    • This paper addresses issues in closed loop performance monitoring. Particular attention is paid to detecting whether an observed deviation from nominal performance is due to a disturbance or due to a system change. This is achieved by introducing a novel performance measure that allows feasible application of a standard CUSUM change detector. The paper includes explicit results on the probability of mistaking a disturbance for a system change and demonstrates the algorithm with a simulation study.

  • Nuclear Magnetic Resonance Spectroscopy and Spin Relaxation in Amphiphilic Liquid Crystals

    • Spin relaxation theory and nuclear magnetic resonance spectroscopy in amphiphilic liquid crystals are presented. Group theory is used to determine the information content and the orientation dependence in spin relaxation experiments, and to investigate the combined effect of crystal and local symmetries and the effect of symmetry on multiple timescales. From lineshapes of a deuterated decanol and SdS molecules the distribution within and aspect ratio of rodlike aggregates in the rectangular phase of the system SdS/decanol/water are determined. The results reveal the importance of electrostatic interactions in stabilisation of the rectangular phase. A geometry independent theory is developed for spin relaxation by diffusion on a biaxial rod. The most striking result of this theory is the strong geometry dependence of the spinrelaxation functions. A theoretical investigation of spinrelaxation in coupled membrane system are presented. The coupled membrane theory reveals a strong dependence of spectral density functions on membrane coupling. The second and fourth order spectral density functions are calculated. The effective diffusion coefficient for a particle diffusing on a random surface is related to the degree of roughness of the surface. An exact result for the diffusion coefficient is obtained in the limit of fast surface fluctuations, for a static surface upper and lower bounds and an effective medium approximation are derived. From diffusion measurements the surface configuration can then be determined. For particle diffusion in one dimensional random geometry an exact results for the diffusion coefficient is obtained for rapidly fluctuating geometry, a static geometry and in the general case.

  • Antenna protein composition of PS I and PS II in thylakoid sub-domains

    • Spinach thylakoids were separated into grana core, grana margin, and two different stromal lamella fractions in the absence of detergents. The levels of all light-harvesting chlorophyll a/b-binding (LHC) proteins were determined in all fractions, and were normalised to the amount of Photosystem I (PS I) and Photosystem II (PS II) centres. PS Iβ in the stroma lamellae was found to have a full complement of Lhca polypeptides and, probably, one attached LHC II trimer. PS Iα binds additional LHC II trimers, but PS I centres located in the inner parts of the grana stack lack Lhca1 and are depleted in Lhca4. PS IIβ, found in grana margins and stroma lamellae, seems to associate one monomer each of Lhcb4, Lhcb5 and Lhcb6 (CP29, CP26 and CP24, respectively) and one LHC II trimer consisting of two Lhcb1 and one Lhcb3 subunit. PS IIα has additional LHC II trimers (consisting of Lhcb1 and Lhcb2) attached. We also find evidence for the existence of both PS I and PS II centres in the extreme stroma (probably centres being synthesised or repaired), that lack all LHC proteins.

    • Monospecific polyclonal antibodies have been raised against synthetic peptides derived from the primary sequences from different plant light-harvesting Chl a/b-binding (LHC) proteins. Together with other monospecific antibodies, these were used to quantify the levels of the 10 different LHC proteins in wild-type and chlorina 12 barley (Hordeum vulgare L.), grown under normal and intermittent light (ImL). Chlorina 12, grown under normal light, lacked Lhcb1 (type I LHC II) and Lhcb6 (CP24) and had reduced amounts of Lhcb2, Lhcb3 (types II and III LHC II), and Lhcb4 (CP 29). Chlorina f2 grown under ImL lacked all LHC proteins, whereas wild-type ImL plants contained Lhcb5 (CP 26) and a small amount of Lhcb2. The chlorina f2 ImL thylakoids were organized in large parallel arrays, but wild-type ImL thylakoids had appressed regions, indicating a possible role for Lhcb5 in grana stacking. Chlorina f2 grown under ImL contained considerable amounts of violaxanthin (2-3/reaction center), representing a pool of phototransformable xanthophyll cycle pigments not associated with LHC proteins. Chlorina f2 and the plants grown under ImL also contained early light-induced proteins (ELIPs) as monitored by western blotting. The levels of both ELIPs and xanthophyll cycle pigments increased during a 1 h of high light treatment, without accumulation of LHC proteins. These data are consistent with the hypothesis that ELIPs are pigment-binding proteins, and we suggest that ELIPs bind photoconvertible xanthophylls and replace ''normal'' LHC proteins under conditions of light stress.

    • The accumulation of compatible solutes, such as glycerol, in the yeast Saccharomyces cerevisiae, is a ubiquitous mechanism in cellular osmoregulation. Here, we demonstrate that yeast cells control glycerol accumulation in part via a regulated, Fps1p-mediated export of glycerol. Fps1p is a member of the MIP family of channel proteins most closely related to the bacterial glycerol facilitators. The protein is localized in the plasma membrane. The physiological role of Fps1p appears to be glycerol export rather than uptake. Fps1 delta mutants are sensitive to hypo-osmotic shock, demonstrating that osmolyte export is required for recovery from a sudden drop in external osmolarity. In wild-type cells, the glycerol transport rate is decreased by hyperosmotic shock and increased by hypo-osmotic shock on a subminute time scale. This regulation seems to be independent of the known yeast osmosensing HOG and PKC signalling pathways. Mutants lacking the unique hydrophilic N-terminal domain of Fps1p, or certain parts thereof, fail to reduce the glycerol transport rate after a hyperosmotic shock. Yeast cells carrying these constructs constitutively release glycerol and show a dominant hyperosmosensitivity, but compensate for glycerol loss after prolonged incubation by glycerol overproduction. Fps1p may be an example of a more widespread class of regulators of osmoadaptation, which control the cellular content and release of compatible solutes.
