    • Bioindication has become an indispensable part of water quality monitoring in most countries of the world, with the presence and abundance of bioindicator taxa, mostly multicellular eukaryotes, used for biotic indices. In contrast, microbes (bacteria, archaea and protists) are seldom used as bioindicators in routine assessments, although they have been recognized for their importance in environmental processes. Recently, the use of molecular methods has revealed unexpected diversity within known functional groups and novel metabolic pathways that are particularly important in energy and nutrient cycling. In various habitats, microbial communities respond to eutrophication, metals, and natural or anthropogenic organic pollutants through changes in diversity and function. In this review, we evaluated the common trends in these changes, documenting that they have value as bioindicators and can be used not only for monitoring but also for improving our understanding of the major processes in lotic and lentic environments. Current knowledge provides a solid foundation for exploiting microbial taxa, community structures and diversity, as well as functional genes, in novel monitoring programs. These microbial community measures can also be combined into biotic indices, improving the resolution of individual bioindicators. Here, we assess particular molecular approaches complemented by advanced bioinformatic analysis, as these are the most promising with respect to detailed bioindication value. We conclude that microbial community dynamics are a missing link important for our understanding of rapid changes in the structure and function of aquatic ecosystems, and should be addressed in the future environmental monitoring of freshwater ecosystems.     • A major challenge in environmental risk assessment of pollutants is establishing a causal relationship between field exposure and community effects that integrates both structural and functional complexity within ecosystems. Pollution-induced community tolerance (PICT) is a concept that evaluates whether pollutants have exerted a selection pressure on natural communities. PICT detects whether a pollutant has eliminated sensitive species from a community and thereby increased its tolerance. PICT has the potential to link assessments of the ecological and chemical status of ecosystems by providing causal analysis for effect-based monitoring of impacted field sites. Using PICT measurements and microbial community endpoints in environmental assessment schemes could give more ecological relevance to the tools that are now used in environmental risk assessment. Here, we propose practical guidance and a list of research issues that should be further considered to apply the PICT concept in the field.

    • A new Arabidopsis thaliana (ecotype Columbia) genomic library has been constructed in Yeast Artificial Chromosomes: the CIC library (for CEPH, INRA and CNRS). Optimization of plant culture conditions and protoplast preparation allowed the recovery of large amounts of viable protoplasts. Mechanical shearing of DNA was minimized by isolation of DNA from protoplasts embedded in agarose. Cloning of large inserts was favored by including two successive size fractionation steps (after partial EcoRI digestion and after ligation with the vector arms), which selected DNA fragments larger than 350 kb. The library consists of 1152 clones with an average insert size of 420 kb. Clones carrying chloroplast DNA and various nuclear repeated sequences have been identified. Twenty-one per cent of the clones are found to contain chloroplast DNA. Therefore, the library represents around four nuclear genome equivalents. The clones containing 5S rDNA genes, 18S-25S rDNA sequences and the 180 bp paracentromeric repeated element account for 3.6%, 8.9% and 5.8%, respectively. Only one clone was found to carry the 160 bp paracentromeric repeated element. Given the smaller size of clones carrying Arabidopsis repeated DNA, the average size of remaining clones is around 480 kb. The library was screened by PCR amplification using pairs of primers corresponding to sequences dispersed in the genome. Seventy out of 76 pairs of primers identified from one to seven YAC clones. Thus at least 92% of the genome is represented in the CIC library. The survey of the library for clones containing unlinked DNA sequences indicates that the proportion of chimeric clones is lower than 10%.

  • A Slowly Precessing Disk in the Nucleus of M31 as the Feeding Mechanism for a Central Starburst

    • We present a kinematic study of the nuclear stellar disk in M31 at infrared wavelengths using high spatial resolution integral field spectroscopy. The spatial resolution achieved, FWHM = 0.'' 12 (0.45 pc at the distance of M31), has only previously been equaled in spectroscopic studies by space-based long-slit observations. Using adaptive optics-corrected integral field spectroscopy from the OSIRIS instrument at the W. M. Keck Observatory, we map the line-of-sight kinematics over the entire old stellar eccentric disk orbiting the supermassive black hole (SMBH) at a distance of r<4 pc. The peak velocity dispersion is 381 +/- 55 km/s(-1), offset by 0.'' 13 +/- 0.'' 03 from the SMBH, consistent with previous high-resolution long-slit observations. There is a lack of near-infrared (NIR) emission at the position of the SMBH and young nuclear cluster, suggesting a spatial separation between the young and old stellar populations within the nucleus. We compare the observed kinematics with dynamical models from Peiris & Tremaine (2003). The best-fit disk orientation to the NIR flux is [theta(l), theta(i), theta(a)] = [-33 +/- 4 degrees, 44 +/- 2 degrees, -15 +/- 15 degrees}], which is tilted with respect to both the larger-scale galactic disk and the best-fit orientation derived from optical observations. The precession rate of the old disk is Omega(P) = 0.0 +/- 3.9 km/s(-1)pc(-1), lower than the majority of previous observations. This slow precession rate suggests that stellar winds from the disk will collide and shock, driving rapid gas inflows and fueling an episodic central starburst as suggested in Chang et al.

    • Levosimendan was first approved for clinical use in 2000, when authorization was granted by Swedish regulatory authorities for the hemodynamic stabilization of patients with acutely decompensated chronic heart failure (HF). In the ensuing 20 years, this distinctive inodilator, which enhances cardiac contractility through calcium sensitization and promotes vasodilatation through the opening of adenosine triphosphate-dependent potassium channels on vascular smooth muscle cells, has been approved in more than 60 jurisdictions, including most of the countries of the European Union and Latin America. Areas of clinical application have expanded considerably and now include cardiogenic shock, takotsubo cardiomyopathy, advanced HF, right ventricular failure, pulmonary hypertension, cardiac surgery, critical care, and emergency medicine. Levosimendan is currently in active clinical evaluation in the United States. Levosimendan in IV formulation is being used as a research tool in the exploration of a wide range of cardiac and noncardiac disease states. A levosimendan oral form is at present under evaluation in the management of amyotrophic lateral sclerosis. To mark the 20 years since the advent of levosimendan in clinical use, 51 experts from 23 European countries (Austria, Belgium, Croatia, Cyprus, Czech Republic, Estonia, Finland, France, Germany, Greece, Hungary, Italy, the Netherlands, Norway, Poland, Portugal, Russia, Slovenia, Spain, Sweden, Switzerland, the United Kingdom, and Ukraine) contributed to this essay, which evaluates one of the relatively few drugs to have been successfully introduced into the acute HF arena in recent times and charts a possible development trajectory for the next 20 years.
