  • Earliest Acheulian paleolandscape reveals a 1.7 million-year-old megasite at Olduvai Gorge (Tanzania)

    • FLK West (Bed II, Olduvai Gorge) contains the oldest association of Acheulian stone tools and exploitation of fauna (including megafauna) by hominins in the Pleistocene. Recently, the FLK West paleolandscape has been intensively studied, unveiling a spatial association between archaeological materials and hydrothermal resources. A new type of landscape use by hominins has also been documented around the area where the penecontemporaneous FLK West and HWK site complex were formed, resulting in an array of habitats spanning thousands of square meters covered with large amounts of lithic artefacts. Here, we show how the intensive use of certain environments by hominins resulted in these megasites, in which hominins engaged in a variety of activities, complementary to those performed at discrete archaeological clusters like FLK West. Despite using these habitats redundantly as quarries, hominins performed extensive core reduction of several types of raw materials indicating a dexterity and careful planning undocumented in earlier periods. Here, we also show how palaeoecological reconstructions must be based on fine-scale geological analyses, given the palimpsestic nature of both geological and anthropogenic processes. The research reported here also uncovered an additional unknown source of metamorphic rocks for hominins at Olduvai, which questions a large part of previous modeling based on hominin provisioning exclusively at Naibor Soit. We also show that the manufacture of handaxes was slightly older than documented at FLK West and that they occur in isolation on the landscape in addition to being clustered at sites. This implies that at the beginning of the Acheulian, hominins were not only using and discarding handaxes at specific loci, but they also transported these tools for various activities across the landscape as part of their strategized adaptation to those environments. The occurrence of these intensively-used megasites hints at some territorial behaviors by early Acheulian hominins.

  • High IGKC-Expressing Intratumoral Plasma Cells Predict Response to Immune Checkpoint Blockade

    • Resistance to Immune Checkpoint Blockade (ICB) constitutes the current limiting factor for the optimal implementation of this novel therapy, which otherwise demonstrates durable responses with acceptable toxicity scores. This limitation is exacerbated by a lack of robust biomarkers. In this study, we have dissected the basal TME composition at the gene expression and cellular levels that predict response to Nivolumab and prognosis. BCR, TCR and HLA profiling were employed for further characterization of the molecular variables associated with response. The findings were validated using a single-cell RNA-seq data of metastatic melanoma patients treated with ICB, and by multispectral immunofluorescence. Finally, machine learning was employed to construct a prediction algorithm that was validated across eight metastatic melanoma cohorts treated with ICB. Using this strategy, we have unmasked a major role played by basal intratumoral Plasma cells expressing high levels of IGKC in efficacy. IGKC, differentially expressed in good responders, was also identified within the Top response-related BCR clonotypes, together with IGK variants. These results were validated at gene, cellular and protein levels; CD138+ Plasma-like and Plasma cells were more abundant in good responders and correlated with the same RNA-seq-defined fraction. Finally, we generated a 15-gene prediction model that outperformed the current reference score in eight ICB-treated metastatic melanoma cohorts. The evidenced major contribution of basal intratumoral IGKC and Plasma cells in good response and outcome in ICB in metastatic melanoma is a groundbreaking finding in the field beyond the role of T lymphocytes.

  • Determining the diagenetic paths of archaeofaunal assemblages and their palaeoecology through artificial intelligence : an application to Oldowan sites from Olduvai Gorge (Tanzania)

    • The implementation of deep-learning methods to the taphonomic analysis of the microscopic modification of bone-surface modifications exposed to different chemical diagenetic pathways can effectively discriminate between acidic and alkaline soil properties, indirectly reflecting different ecological conditions. Here we use this novel method to assess the sedimentary conditions of two of the oldest Oldowan archaeofaunal records (DS and PTK, Bed I) from Olduvai Gorge Bed I in Tanzania. We show how the results support different diagenetic conditions for both penecontemporaneous sites, which are appropriate for their respective locations on the palaeolandscape to which they belonged. We also show how geochemical analyses of the clay deposit that embedded both sites indicate a similar soil pH divergence. PTK was formed on an alluvial sloping surface affected by rills but draining efficiently, which resulted in alkaline soil conditions, that optimised bone-surface preservation. DS occurred in a more depressed area that underwent intermittent flooding, affecting soil chemistry by creating more acidic conditions. This impacted on bone surfaces by dynamically modifying mark morphology. This deep-learning approach has relevance for the interpretation of the local palaeoecological conditions of both assemblages and their respective depositional loci. The results presented here open a new window to the incremental information gain through the use of artificial intelligence methods in taphonomic and palaeoecological research.

  • Tailoring Dielectric Properties using Designed Polymer-Grafted ZnO Nanoparticles in Silicone Rubber

    • Polymer grafts were used to tailor the interphases between ZnO nanoparticles (NPs) and silicone matrices. The final electrical properties of the nanocomposites were tuned by the grafted interphases, by controlling the inter-particle distance and the NP-morphology. The nanocomposites can be used in electrical applications where control of the resistivity is desired. Hansen's solubility parameters were used to select a semi-compatible polymer for grafting to obtain anisotropic NP morphologies in silicone, and the grafted NPs self-assembled into various morphologies inside the silicone matrices. The morphologies in the semi-compatible nanocomposites could be tuned by steering the graft length of poly(n-butyl methacrylate) via entropic matrix-graft wetting using surface-initiated atom-transfer radical polymerization. Image analysis models were developed to calculate the radius of primary NPs, the fraction of aggregates, the dispersion, and the face-to-face distance of NPs. The dielectric properties of the nanocomposites were related to the morphology and the face-to-face distance of the NPs. The dielectric losses, above 100 Hz, for nanocomposites with grafted NPs were approximately one decade lower than those of pristine NPs. The isotropic nanocomposites increased the resistivity up to 100 times compared to that of neat silicone rubber, due to the trapping of charge carriers by the interphase of dispersed NPs and nanoclusters. On the other hand, the resistivity of anisotropic nanocomposites decreased 10–100 times when the inter-particle distance in continuous agglomerates was close to the hopping distance of charge carriers. The electrical breakdown strength increased for compatible isotropic nanocomposites, and the temperature dependence of the resistivity and the activation energy were ∼50% lower in the nanocomposites with grafted NPs. These flexible dielectric nanocomposites are promising candidates for low-loss high-voltage transmission cable accessories, mobile electronic devices, wearables and sensors.
