  • Kallikrein genes are associated with lupus and glomerular basement membrane-specific antibody-induced nephritis in mice and humans

    • Immune-mediated nephritis contributes to disease in systemic lupus erythematosus, Goodpasture syndrome (caused by antibodies specific for glomerular basement membrane [anti-GBM antibodies]), and spontaneous lupus nephritis. Inbred mouse strains differ in susceptibility to anti-GBM antibody-induced and spontaneous lupus nephritis. This study sought to clarify the genetic and molecular factors that maybe responsible for enhanced immune-mediated renal disease in these models. When the kidneys of 3 mouse strains sensitive to anti-GBM antibody-induced nephritis were compared with those of 2 control strains using microarray analysis, one-fifth of the underexpressed genes belonged to the kallikrein gene family,which encodes serine esterases. Mouse strains that upregulated renal and urinary kallikreins exhibited less evidence of disease. Antagonizing the kallikrein pathway augmented disease, while agonists dampened the severity of anti-GBM antibody-induced nephritis. In addition, nephritis-sensitive mouse strains had kallikrein haplotypes that were distinct from those of control strains, including several regulatory polymorphisms,some of which were associated with functional consequences. Indeed, increased susceptibility to anti-GBM antibody-induced nephritis and spontaneous lupus nephritis was achieved by breeding mice with a genetic interval harboring the kallikrein genes onto a disease-resistant background. Finally, both human SLE and spontaneous lupus nephritis were found to be associated with kallikrein genes, particularly KLK1 and the KLK3 promoter, when DNA SNPs from independent cohorts of SLE patients and controls were compared. Collectively, these studies suggest that kallikreins are protective disease-associated genes in anti-GBM antibody-induced nephritis and lupus.

  • Privacy of Value-Added Context-Aware Service Cloud

    • In the cloud computing era, service provider cloud and context service cloud store all your personal context data. This is a positive aspect for value-added context-aware service cloud as it makes that context information collection are easier than was the case previously. However, this computing environment does add a series of threats in relation to privacy protection. Whoever receives the context information is able to deduce the status of the owners and, generally owners are not happy to share this information. In this paper, we propose a privacy preserved framework which can be utilized by value-added context-aware service cloud. Context data and related services access privileges are determined by context-aware role-based access control (CRAC) extended from role-based access control (RAC). Privacy preserved context service protocol (PPCS) is designed to protect user privacy from exposed context information. Additionally, user network and information diffusion is combined to evaluate the privacy protection effect.

    • There is a growing demand for the efficient treatment of organic polluted wastewaters by advanced oxidation processes (AOPs) which calls for the determination of the mineralization order of ease for the organic contaminants with oxidants. The mineralization abilities of organic contaminants in AOPs are investigated in this work. Photocatalytic experiments for three representative organic contaminants are carried out, and their corresponding reaction rates are determined experimentally. Meanwhile, molar Gibbs free energy changes Delta(r)G(m)degrees for the reactions of 31 organic contaminants (10 chlorinated hydrocarbons, four brominated hydrocarbons, I I aromatic hydrocarbons and their derivatives, three chloroacetic acid, and three chloroacetyl chloride) with oxidants of (OH)-O-center dot, H2O2, O-center dot(-), O-3, and O-2 are calculated, and the mineralization order of ease is determined theoretically on the basis of Delta(r)G(m)degrees. The agreement of the theoretical and experimental mineralization abilities for most of the organic contaminants investigated implies the reliability of the determination of the mineralization ability from the magnitude of Delta(r)G(m)degrees for the mineralization of trace organic contaminants. Results also show that for most of the organic contaminants studied, the mineralization abilities are (OH)-O-center dot > H2O2 > O-center dot(-) > O-3 > O-2, and the mineralization ability of the organic contaminants depends on not only the oxidants but also the structure and properties of the organic contaminants themselves, and the degradation reaction products.

  • Step enhanced dehydrogenation of ethanol on Rh

    • We have investigated the adsorption and decomposition of ethanol on the Rh(111) and Rh(5 5 3) surfaces at room temperature with special emphasis on the dehydrogenation. We use high resolution Core level photoemission and density functional theory (DFT) based simulations. A detailed analysis of the C1s core level spectra, including analysis of the vibrational fine-structure and comparison to calculated C1s binding energy shifts, shows that the ethanol decomposes into CO, ethylidyne (C2H3) methylidyne (CH), atomic C, and hydrogen. At low ethanol exposures, CH is the dominating hydrocarbon fragment on Rh(111), whereas on Rh(553) atomic C dominates over CH, indicating an enhanced dehydrogenation due to the steps present on the latter surface. At higher ethanol exposures we find a similar behavior of atomic C dominating over hydrocarbons on Rh(553), while on Rh(111) atomic carbon remains a minority species. Our DFT based simulations show that the enhanced dehydrogenation results from a significant lowering of the CH dissociation barrier from Rh(111) to Rh(553), as well as from the dissociation changing from endothermic on Rh(111) to exothermic on Rh(553).     • Purpose: p33ING1b, as a candidate tumour suppressor gene, has been found to be expressed a proportion of oral squamous cell carcinomas (OSCCs), however, its clinicopathological significance is not studied yet. Our aim was to investigate association of p33ING1b expression with clinicopathological variables and particularly interesting new cysteine-histidine rich protein (PINCH) in OSCCs. Methods: p33ING1b expression was immumohistochemically examined in 20 normal oral mucosa specimens and 49 OSCCs. Results: Normal squamous cells showed only p33ING1b nuclear expression (no cytoplasmic expression), with a rate of 90% positive cases. While 24% of OSCCs appeared cytoplasmic expression (11 of them with weak nuclear staining) and the rest tumours (76%) were negative for p33 ING1b. Furthermore, the cases having lymph node metastasis showed a higher frequency of positive cytoplasmic expression than those without metastasis (P = 0.03). The p33ING1b cytoplasmic expression was positively related to PINCH expression (P = 0.04), the cases positive for both proteins had a high rate of the metastasis (P = 0.03). Conclusions: The transfer of p33ING1b protein from the nucleus to the cytoplasm may result in loss of normal cellular function of the protein, which might play a role in the tumourigenesis and metastasis of OSCCs. © 2007 Springer-Verlag.
