  • Identification of human S100A9 as a novel target for treatment of autoimmune disease via binding to quinoline-3-carboxamides.

    • Despite more than 25 years of research, the molecular targets of quinoline-3-carboxamides have been elusive although these compounds are currently in Phase II and III development for treatment of autoimmune/inflammatory diseases in humans. Using photoaffinity cross-linking of a radioactively labelled quinoline-3-carboxamide compound, we could determine a direct association between human S100A9 and quinoline-3-carboxamides. This interaction was strictly dependent on both Zn++ and Ca++. We also show that S100A9 in the presence of Zn++ and Ca++ is an efficient ligand of receptor for advanced glycation end products (RAGE) and also an endogenous Toll ligand in that it shows a highly specific interaction with TLR4/MD2. Both these interactions are inhibited by quinoline-3-carboxamides. A clear structure-activity relationship (SAR) emerged with regard to the binding of quinoline-3-carboxamides to S100A9, as well as these compounds potency to inhibit interactions with RAGE or TLR4/MD2. The same SAR was observed when the compound's ability to inhibit acute experimental autoimmune encephalomyelitis in mice in vivo was analysed. Quinoline-3-carboxamides would also inhibit TNFalpha release in a S100A9-dependent model in vivo, as would antibodies raised against the quinoline-3-carboxamide-binding domain of S100A9. Thus, S100A9 appears to be a focal molecule in the control of autoimmune disease via its interactions with proinflammatory mediators. The specific binding of quinoline-3-carboxamides to S100A9 explains the immunomodulatory activity of this class of compounds and defines S100A9 as a novel target for treatment of human autoimmune diseases.

    • Det här är en handbok som skrivits för att användas praktiskt i kampen om hyresrättens framtid. Boken har växt fram ur gemensamma erfarenheter av att ta strid mot renoveringar med höga hyreshöjningar som följd. Det har gett oss både kunskaper om lokal organisering och en övertygelse om vikten av att hyresgäster ska ha rätt till att påverka framtiden i de bostadsområden de bor i. De senaste åren har vi kunnat se hur hyreslägenheter renoveras med omfattande hyreshöjningar som följd. Konsekvenserna har blivit att många hyresgäster har tvingats flytta, ibland från bostadsområden de bott i under lång tid, då de inte längre har råd att betala hyran. Om inte hyresgäster går samman och kräver sina rättigheter kommer inget förändras och flera tusentals människor tvingas flytta. Den här handboken ska användas som ett praktiskt verktyg för lokal organisering. Vi som har gjort den här boken är engagerade i olika rörelser, några av oss är också forskare med inriktning mot bostad- spolitik och stadsutveckling. Boken har tillkommit genom en kollektiv process, där många varit inblandade för att bidra med sina erfarenheter och perspektiv. En lista med alla medverkande inklusive de rörelser som står bakom den här boken finns lägst bak.

  • Protein and DNA methylation-based scores as surrogate markers for interferon system activation in patients with primary Sjögren's syndrome

    • Objective: Standard assessment of interferon (IFN) system activity in systemic rheumatic diseases depends on the availability of RNA samples. In this study, we describe and evaluate alternative methods using plasma, serum and DNA samples, exemplified in the IFN-driven disease primary Sjogren's syndrome (pSS).Methods: Patients with pSS seropositive or negative for anti-SSA/SSB and controls were included. Protein-based IFN (pIFN) scores were calculated from levels of PD-1, CXCL9 and CXCL10. DNA methylation-based (DNAm) IFN scores were calculated from DNAm levels at RSAD2, IFIT1 and IFI44L. Scores were compared with mRNA-based IFN scores measured by quantitative PCR (qPCR), Nanostring or RNA sequencing (RNAseq).Results: mRNA-based IFN scores displayed strong correlations between B cells and monocytes (r=0.93 and 0.95, p<0.0001) and between qPCR and Nanostring measurements (r=0.92 and 0.92, p<0.0001). The pIFN score in plasma and serum was higher in patients compared with controls (p<0.0001) and correlated well with mRNA-based IFN scores (r=0.62-0.79, p<0.0001), as well as with each other (r=0.94, p<0.0001). Concordance of classification as 'high' or 'low' IFN signature between the pIFN score and mRNA-based IFN scores ranged from 79.5% to 88.6%, and the pIFN score was effective at classifying patients and controls (area under the curve, AUC=0.89-0.93, p<0.0001). The DNAm IFN score showed strong correlation to the RNAseq IFN score (r=0.84, p<0.0001) and performed well in classifying patients and controls (AUC=0.96, p<0.0001).Conclusions: We describe novel methods of assessing IFN system activity in plasma, serum or DNA samples, which may prove particularly valuable in studies where RNA samples are not available.

    • This essay takes its empirical starting point in the cultural and library policy debate that took place right before the governmental election in Sweden, September 2006. The concern of the essay is two-folded. At first, we want to show how the appropriation of certain ideological and political concepts, in times of election, tends to organize the multiplicity of rationalities and subjects into binary relations. Such binary relation is often in Swedish cultural political climate defined as social-conservative versus neoliberal or conservative. As for the social-conservative formation, we connect the uses of concepts to an immanent idea of a national subject and historical coherence; whereas for the political right we connect the use of concepts to a wider neo-liberal turn in the Swedish political language. Secondly, in a political-philosophical reflection, we will argue that the contemporary decline of cultural policy discourse on the level of governmentality is not at all adequate. Rather, we observe tendencies that points to a more complex, web like, cultural policy that have taken it’s turn into other institutional positions, from where it can continue following a somewhat social-conservative agenda. However, we are convinced that the cultural policy debate, if it wants to remain vital, needs to discuss it’s meta-political, conceptual and form related issues, rather than keep dwelling on factual matters.
