic, epigenomic and gene regulatory basis of neuronal phenotypes such as their physiological and anatomical properties, demonstrating the biological validity and genomic underpinning of neuron types. We further present an extensive genetic toolset for targeting glutamatergic neuron types towards linking their molecular and developmental identity to their circuit function. Together, our results establish a unifying and mechanistic framework of neuronal cell-type organization that integrates multi-layered molecular genetic and spatial information with multi-faceted phenotypic properties.

    • Polymer crosslinking is crucial for the preparation and consolidation of hierarchical nano- and micro-structures, hybrid interfaces, and collective assemblies. Here, for the first time, we showed that a cation-methylene-nitrile (CMN) functionality sequence encoded within repeating units of poly(ionic liquid)s (PILs) allowed for mild cyclizations of nitriles, processes otherwise requiring high temperatures and harsh catalysts. These new reactions facilitated by the CMN sequence were readily translated into freestanding nanomembranes (similar to 19 nm in thickness) and nanocomposite membranes by treating the PILs with mild ammonia vapor (0.2 bar, 20 degrees C). These materials were observed to be stable in various solvents, at different pH levels, and even in boiling water, exhibiting exceptional mechanical strength and solar-thermal desalination performance. The sequence was easy to synthesize, transferable in copolymers, and applicable to various cations, such as imidazolium, pyridinium, and triazolium. We expect it to provide a molecular code promoting programmable polymer crosslinking and the formation of hybrid structures for sustainable energy and water applications.

  • Iron-chelated thermoresponsive polymer brushes on bismuth titanate nanosheets for metal affinity separation of phosphoproteins

    • Separation of phosphoproteins plays an important role for identification of biomarkers in life science. In this work, bismuth titanate supported, iron-chelated thermoresponsive polymer brushes were prepared for selective separation of phosphoproteins. The iron-chelated thermoresponsive polymer brushes were synthesized by surface-initiated atom transfer radical polymerization of N-isopropylacrylamide and glycidyl methacrylate, followed by a ring opening reaction of epoxy group, and chelation of the obtained cis-diols with Fe3+ ions. The composite material was characterized to determine the size and thickness, the content of the organic polymer and the metal loading. The bismuth titanate supported, iron-chelated thermoresponsive polymer brushes showed selective binding for phosphoproteins in the presence of abundant interfering proteins, and a high binding capacity for phosphoproteins by virtue of the metal affinity between the metal ions on the polymer brushes and the phosphate groups in the phosphoproteins (664 mg β-Casein per g sorbent). The thermoresponsive property of the polymer brushes made it possible to adjust phosphoprotein binding by changing temperature. Finally, separation of phosphoproteins from a complex biological sample (i.e. milk) was demonstrated using the nanosheet-supported thermoresponsive polymer brushes.

  • Identifying a Safe and Just Corridor for People and the Planet

    • Keeping the Earth system in a stable and resilient state, to safeguard Earth's life support systems while ensuring that Earth's benefits, risks, and related responsibilities are equitably shared, constitutes the grand challenge for human development in the Anthropocene. Here, we describe a framework that the recently formed Earth Commission will use to define and quantify target ranges for a safe and just corridor that meets these goals. Although safe and just Earth system targets are interrelated, we see safe as primarily referring to a stable Earth system and just targets as being associated with meeting human needs and reducing exposure to risks. To align safe and just dimensions, we propose to address the equity dimensions of each safe target for Earth system regulating systems and processes. The more stringent of the safe or just target ranges then defines the corridor. Identifying levers of social transformation aimed at meeting the safe and just targets and challenges associated with translating the corridor to actors at multiple scales present scope for future work.

  • Magnetic topological insulator MnBi6 Te10 with a zero-field ferromagnetic state and gapped Dirac surface states

    • Magnetic topological insulators (TIs) with nontrivial topological electronic structure and broken time-reversal symmetry exhibit various exotic topological quantum phenomena. The realization of such exotic phenomena at high temperature is one of the central topics in this area. We reveal that MnBi6Te10 is a magnetic TI with an antiferromagnetic ground state below 10.8 K whose nontrivial topology is manifested by Dirac-like surface states. The ferromagnetic axion insulator state with Z4=2 emerges once spins are polarized at a field as low as 0.1 T, accompanied with saturated anomalous Hall resistivity up to 10 K. Such a ferromagnetic state is preserved even with an external field down to zero at 2 K. Theoretical calculations indicate that the few-layer ferromagnetic MnBi6Te10 is also topologically nontrivial with a nonzero Chern number. Angle-resolved photoemission spectroscopy experiments further reveal three types of Dirac surface states arising from different terminations on the cleavage surfaces, one of which has insulating behavior with an energy gap of ∼28 meV at the Dirac point. These outstanding features suggest that MnBi6Te10 is a promising system to realize various topological quantum effects at zero field and high temperature.

    • The Lancet Countdown is an international collaboration established to provide an independent, global monitoring system dedicated to tracking the emerging health profile of the changing climate.The 2020 report presents 43 indicators across five sections: climate change impacts, exposures, and vulnerabilities; adaptation, planning, and resilience for health; mitigation actions and health co-benefits; economics and finance; and public and political engagement. This report represents the findings and consensus of the 35 leading academic institutions and UN agencies that make up The Lancet Countdown, and draws on the expertise of climate scientists, geographers, engineers, experts in energy, food, and transport, economists, social, and political scientists, data scientists, public health professionals, and doctors.

    • In 2018, a total of US$166 billion global economic losses and a new high of 55.3 Gt of CO2 equivalent emission were generated by 831 climate-related extreme events. As the world's largest CO2 emitter, we reported China's recent progresses and pitfalls in climate actions to achieve climate mitigation targets (i.e., limit warming to 1.52 degrees C above the pre-industrial level). We first summarized China's integrated actions (2015 onwards) that benefit both climate change mitigation and Sustainable Development Goals (SDGs). These projects include re-structuring organizations, establishing working goals and actions, amending laws and regulations at national level, as well as increasing social awareness at community level. We then pointed out the shortcomings in different regions and sectors. Based on these analyses, we proposed five recommendations to help China improving its climate policy strategies, which include: 1) restructuring the economy to balance short-term and long-term conflicts; 2) developing circular economy with recycling mechanism and infrastructure; 3) building up unified national standards and more accurate indicators; 4) completing market mechanism for green economy and encouraging green consumption; and 5) enhancing technology innovations and local incentives via bottom-up actions.
