  • Terbinafine prevents colorectal cancer growth by inducing dNTP starvation and reducing immune suppression

    • Existing evidence indicates that gut fungal dysbiosis might play a key role in the pathogenesis of colorectal cancer (CRC). We sought to explore whether reversing the fungal dysbiosis by terbinafine, an approved antifungal drug, might inhibit the development of CRC. A population-based study from Sweden identified a total of 185 patients who received terbinafine after their CRC diagnosis and found that they had a decreased risk of death (hazard ratio=0.50) and metastasis (hazard ratio=0.44) compared with patients without terbinafine administration. In multiple mouse models of CRC, administration of terbinafine decreased the fungal load, the fungus-induced myeloid-derived suppressor cell (MDSC) expansion, and the tumor burden. Fecal microbiota transplantation from mice without terbinafine treatment reversed MDSC infiltration and partially restored tumor proliferation. Mechanistically, terbinafine directly impaired tumor cell proliferation by reducing the ratio of nicotinamide adenine dinucleotide phosphate (NADP+) to reduced form of nicotinamide adenine dinucleotide phosphate (NADPH), suppressing the activity of glucose-6-phosphate dehydrogenase (G6PD), resulting in nucleotide synthesis disruption, deoxyribonucleotide (dNTP) starvation and cell cycle arrest. Collectively, terbinafine can inhibit CRC by reversing fungal dysbiosis, suppressing tumor cell proliferation, inhibiting fungus-induced MDSC infiltration, and restoring antitumor immune response.

  • Low-Carbohydrate Diet Scores and Mortality Among Adults With Incident Type 2 Diabetes

    • OBJECTIVE: The current study aims to prospectively examine the association between postdiagnosis low-carbohydrate diet (LCD) patterns and mortality among individuals with type 2 diabetes (T2D). RESEARCH DESIGN AND METHODS: Among participants with incident diabetes identified in the Nurses' Health Study and Health Professionals Follow-up Study, an overall total LCD score (TLCDS) was calculated based on the percentage of energy as total carbohydrates. In addition, vegetable (VLCDS), animal (ALCDS), healthy (HLCDS), and unhealthy (ULCDS) LCDS were further derived that emphasized different sources and quality of macronutrients. Multivariable-adjusted Cox models were used to assess the association between the LCDS and mortality. RESULTS: Among 10,101 incident T2D cases contributing 139,407 person-years during follow-up, we documented 4,595 deaths of which 1,389 cases were attributed to cardiovascular disease (CVD) and 881 to cancer. The pooled multivariable-adjusted hazard ratios (HRs, 95% CIs) of total mortality per 10-point increment of postdiagnosis LCDS were 0.87 (0.82, 0.92) for TLCDS, 0.76 (0.71, 0.82) for VLCDS, and 0.78 (0.73, 0.84) for HLCDS. Both VLCDS and HLCDS were also associated with significantly lower CVD and cancer mortality. Each 10-point increase of TLCDS, VLCDS, and HLCDS from prediagnosis to postdiagnosis period was associated with 12% (7%, 17%), 25% (19%, 30%), and 25% (19%, 30%) lower total mortality, respectively. No significant associations were observed for ALCDS and ULCDS. CONCLUSIONS: Among people with T2D, greater adherence to LCD patterns that emphasize high-quality sources of macronutrients was significantly associated with lower total, cardiovascular, and cancer mortality.

  • Chromosome-scale assembly and evolution of the tetraploid Salvia splendens (Lamiaceae) genome

    • Polyploidization plays a key role in plant evolution, but the forces driving the fate of homoeologs in polyploid genomes, i.e., paralogs resulting from a whole-genome duplication (WGD) event, remain to be elucidated. Here, we present a chromosome-scale genome assembly of tetraploid scarlet sage (Salvia splendens), one of the most diverse ornamental plants. We found evidence for three WGD events following an older WGD event shared by most eudicots (the γ event). A comprehensive, spatiotemporal, genome-wide analysis of homoeologs from the most recent WGD unveiled expression asymmetries, which could be associated with genomic rearrangements, transposable element proximity discrepancies, coding sequence variation, selection pressure, and transcription factor binding site differences. The observed differences between homoeologs may reflect the first step toward sub- and/or neofunctionalization. This assembly provides a powerful tool for understanding WGD and gene and genome evolution and is useful in developing functional genomics and genetic engineering strategies for scarlet sage and other Lamiaceae species.

  • Risk of SARS-CoV-2 exposure among hospital healthcare workers in relation to patient contact and type of care

    • We aimed to assess prevalence of IgG antibodies to severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) and factors associated with seropositivity in a large cohort of healthcare workers (HCWs). Methods: From 11 May until 11 June 2020, 3981 HCWs at a large Swedish emergency care hospital provided serum samples and questionnaire data. Presence of IgG antibodies to SARS-CoV-2 was measured as an indicator of SARS-CoV-2 exposure. Results: The total seroprevalence was 18% and increased during the study period. Among the seropositive HCWs, 11% had been entirely asymptomatic. Participants who worked with COVID-19 patients had higher odds for seropositivity: adjusted odds ratio 1.96 (95% confidence intervals 1.59–2.42). HCWs from three of the departments managing COVID-19 patients had significantly higher seroprevalences, whereas the prevalence among HCWs from the intensive care unit (also managing COVID-19 patients) was significantly lower. Conclusions: HCWs in contact with SARS-CoV-2 infected patients had a variable, but on average higher, likelihood for SARS-CoV-2 infections.

    • Many efforts are being made to mitigate the main disadvantage of most phase change materials - their low thermal conductivities - in order to deliver latent heat energy storage systems (LHESS) with adequate perfor-mance. However, the effect of applied methods is difficult to compare as they are mostly tested for different storage types and sizes and/or different boundary and initial conditions, which hinders rapid progress in the optimization of these approaches. In this work, a previously developed method for comparing the performance of LHESS is applied to experimental results of different storage systems under different conditions and subsequently analyzed and further refined. The main idea of the method is to normalize the power with the volume and a reference temperature difference and compare its mean value plotted over the normalized mean capacity flow of the heat transfer fluid (HTF). This enables the presentation of the results in a compact and easily comparative way. Attention has to be paid when it comes to the choice of the reference temperature difference, the reference volume and the method for calculating the mean value. Two variants of calculating the mean value (time-weighted and energy-weighted) and two variants of reference temperatures for determining the temperature difference to the inlet temperature of the HTF (initial temperature and melting temperature) are applied and discussed in detail. While the method significantly increases the comparability of results, none of the options listed above are without drawbacks. Approaches are shown to reduce or eliminate these drawbacks in the future. The recommendation for comparing different LHESS under different conditions is to use the method described here and clearly state the chosen reference temperature, reference volume and method for calculating the mean value.

  • Bayesian genome scale modelling identifies thermal determinants of yeast metabolism

    • The molecular basis of how temperature affects cell metabolism has been a long-standing question in biology, where the main obstacles are the lack of high-quality data and methods to associate temperature effects on the function of individual proteins as well as to combine them at a systems level. Here we develop and apply a Bayesian modeling approach to resolve the temperature effects in genome scale metabolic models (GEM). The approach minimizes uncertainties in enzymatic thermal parameters and greatly improves the predictive strength of the GEMs. The resulting temperature constrained yeast GEM uncovers enzymes that limit growth at superoptimal temperatures, and squalene epoxidase (ERG1) is predicted to be the most rate limiting. By replacing this single key enzyme with an ortholog from a thermotolerant yeast strain, we obtain a thermotolerant strain that outgrows the wild type, demonstrating the critical role of sterol metabolism in yeast thermosensitivity. Therefore, apart from identifying thermal determinants of cell metabolism and enabling the design of thermotolerant strains, our Bayesian GEM approach facilitates modelling of complex biological systems in the absence of high-quality data and therefore shows promise for becoming a standard tool for genome scale modeling.

    • The reprogramming of metabolism in response to switching the carbon source from glucose to non-preferred carbon sources is well-studied for yeast. However, understanding how metabolic networks respond to utilize a non-natural carbon source such as xylose is limited due to the incomplete knowledge of cellular response mechanisms. Here we applied a combination of metabolic engineering, systems biology and adaptive laboratory evolution to gain insights into how yeast can perform a global rewiring of cellular processes to efficiently accompany metabolic transitions. Through metabolic engineering, we substantially enhanced the cell growth on xylose after the growth on glucose. Transcriptome analysis of the engineered strains demonstrated that multiple pathways were involved in the cellular reprogramming. Through genome resequencing of the evolved strains and reverse engineering, we further identified that SWI/SNF chromatin remodelling, osmotic response and aldehyde reductase were responsible for the improved growth. Combined, our analysis showed that glycerol-3-phosphate and xylitol serve as two key metabolites that affect cellular adaptation to growth on xylose. [Figure not available: see fulltext.].

  • Quantification of Cold-Ion Beams in a Magnetic Reconnection Jet

    • Cold (few eV) ions of ionospheric origin are widely observed in the lobe region of Earth's magnetotail and can enter the ion jet region after magnetic reconnection is triggered in the magnetotail. Here, we investigate a magnetotail crossing with cold ions in one tailward and two earthward ion jets observed by the Magnetospheric Multiscale (MMS) constellation of spacecraft. Cold ions co-existing with hot plasma-sheet ions form types of ion velocity distribution functions (VDFs) in the three jets. In one earthward jet, MMS observe cold-ion beams with large velocities parallel to the magnetic fields, and we perform quantitative analysis on the ion VDFs in this jet. The cold ions, together with the hot ions, are reconnection outflow ions and are a minor population in terms of number density inside this jet. The average bulk speed of the cold-ion beams is approximately 38% larger than that of the hot plasma-sheet ions. The cold-ion beams inside the explored jet are about one order of magnitude colder than the hot plasma-sheet ions. These cold-ion beams could be accelerated by the Hall electric field in the cold ion diffusion region and the shrinking magnetic field lines through the Fermi effect.

    • A novel metallic carbon allotrope, Q-carbon, was discovered using first-principles calculations. The named Q-carbon possessed a three-dimensional (3D) cage structure formed by carbon atoms with three ligands. The energy distribution of electrons in different orbitals revealed that Q-carbon has a low degree of s-p orbital hybridization. The calculated Li+ binding energies suggested Li+ aggregation inside Q-carbon during lithiation. As a result, a Li8C32 phase was formed and gradually expanded in Q-carbon, implying a typical two-phase transition. This allowed Q-carbon to have a constant theoretical voltage of 0.40 V, which effectively inhibited Li dendrite formation. A stable Li8C32/C-32 two-phase interface was confirmed by stress-strain analysis, and a calculated Li+ diffusion barrier of similar to 0.50 eV ensured effective Li+ diffusion along a 3D pathway. This study was of great significance for the understanding of two-phase transition of Li+ storage materials and provided a new insight into the design of new carbon materials for energy storage applications.

  • A predictive model unifying hydrogen enhanced plasticity and decohesion

    • The detrimental effect of hydrogen on metals which manifests itself as a transition from a ductile to a brittle failure mode is, for the first time, incorporated into a unified continuum-scale predictive framework. The complete Gurson model, designed to predict ductile failure by voiding, is extended to include failure by deco-hesion. Hydrogen enhanced plasticity is accounted for through acceleration of the voiding process while hydrogen induced decohesion is realized by a degradation of the decohesion threshold. The interplay between these two failure modes driven by hydrogen concentration are well captured. This model can predict a realistic level of embrittlement as well as the suppression of dimples in a hydrogen induced fracture surface. Being generic, versatile, and easy to implement, the model may serve as a basis for interpretation of laboratory ex-periments and enable the transferability of the laboratory results to the integrity assessment of engineering components in hydrogen environment.
