  • Comparison between Chicken and Rabbit Antibody Based Particle Enhanced Cystatin C Reagents for Immunoturbidimetry
  • 2008
  • Ingår i: Journal of immunoassay & immunochemistry. - : Informa UK Limited. - 1532-1819 .- 1532-4230. ; 29:1, s. 1-9
  • Tidskriftsartikel (refereegranskat)abstract
    • We have compared three commercial particle enhanced cystatin C reagents. One of the reagents utilizes chicken antibodies and the other two reagents are rabbit antibody based. We show that the chicken antibody based reagent yields a higher delta absorbance when reacting with the antigen. IgY coupled to latex particles show a strong scatter response even at high antigen concentrations in contrast to the steep decline in scatter previously reported for IgY antibodies in solution. The reagent also showed a low CV for duplicate samples. Laying hens thus seems as an interesting source of antibodies for particle-enhanced immunoassays.

  • Exceptional overproduction of a functional human membrane protein
  • 2007
  • Ingår i: Protein Expression and Purification. - : Elsevier BV. - 1046-5928 .- 1096-0279. ; 56:1, s. 110-120
  • Tidskriftsartikel (refereegranskat)abstract
    • Eukaryotic-especially human-membrane protein overproduction remains a major challenge in biochemistry. Heterologously overproduced and purified proteins provide a starting point for further biochemical, biophysical and structural studies, and the lack of sufficient quantities of functional membrane proteins is frequently a bottleneck hindering this. Here, we report exceptionally high production levels of a correctly folded and crystallisable recombinant human integral membrane protein in its active form; human aquaporin 1 (hAQP1) has been heterologously produced in the membranes of the methylotrophic yeast Pichia pastoris. After solubilisation and a two step purification procedure, at least 90 mg hAQP1 per liter of culture is obtained. Water channel activity of this purified hAQP was verified by reconstitution into proteoliposomes and performing stopped-flow vesicle shrinkage measurements. Mass spectrometry confirmed the identity of hAQPI in crude membrane preparations, and also from purified protein reconstituted into proteoliposomes. Furthermore, crystallisation screens yielded diffraction quality crystals of untagged recombinant hAQP1. This study illustrates the power of the yeast P. pastoris as a host to produce exceptionally high yields of a functionally active, human integral membrane protein for subsequent functional and structural characterization. • Slag inclusion formation during solidification of steel alloys and in cast iron
  • 2007
  • Licentiatavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • This thesis explores the formation of segregation and inclusions during solidification of steel and cast iron. A better understanding of the formation mechanism should result in decreasing fraction of defects during solidification of ingot and strand material. Density driven macrosegregation was studied both experimentally and theoretically to see the effect of channel segregation on the total segregation. Formation of these pencil-like segregations is due to natural convection in the solidifying metal caused by liquid enrichment of elements with lower density compared to the bulk. It is suggested to change the composition to compensate for this density difference. Inclusion precipitation can be finite by limitations in segregation. Saturated liquid is found in the last solidified areas, often between dendrites. Here the enrichment of the liquid is possible due to microsegregation. Meanwhile crystals form and solidify the elements with low solubility in the solid is pushed out in the remaining liquid. Soon the liquid is saturated to the level where spontaneous formation of inclusions occurs. Microstructure studies by aid of SEM and micro-probe measurements are analysed to find at what point during solidification process the inclusions start to form. In steel making this formation has a detrimental effect on the mechanical properties in contrary to the production of nodular cast iron where the inclusions have a beneficial effect on the graphite formation. Inoculation of cast iron aims at reaching higher number density of graphite nodules, nodule morphology modification and control of nodule distribution during solidification. Late precipitation of nucleation sites has shown to have a positive impact on preventing chill. To find the most potent inoculation agent different additives were tested. Special effort has been made to analyse the effect of oxides and sulphides as nucleation sites.

  • Analysis of Gas6 in Human Platelets and Plasma.
  • 2005
  • Ingår i: Arteriosclerosis, Thrombosis and Vascular Biology. - 1524-4636. ; 25:6, s. 1280-1286
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective - Gas6 is a member of the vitamin K-dependent protein family. Gas6- deficient mice were found to be resistant to thrombosis because of defective platelet function. Mouse Gas6 was demonstrated to be present in platelets and found to be involved in platelet aggregation. The aim of this study was to investigate the presence of Gas6 in human platelets and plasma and determine its role in platelet function. Methods and Results - The presence of Gas6 in human platelets and plasma was analyzed using sensitive immunologic methods. Mass spectrometry and ELISA were used to identify and quantify Gas6 in plasma. Gas6 was demonstrated to be present in human plasma, at a concentration determined to be 13 to 23 ng/mL (0.16 to 0.28 nM). Furthermore, plasma Gas6 levels were found to be lower in patients administered with warfarin. However, Gas6 was undetectable in human platelets. Conclusions - This is the first report to identify and quantify Gas6 in human plasma. However, Gas6 protein was not detected in human platelets, suggesting that any potential platelet-specific function could be because of Gas6 from the circulation. These findings open up new directions regarding the role of Gas6 in normal and pathophysiological situations such as inflammation, autoimmune disease, thrombosis and arteriosclerosis.

  • Clinical evaluation of a dual-side readout technique computed radiography system in chest radiography of premature neonates.
  • 2008
  • Ingår i: Acta radiologica (Stockholm, Sweden : 1987). - : SAGE Publications. - 1600-0455 .- 0284-1851. ; 49:4, s. 468-74
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND: Recently, the dual-side readout technique has been introduced in computed radiography, leading to an increase in detective quantum efficiency (DQE) compared with the single-side readout technique. PURPOSE: To evaluate if the increase in DQE with the dual-side readout technique results in a higher clinical image quality in chest radiography of premature neonates at no increase in radiation dose. MATERIAL AND METHODS: Twenty-four chest radiographs of premature neonates were collected from both a single-side readout technique system and a double-side readout technique system. The images were processed in the same image-processing station in order for the comparison to be only dependent on the difference in readout technique. Five radiologists rated the fulfillment of four image quality criteria, which were based on important anatomical landmarks. The given ratings were analyzed using visual grading characteristics (VGC) analysis. RESULTS: The VGC analysis showed that the reproduction of the carina with the main bronchi and the thoracic vertebrae behind the heart was better with the dual-side readout technique, whereas no significant difference for the reproduction of the central vessels or the peripheral vessels could be observed. CONCLUSIONS: The results indicate that the higher DQE of the dual-side readout technique leads to higher clinical image quality in chest radiography of premature neonates at no increase in radiation dose.

  • Cross-disciplinary research in analytic decision support systems
  • 2006
  • Ingår i: ITI 2006. - Zagreb : University Computing Centre SRCE, University of Zagreb. - 9537138054 ; , s. 123-128
  • Konferensbidrag (refereegranskat)abstract
    • A main problem in nearly all contexts is that unguided decision making is tremendously difficult and can lead to inefficient decision processes and undesired consequences. Therefore, decision support systems (DSSs) are of prime concern to any organization and there have been numerous approaches to such from, e.g., computational, mathematical, financial, philosophical, psychological, and sociological angles. However, a key observation is that efficient decision making is not easily performed by using methods from one discipline only. The case is rather that if real world decision making is taken seriously, several aspects must be included. This article describes some efforts of the DECIDE research group for approaching decision making and developing DSSs in a cross-disciplinary environment.
