    • In this thesis, I examine the variation in plant species richness along gradients of productivity and disturbance in grasslands and forest habitats in southern Sweden, and I compare the documented patterns with theoretical predictions. Moreover, I evaluate the relative importance of habitat quality and habitat configuration for the occurrence of field layer species in deciduous forests. Finally, I present a new method for the determination of the regional species pool. To examine regional and local variation in plant species richness, I gathered data on species composition in plots of different size (0.001 - 1000 m2) in three vegetation types (deciduous forests, dry grasslands and coastal meadows) in four regions of southern Sweden (Öland, Gotland, Småland and Uppland). As predicted by the species pool hypothesis, differences in small-scale species richness of deciduous forests and dry grasslands were correlated with differences in the size of the regional species pool. Moreover, among plots large-scale diversity was predictive of small-scale diversity. Species diversity showed a hump-shaped relationship with productivity in forests, and was related to environmental heterogeneity and the size of the 'habitat-specific' species pool. In the two types of grassland examined, grazed sites were richer in species than abandoned sites. Moreover, both species composition and the representation of plants with different life-history characteristics differed between grazed and abandoned sites. As predicted by the intermediate disturbance hypothesis, species richness was highest at intermediate levels of grazing in coastal meadows. However, all the above patterns were scale-dependent, and not observed at all plot sizes. The occurrence of field layer species in deciduous forests was more strongly related to habitat quality (mainly soil factors) than to habitat configuration (forest area and isolation). Across species, low seed production, clonal reproduction and habitat specificity were negatively associated with isolation.

  • Association between depressive symptoms and parental stress among mothers and fathers in early parenthood : A Swedish cohort study

    • Aim To determine whether there is an association between depressive symptoms and parental stress among mothers and fathers during early parenthood in Sweden. Methods In this study, 401 mothers and 396 fathers (393 couples) were included; the Edinburgh Postnatal Depression Scale and the Sense of Coherence Scale were measured 3 months after childbirth, and the Swedish Parenthood Stress Questionnaire and the Sense of Coherence Scale after 18 months. Complete data for multivariable analysis were available for 264 mothers and 252 fathers. Results The mothers estimated greater total depressive symptoms and parental stress than the fathers did. Both the mothers and the fathers had the greatest level of stress in the sub-area 'Role restriction'. The mothers had the lowest level of stress in the sub-area 'Social isolation' and the fathers in the sub-area 'Incompetence'. The mothers perceived greater levels of stress than the fathers did in all sub-areas except for 'Social isolation', where the fathers perceived higher stress. There was an association between the parents' depressive symptoms and parental stress. The parents' own depressive symptoms at 3 months and sense of coherence and the partners' parental stress at 18 months were positively associated with the parental stress at 18 months in univariable and multivariable analyses. Conclusions Understanding the relationship between depressive symptoms and parental stress is important for health professionals so they can offer parents adequate support in early parenthood to optimize the conditions for raising a child. This knowledge should also be communicated to the parents.

  • AC losses in HTS as a function of magnetic fields with arbitrary directions

    • Although a superconductor has zero resistivity when carrying a direct current, losses do occur when it is exposed to an alternating magnetic field and/or is carrying an alternating current. The magnitude of these so-called AC losses depends on the operating temperature, the amplitude and the direction of the magnetic field, the transport current, and the frequency. Therefore, the use of high-temperature superconductors, HTSs, in electric power components such as cables, transformers or reactors, requires knowledge of the AC losses.This thesis deals with the development of AC loss models for HTSs, mainly for Bi-2223 tapes. In particular, the orientation of the applied magnetic field is taken into account in the modelling. The basis for the models is the results of experimental investigations.The basic concepts of HTSs with special emphasis on the modelling of AC losses are presented. These can be broken down into several components. Their sources and natures are described. One of the components is the hysteretic loss and it is the dominating loss in AC applications at power frequencies. Therefore, the other loss components are neglected in the modelling.Models are presented and the associated parameters are investigated with respect to their dependence of the magnetic field as well as the temperature. The AC losses for parallel and perpendicular magnetic field with respect to the wide side of the tape are calculated numerically. Moreover, a semi-empirical model for intermediate angels of the applied magnetic field is proposed.   • Context-Dependent Regulation of Hematopoietic Lineage Choice by HEBAlt

    • Hematopoietic development is controlled by combinatorial interactions between E-protein transcription factors and other lineage regulators that operate in the context of gene-regulatory networks. The E-proteins HEB and E2A are critical for T cell and B cell development, but the mechanisms by which their activities are directed to different genes in each lineage are unclear. We found that a short form of HEB, HEBAlt, acts downstream of Delta-like (DL)-Notch signaling to promote T cell development. In this paper, we show that forced expression of HEBAlt in mouse hematopoietic progenitors inhibited B cell development, but it allowed them to adopt a myeloid fate. HEBAlt interfered with the activity of E2A homodimers and with the expression of the transcription factor Pax5, both of which are critical for B cell development. However, when combined with DL-Notch signaling, HEBAlt enhanced the generation of T cell progenitors at the expense of myeloid cells. The longer form of HEB, HEBCan, also inhibited E47 activity and Pax5 expression, but it did not collaborate with DL-Notch signaling to suppress myeloid potential. Therefore, HEBAlt can suppress B cell or myeloid potential in a context-specific manner, which suggests a role for this factor in maintaining T lineage priming prior to commitment.

    • Sjogren 's syndrome is a chronic autoimmune disorder characterised by lymphocytic infiltration and hypofunction of salivary and lacrimal glands. This loss of salivary function leads to oral dryness, impaired swallowing and speech and increased infection and is associated with other autoimmune diseases and an increased risk of certain cancers. Despite the implications of this prevalent disease, diagnosis currently takes years, partly due to the diversity in patient presentation. Saliva is a complicated biological fluid with major constituents including heavily glycosylated mucins MUC5B and MUC7, important for its viscoelastic, hydrating and lubricating properties. This study investigated Sjogren 's patient 's perception of dryness (bother index questionnaires) along with the rheological, protein composition and glycan analysis of whole mouth saliva and the saliva on the mucosal surface (residual mucosal saliva) to understand the properties that most affect patient wellbeing. Sjogrenvs patients exhibited a statistically significant reduction in residual mucosal saliva, salivary flow rate and extensional rheology, spinnbarkeit (stringiness). Although the concentration of mucins MUC5B and MUC7 were similar between patients and controls, a comparison of protein Western blotting and glycan staining identified a reduction in mucin glycosylation in Sjogren 's, particularly on MUC7. LC-MS/MS analysis of O -glycans released from MUC7 by β-elimination revealed patients had an increase in core 1 sulfation and an overall reduction in sialylation resulting in a global decline of charged glycans. This was primarily due to the loss of the extended core 2 disialylated structure, with and without fucosylation. A decrease in the extended, fucosylated core 2 disialylated structure on MUC7, residual mucosal wetness and whole mouth saliva flow rate appeared to have a negative and cumulative effect on the perception of oral dryness. The observed changes in MUC7 glycosylation could be a potential diagnostic tool for saliva quality and taken into consideration for future therapies for this multifactorial syndrome.
