    • Instead of sole nutrient starvation to boost algal lipid production, we addressed nutrient limitation at two different seasons (autumn and spring) during outdoor cultivation in flat panel photobioreactors. Lipid accumulation, biomass and lipid productivity and changes in fatty acid composition of Nannochloropsis oculata were investigated under nitrogen (N) limitation (nitrate:phosphate N:P 5, N:P 2.5 molar ratio). N. oculata was able to maintain a high biomass productivity under N-limitation compared to N-sufficiency (N:P 20) at both seasons, which in spring resulted in nearly double lipid productivity under N-limited conditions (0.21 g L−1 day−1) compared to N-sufficiency (0.11 g L−1 day−1). Saturated and monounsaturated fatty acids increased from 76% to nearly 90% of total fatty acids in N-limited cultures. Higher biomass and lipid productivity in spring could, partly, be explained by higher irradiance, partly by greater harvesting rate (~30%). Our results indicate the potential for the production of algal high value products (i.e., polyunsaturated fatty acids) during both N-sufficiency and N-limitation. To meet the sustainability challenges of algal biomass production, we propose a dual-system process: Closed photobioreactors producing biomass for high value products and inoculum for larger raceway ponds recycling waste/exhaust streams to produce bulk chemicals for fuel, feed and industrial material.

    • In the continual desire to reduce the environmental footprints of human activities, research efforts to provide cleaner energy is increasingly becoming vital. The effect of climate change on present and future existence, sustainable processes, and utilizations of renewable resources have been active topics within international discourse. In order to reduce the greenhouse gases emissions from traditional materials and processes, there has been a shift to more environmental friendly alternatives. The conversion of biomass to bioenergy, including biofuels has been considered to contribute to the future of climate change mitigation, although there are concerns about carbon balance from forest utilization. Bioenergy accounts for more than one-third of all energy used in Sweden and biomass has provided about 60% of the fuel for district heating. Apart from heat and electricity supply, the transport sector, with about 30% of global energy use, has a significant role in a sustainable bioenergy system. This review presents the state of the art in the Swedish bioenergy sector based on literature and Swedish Energy Agency’s current statistics. The review also discusses the overall bioenergy production and utilization in different sectors in Sweden. The current potential, challenges, and environmental considerations of bioenergy production are also discussed.

  • Dry Anaerobic Digestion of Food and Paper Industry Wastes at Different Solid Contents

    • A large volume of food is being wasted every year, while the pulp and paper industry also generate a large amount of solid wastes on a daily basis, causing environmental challenges around the world. Dry anaerobic digestion (AD) of these solid wastes is a cost-effective method for proper management. However, dry digestion of these waste streams has been restricted due to their complex structure, the presence of possible inhibitors and inappropriate operating conditions. In light of this fact, dry digestion of food waste (FW) and paper wastes (PW) was conducted at different total solid (TS) concentrations of reactor mixtures of 14%, 16%, 18% and 20% TS, corresponding to substrate to inoculum (S/I) ratio of 0.5 and 1; investigating the optimum operating conditions for effective dry digestion of these complex wastes. The highest methane yields of 402 NmlCH(4)/gVS and 229 NmlCH(4)/gVS were obtained from digestion of FW and PW, respectively at 14%TS corresponding to an S/I ratio of 0.5. Increasing the S/I ratio from 0.5 to 1 and thereby having a TS content of 20% in the reactor mixtures was unfavorable to the digestion of both substrates.

    • Introduction: In the past few decades, new methods for drug and gene delivery have been developed, among which electroporation and electrofusion have gained noticeable attention. Lately, advances in the field of immunotherapy have enabled new cancer therapies based on immune response, including monoclonal antibodies and cell vaccines. Efficient cell fusion is needed for both hybridoma production and cell vaccine preparation, and electrofusion is a promising method to achieve this goal.Areas covered: In the present review, we cover new strategies of cancer treatment related to antibody production and cell vaccines. In more detail, cell electroporation and electrofusion are addressed. We briefly describe principles of cell electroporation and focus on electrofusion and its influential factors, with special attention on the fusogenic state of the cell membrane, contact formation, the effect of electrofusion media and cell viability. We end the review with an overview of the very promising field of microfluidic devices for electrofusion.Expert opinion: In our opinion, electrofusion can be a very efficient method for hybridoma and cell vaccine production. Advances in the development of microfluidic devices and a better understanding of the underlying (biological) mechanisms will overcome the current limitations.

    • The artificially induced cell fusion is a useful experimental tool in biology, biotechnology and medicine. The electrofusion is a physical method for cell fusion that applies high-voltage electric pulses. The use of electric pulses causes cell membrane structural changes which bring the cell membrane in the so-called fusogenic state. When such fusogenic membranes are in close contact cell fusion takes place. Physical contact between fusion partners can be achieved by various methods and one of them is modified adherence method (MAM) described in detail here on B16-F1 cell line. The method is based on the fact that living cells form contacts in confluent culture. However, instead of using confluent cell culture, in modified adherence method cells are plated in suitable concentration and allowed to form contacts for only short predetermined period of time. During that time the cells are only slightly attached to the dish surface maintaining the spherical shape. Observed high fusion yields up to 50 % obtained by MAM in situ by dual-color fluorescence microscopy are among the highest in field of electrofusion. The method can be readily adapted to other anchorage-dependent cell lines.

    • Yellow pea and faba bean are potential candidates to replace soybean-based ingredients due to their suitability for cultivation in the northern hemisphere, non-genetically modified organisms cultivation practice and low risk of allergenicity. This study examined the functionality of local yellow pea and faba bean protein isolates/concentrate as meat analogue products. The most critical factors affecting the texture properties of meat analogue were also determined. Extrusion was used to produce high-moisture meat analogues (HMMAs) from yellow pea and faba bean protein isolates/concentrates and HMMAs with fibrous layered structures was successfully produced from both imported commercial and local sources. The texture properties of the HMMA produced were mainly affected by the ash, fiber and protein content and water-holding capacity of the source protein. Three extrusion process parameters (target moisture content, extrusion temperature, screw speed), also significantly affected HMMA texture. In conclusion, functional HMMA can be produced using protein isolates derived from locally grown pulses.

    • The main objective of this investigation is to determine the effect of different physical parameters on the performance of biofilters, treating hydrophobic compounds. In this respect, the effects of irrigation and water content of packings on the removal efficiency of bed in different pollutant loading rates, and gas phase flow rates, is studied. Alpha-pinene, which is produced from variety of industrial wood products, pulp and paper industries, and fragrance production units, has been selected as a model compound. Since the effectiveness of biofiltration depends strongly upon water solubility of compounds, in the case of alpha-pinene (2.5 ppm, at 25 ◦C), the process of waste gas treatment is faced with difficulties. In this paper, it is shown that performance of biofilters, treating hydrophobic contaminants, declines due to irrigation. This reduction is detected by an increase in the outlet concentration from 11% up to 22.5%. Its magnitude depends on the gas velocity inside the biofilter and outlet concentration of the bed. The result indicated that pore blocking along the bed has less effect on the performance reduction than diffusion coefficient. Also the inhibitory effects of velocity on biodegradation are considerably higher than the effects of concentration. In addition, this compost-based biofilter shows noteworthy higher elimination capacities in comparison with previous studied biofiltration systems. In this study, a maximum elimination capacity of 227 gm−3 of packing h−1 is achieved by 95% of removal efficiency. The maximum concentration in the inlet gas was 650 mgm−3.

    • A label free biosensor for direct detection of inorganic phosphate based on potential-step capacitance measurements has been developed. The high-affinity Pho84 plasma membrane phosphate/proton symporter of Saccharomyces cerevisiae was used as a sensing element. Heterologously expressed and purified Pho84 protein was immobilized on a self-assembled monolayer (SAM) on a capacitance electrode. Changes in capacitance were recorded upon exposure to phosphate compared to the control substance, phosphate analogue methylphosphonate. Hence, even without the explicit use of lipid membranes, the Pho84 membrane protein could retain its capacity of selective substrate binding, with a phosphate detection limit in the range of the apparent in vivo K(m). A linear increase in capacitance was monitored in the phosphate concentration range of 5-25 mu M. The analytical response of the capacitive biosensor is in agreement with that the transporter undergoes significant conformational changes upon exposure to inorganic phosphate, while exposure to the analogue only causes minor responses.
