    •  The aim of this study was to investigate whether there are differences between women and men in how chemosensory stimuli are processed. Event-related potentials from 36 participants (18 men) showed that women had larger P3 amplitudes when attending, but not when ignoring CO 2 but not for n-butanol, compared with men. The main finding was that women and men differ in cognitive measures of chemosensory processing.

  • Alterations in amino acid concentrations in the plasma and muscle in human subjects during 24 h of simulated adventure racing

    • This investigation was designed to evaluate changes in plasma and muscle levels of free amino acids during an ultra-endurance exercise and following recovery. Nine male ultra-endurance trained athletes participated in a 24-h standardized endurance trial with controlled energy intake. The participants performed 12 sessions of running, kayaking and cycling (4 x each discipline). Blood samples were collected before, during and after exercise, as well as after 28 h of recovery. Muscle biopsies were taken 1 week before the test and after exercise, as well as after 28 h of recovery. During the 24-h exercise, plasma levels of branched-chain (BCAA), essential amino acids (EAA) and glutamine fell 13%, 14% and 19% (P<0.05) respectively, whereas their concentrations in muscle were unaltered. Simultaneously, tyrosine and phenylalanine levels rose 38% and 50% (P<0.05) in the plasma and 66% and 46% (P<0.05) in muscle, respectively. After the 24-h exercise, plasma levels of BCAA were positively correlated with muscle levels of glycogen (r2=0.73, P<0.05), as was the combined concentrations of muscle tyrosine and phenylalanine with plasma creatine kinase (r2=0.55, P<0.05). Following 28-h of recovery, plasma and muscle levels of amino acids had either returned to their initial levels or were elevated. In conclusion, ultra-endurance exercise caused significant changes elevations in plasma and muscle levels of tyrosine and phenylalanine, which suggest an increase in net muscle protein breakdown during exercise. There was a reduction in plasma concentrations of EAA and glutamine during exercise, whereas no changes were detected in their muscle concentration after exercise.

    • In 1637 the Swedish Crown, encouraged by Dutch merchants, developed a plan to establish a colonial outpost in America to tap into profitable tobacco and beaver pelt trade. The same year the first cargo ships left Sweden and sailed westwards to claim their piece of America along the Delaware River. Although in many ways unsuccessful and short-lived (the colony collapsed in 1656), New Sweden became a home for generations of colonists. This chapter focuses on the different aspects of their daily life: their longing and desperation, practices of homemaking and domesticating the landscape, their perception and interactions with the neighbouring Native American groups. It discusses the ways material culture was used, exchanged and appropriated by the colonists and the local Lenape and Susquehannock in the processes of meeting, negotiations and daily coexistence.

  • Urbanism Under Sail : An Archaeology of Fluit Ships in Early Modern Everyday Life

    • In the seventeenth- and early eighteenth centuries, fluits were the most common type of merchant ship used in Baltic trade. Originally a Dutch design, the majority of all goods transported between Sweden and the Republic was carried on board such vessels. Far from all voyages reached their destination. Down in the cold brackish water of the Baltic, the preservation conditions are optimal, and several of these unfortunate vessels remain nearly intact today. Although thousands of more or less identical fluits were built, surprisingly little is known about the arrangement of space on board, their sculptural embellishment and other aspects that formed the physical component of everyday life on and alongside these ships. Fluits were a fixture in early modern society, so numerous that they became almost invisible. The study of wrecks thus holds great potential for revealing vital components of early modern life. Inspired by phenomenological approaches in archaeology, this thesis aims to focus on the lived experience of fluits. It sets out to grasp for seemingly mundane everyday activities relating to these ships, from the physical arrangements for eating, sleeping and answering nature’s call, to their rearrangement for naval use, and ends with a consideration of the architectonical contribution of the fluit to the urban landscape.

    • BACKGROUND:Many different cystatin C-based equations exist for estimating glomerular filtration rate. Major reasons for this are the previous lack of an international cystatin C calibrator and the nonequivalence of results from different cystatin C assays.METHODS:Use of the recently introduced certified reference material, ERM-DA471/IFCC, and further work to achieve high agreement and equivalence of 7 commercially available cystatin C assays allowed a substantial decrease of the CV of the assays, as defined by their performance in an external quality assessment for clinical laboratory investigations. By use of 2 of these assays and a population of 4690 subjects, with large subpopulations of children and Asian and Caucasian adults, with their GFR determined by either renal or plasma inulin clearance or plasma iohexol clearance, we attempted to produce a virtually assay-independent simple cystatin C-based equation for estimation of GFR.RESULTS:We developed a simple cystatin C-based equation for estimation of GFR comprising only 2 variables, cystatin C concentration and age. No terms for race and sex are required for optimal diagnostic performance. The equation, [Formula: see text] is also biologically oriented, with 1 term for the theoretical renal clearance of small molecules and 1 constant for extrarenal clearance of cystatin C.CONCLUSIONS:A virtually assay-independent simple cystatin C-based and biologically oriented equation for estimation of GFR, without terms for sex and race, was produced.

    • Bacterial colonization of the lower respiratory tract is frequently seen in chronic obstructive pulmonary disease (COPD), and may cause exacerbations leading to disease progression. Antimicrobial peptides comprise an important part of innate lung immunity, and not least the cathelicidin human cationic antimicrobial protein-18/LL-37. Peptidylarginine deiminases (PADIs) post-translationally modify proteins by converting cationic peptidylarginine residues to neutral peptidylcitrulline. An increased presence of PADI2 and citrullinated proteins was demonstrated in the lungs of smokers. In this study, preformed PADI4, stored in granulocytes and extracellularly in the lumina of bronchi, was found in lung tissue of individuals suffering from COPD. In vitro, recombinant human PADI2 and PADI4 both caused a time- and dose-dependent citrullination of LL-37. The citrullination resulted in impaired antibacterial activity against Staphylococcus aureus, Streptococcus pneumoniae, and nontypable Haemophilus influenzae, but less so against Pseudomonas aeruginosa. Using artificial lipid bilayers, we observed discrete differences when comparing the disrupting activity of native and citrullinated LL-37, suggesting that differences in cell wall composition are important during interactions with whole bacteria. Furthermore, citrullinated LL-37 showed higher chemotactic activity against mononuclear leukocytes than did native LL-37, but was less efficient at neutralizing lipolysaccharide, and also in converting apoptotic neutrophils into a state of secondary necrosis. In addition, citrullinated LL-37 was more prone to degradation by proteases, whereas the V8 endopetidase of S. aureus cleaved the modified peptide at additional sites, compared with native LL-37. Together, these findings demonstrate novel mechanisms whereby the inflammation-dependent deiminases PADI2 and PADI4 can alter the activites of antibacterial polypeptides, affecting the course of inflammatory disorders such as COPD.
