Identification of Mutations Causing the White Spotted Coat Color in Swamp Buffalo

- Candidate gene analysis was used to identify mutations that is associated with white-spotted coat color phenotype in the Asian swamp buffalo (Bubalus bubalis carabanensis). The spotted coat color phenotype is found exclusively in a subpopulation of swamp buffalo that exists in the Tana Toraja region, South Sulawesi, Indonesia. Spotted buffaloes are culturally important and male spotted buffaloes are sacrificed during funeral ceremonies. Two distinct loss-of-function mutations that both cause white spotted coat color in swamp buffaloes, were identified. Positive selection has led to the accumulation of multiple mutations causing a white spotting phenotype in swamp buffalo. Our results show that genetic variation causing white spotting causes a tie to the culturally important use of these buffaloes in Tana Toraja, Indonesia.
