  • Ancient microbial activity in deep hydraulically conductive fracture zones within the Forsmark target area for deep geological nuclear waste disposal, Sweden

    • Recent studies reveal that organisms from all three domains of life—Archaea, Bacteria, and even Eukarya—can thrive under energy-poor, dark, and anoxic conditions at large depths in the fractured crystalline continental crust. There is a need for an increased understanding of the processes and lifeforms in this vast realm, for example, regarding the spatiotemporal extent and variability of the different processes in the crust. Here, we present a study that set out to detect signs of ancient microbial life in the Forsmark area—the target area for deep geological nuclear waste disposal in Sweden. Stable isotope compositions were determined with high spatial resolution analyses within mineral coatings, and mineralized remains of putative microorganisms were studied in several deep water-conducting fracture zones (down to 663 m depth), from which hydrochemical and gas data exist. Large isotopic variabilities of 13Ccalcite (?36.2 to +20.2‰V-PDB) and 34Spyrite (?11.7 to +37.8‰V-CDT) disclose discrete periods of methanogenesis, and potentially, anaerobic oxidation of methane and related microbial sulfate reduction at several depth intervals. Dominant calcite–water disequilibrium of 18O and 87Sr/86Sr precludes abundant recent precipitation. Instead, the mineral coatings largely reflect an ancient archive of episodic microbial processes in the fracture system, which, according to our microscale Rb–Sr dating of co-genetic adularia and calcite, date back to the mid-Paleozoic. Potential Quaternary precipitation exists mainly at ~400 m depth in one of the boreholes, where mineral–water compositions corresponded.

    • Human-induced expansion of oxygen-deficient zones can have dramatic impacts on marine systems and its resident biota. One example is the formation of the potent neurotoxic methylmercury (MeHg) that is mediated by microbial methylation of inorganic divalent Hg (HgII) under oxygen-deficient conditions. A negative consequence of the expansion of oxygen-deficient zones could be an increase in MeHg production due to shifts in microbial communities in favor of microorganisms methylating Hg. There is, however, limited knowledge about Hg-methylating microbes, i.e., those carrying hgc genes critical for mediating the process, from marine sediments. Here, we aim to study the presence of hgc genes and transcripts in metagenomes and metatranscriptomes from four surface sediments with contrasting concentrations of oxygen and sulfide in the Baltic Sea. We show that potential Hg methylators differed among sediments depending on redox conditions. Sediments with an oxygenated surface featured hgc-like genes and transcripts predominantly associated with uncultured Desulfobacterota (OalgD group) and Desulfobacterales (including Desulfobacula sp.) while sediments with a hypoxic-anoxic surface included hgc-carrying Verrucomicrobia, unclassified Desulfobacterales, Desulfatiglandales, and uncharacterized microbes. Our data suggest that the expansion of oxygen-deficient zones in marine systems may lead to a compositional change of Hg-methylating microbial groups in the sediments, where Hg methylators whose metabolism and biology have not yet been characterized will be promoted and expand.

  • Defining potential valuables through the characterisation of lake sediments : case study in Arkelstorp bay, Sweden

    • This study aims to characterise and discuss the potential use of sediment from the Arkelstorp bay located in the south of Sweden. Sediments from the Arkelstorp bay is collected and analysed on nutrient content, age, and potential contaminants. No organic environmental pollutants are found and the metal content are not elevated but still problematically high. For example, the amount of cadmium per phosphorus is 480 mg Cd kg(-1) P. However, as the carbon 14-datings showed, the sediment is preindustrial. Therefore, the amount of registered cadmium comes naturally from the surrounding environment. Arkelstorp sediments present the potential to become a source of nutrition in agriculture. The results show that the material is a good source of nutrients, with a nitrogen content of 18 g kg(-1) dry matter (DM), phosphorus 0.8 g kg(-1) DM and potassium 2.4 g kg(-1) DM respectively. However, the metal content is problematic to use the material without any pre-treatment. On the other hand, bioenergy production is expected to increase in the future, where this resource could potentially be helpful for the cultivation of bioenergy crops.

  • Influence of allochthonous dissolved organic matter on pelagic basal production in a northerly estuary

    • Phytoplankton and heterotrophic bacteria are key groups at the base of aquatic food webs. In estuaries receiving riverine water with a high content of coloured allochthonous dissolved organic matter (ADOM), phytoplankton primary production may be reduced, while bacterial production is favoured. We tested this hypothesis by performing a field study in a northerly estuary receiving nutrient-poor, ADOM-rich riverine water, and analyzing results using multivariate statistics. Throughout the productive season, and especially during the spring river flush, the production and growth rate of heterotrophic bacteria were stimulated by the riverine inflow of dissolved organic carbon (DOC). In contrast, primary production and photosynthetic efficiency (i.e. phytoplankton growth rate) were negatively affected by DOC. Primary production related positively to phosphorus, which is the limiting nutrient in the area. In the upper estuary where DOC concentrations were the highest, the heterotrophic bacterial production constituted almost 100% of the basal production (sum of primary and bacterial production) during spring, while during summer the primary and bacterial production were approximately equal. Our study shows that riverine DOC had a strong negative influence on coastal phytoplankton production, likely due to light attenuation. On the other hand DOC showed a positive influence on bacterial production since it represents a supplementary food source. Thus, in boreal regions where climate change will cause increased river inflow to coastal waters, the balance between phytoplankton and bacterial production is likely to be changed, favouring bacteria. The pelagic food web structure and overall productivity will in turn be altered. (C) 2018 The Authors. Published by Elsevier Ltd.

  • Needs and means to advance science, policy and management understanding of the freshwater system : A synthesis report

    • Fragmented and inconsistent understanding of the freshwater system limits our ability to achieve water security and sustainability under the human-driven changes occurring in the Anthropocene. To advance system-level understanding of freshwater, gaps and inconsistencies in knowledge, data, representations and links of processes and subsystems need to be identified and bridged under consideration of the freshwater system as a continuous whole. Based on such identification, a freshwater system conceptualization is developed in this report, which emphasizes four essential, yet often neglected system aspects:i) Distinction of coastal divergent catchments.ii) Four main zones (surface, subsurface, coastal, observation) of different types of freshwater change.iii) Water pathways as system-coupling agents that link and partition water change among the four change zones.iv) Direct interactions with the anthroposphere as integral system pathways across the change zones.We explain and exemplify some key implications of these aspects, identifying in the process also distinct patterns of human-driven changes in large-scale water fluxes and nutrient loads.The present conceptualization provides a basis for common inter- and trans-disciplinary understanding and systematic characterization of the freshwater system function and its changes, and of approaches to their modeling and monitoring. This can be viewed and used as a unifying checklist that can advance science, policy and management of freshwater and related environmental changes across various scales and world regions.

  • Selective recovery of nickel over iron from a nickel-iron solution using microbial sulfate reduction in a gas-lift bioreactor

    • Process streams with high concentrations of metals and sulfate are characteristic for the mining and metallurgical industries. This study aims to selectively recover nickel from a nickel-iron-containing solution at pH 5.0 using a single stage bioreactor that simultaneously combines low pH sulfate reduction and metal-sulfide formation. The results show that nickel was selectively precipitated in the bioreactor at pH 5.0 and the precipitates consisted of >= 83% of the nickel content. The nickel-iron precipitates were partly crystalline and had a metal/sulfur ratio of 1, suggesting these precipitates were NiS and FeS. Experiments focusing on nickel recovery at pH 5.0 and 5.5 reached a recovery of >99.9%, resulting in a nickel effluent concentration <0.05 mu M. The mixed microbial population included known sulfate reducers and acetogens. This study shows that selective metal precipitation in a single stage sulfate reducing bioreactor operated at low pH has the potential to produce metal-sulfides that can be used by the metallurgical industry as a resource for metal production.

  • Bacterioplankton in the light of seasonality and environmental drivers

    • Bacterioplankton are keystone organisms in marine ecosystems. They are important for element cycles, by transforming dissolved organic carbon and other nutrients. Bacterioplankton community composition and productivity rates change in surface waters over spatial and temporal scales. Yet, many underlying biological processes determining when, why and how bacterioplankton react to changes in environmental conditions are poorly understood. Here, I used experiments with model bacteria and natural assemblages as well as field studies to determine molecular, physiological and ecological responses allowing marine bacteria to adapt to their environment.Experiments with the flavobacterium Dokdonia sp. MED134 aimed to determine how the metabolism of bacteria is influenced by light and different organic matter. Under light exposure, Dokdonia sp. MED134 expressed proteorhodopsin and adjusted its metabolism to use resources more efficiently when growing with lower-quality organic matter. Similar expression patterns were found in oceanic datasets, implying a global importance of photoheterotrophic metabolisms for the ecology of bacterioplankton.Further, I investigated how the composition and physiology of bacterial assemblages are affected by elevated CO2 concentrations and inorganic nutrients. In a large-scale experiment, bacterioplankton could keep productivity and community structure unaltered by adapting the gene expression under CO2 stress. To maintain pH homeostasis, bacteria induced higher expression of genes related to respiration, membrane transport and light acquisition under low-nutrient conditions. Under high-nutrient conditions with phytoplankton blooms, such regulatory mechanisms were not necessary. These findings indicate that open ocean systems are more vulnerable to ocean acidification than coastal waters.Lastly, I used field studies to resolve how bacterioplankton is influenced by environmental changes, and how this leads to seasonal succession of marine bacteria. Using high frequency sampling over three years, we uncovered notable variability both between and within years in several biological features that rapidly changed over short time scales. These included potential phytoplankton-bacteria linkages, substrate uptake rates, and shifts in bacterial community structure. Thus, high resolution time series can provide important insights into the mechanisms controlling microbial communities.Overall, this thesis highlights the advantages of combining molecular and traditional oceanographic methodological approaches to study ecosystems at high resolution for improving our understanding of the physiology and ecology of microbial communities and, ultimately, how they influence biogeochemical processes.

Richly illustrated with twenty four color plates featuring more than one hundred photographs, this volume contains the full results of the expedition’s rapid inventories of the geology, plants, fishes, amphibians, reptiles, birds, and mammals in the Cordillera Escalera, as well as in-depth descriptions of the history, daily life, and natural resource use of local Shawi communities. Contributors also discuss threats to and opportunities for the landscape and its people and offer recommendations for sustaining biodiversity and human well-being in this megadiverse region of Peru. This volume contains the expedition team’s full report in both Spanish and English, as well as an overview in Shawi.

    • The planktonic realm from bacteria to zooplankton provides the baseline for pelagic aquatic food webs. However, multiple trophic levels are seldomly included in time series studies, hampering a holistic understanding of the influence of seasonal dynamics and species interactions on food web structure and biogeochemical cycles. Here, we investigated plankton community composition, focusing on bacterio-, phyto- and large mesozooplankton, and how biotic and abiotic factors correlate at the Linnaeus Microbial Observatory (LMO) station in the Baltic Sea from 2011 to 2018. Plankton communities structures showed pronounced dynamic shifts with recurring patterns. Summarizing the parts of the planktonic microbial food web studied here to total carbon, a picture emerges with phytoplankton consistently contributing > 39% while bacterio- and large mesozooplankton contributed ~ 30% and ~ 7%, respectively, during summer. Cyanophyceae, Actinobacteria, Bacteroidetes, and Proteobacteria were important groups among the prokaryotes. Importantly, Dinophyceae, and not Bacillariophyceae, dominated the autotrophic spring bloom whereas Litostomatea (ciliates) and Appendicularia contributed significantly to the consumer entities together with the more traditionally observed mesozooplankton, Copepoda and Cladocera. Our findings of seasonality in both plankton composition and carbon stocks emphasize the importance of time series analyses of food web structure for characterizing the regulation of biogeochemical cycles and appropriately constraining ecosystem models.
