    • ABSTRACTBackground: Brucellosis is an economically important zoonotic disease with worldwide distribution, with low-income countries being more affected. The disease is endemic in India, a country that house the world’s largest cattle and buffalo population and produce the most milk in the world.Results: Prevalence of the disease in the country is reported as low as 1% to as high as 60% by different researchers but many of the published studies that reported higher prevalence were conducted in non-randomised samples. Based on this review, overall prevalence in the country is likely 12% or less. About 20 different risk factors are reported that contribute/predispose to occurrence of bovine brucellosis. The risk factors could be classified in four groups: host factors, farmer’s factors, managemental factors, and agro-ecological factors.   • Estimates of the Economic Cost Caused by Five Major Reproductive Problems in Dairy Animals in Assam and Bihar, India

    • Reproductive problems in dairy animals reduce fertility, prevent conception, create problems in the delivery of healthy calves, lead to postpartum complications, increase inter-calving periods, reduce milk yield, and lower overall lifetime productivity. This study aimed at understanding the incidence of reproductive problems and the cost caused by these. The study covered 954 dairy animals in Bihar and 1348 dairy animals in Assam that were selected using a multi-stage random sampling method. The costs were calculated as the sum of income losses and expenditures incurred. The major cost incurred resulted from extended calving intervals (46.1% of the total cost), followed by loss through salvage selling (38.1%), expenditure for treatment of repeat breeders (5.9%), loss of milk production (5.3%) and expenditure for extra inseminations (2.0%). About one fifth of the selected reproductive problems were left untreated. The estimated cost of reproductive problems was Indian Rupees (INR) 2424.9 (USD 36.1) per dairy animal per year (of the total dairy animal population) which represented approximately 4.1% of the mean value loss of dairy animals (INR 58,966/USD 877) per year. Reproductive problems were significantly (p < 0.001) higher among improved (exotic breed or cross-bred) dairy animals than indigenous (native breed or nondescript indigenous) dairy animals. The study suggests that with the increase of improved dairy animal population, the loss may further increase. The study concludes that any economic estimation of reproduction problems based on aetiology without confirmatory diagnoses could be highly misleading because of the complex nature of the problems.Simple SummaryLarge ruminant dairy animals (i.e., cattle and buffalo) suffer from several reproductive problems (such as abortion) that reduce ther ability to produce milk and offspring, resulting in huge economic costs to farmers; however, there are few studies in India that estimate such costs. Therefore, an attempt was made to assess the economic cost of five major reproductive problems in two of the poorest Indian states—Assam and Bihar. We estimated the cost by interviewing 534 randomly selected dairy farming households in both the states. Based on this, we found that 32.9% of dairy animals (milking, not-milking and heifer) in Assam and 43.1% dairy animals in Bihar suffered from one or more reproductive problems. The most common reproductive problem was failing to conceive after breeding (23.2% of surveyed dairy animals) followed by retained placenta (6.1%), abortion (4.9%), purulent vaginal discharge (2.9%) and stillbirths (1.0%). It was estimated that the selected reproductive problems caused an annual economic cost of Indian Rupees (INR), 3963.1 million (USD 59.0 million) in Assam, and INR 30,500.0 million (USD 453.9 million) in Bihar. The study concludes that adequate awareness, capacity building, adoption of good reproductive health management practices, proper farm record keeping and improved access to quality veterinary services are essential to address reproductive problems and reduce the cost caused by these reproductive problems.

  • Seroprevalence and Risk Factors of Brucella Infection in Dairy Animals in Urban and Rural Areas of Bihar and Assam, India

    • This study assessed seropositivity of Brucella infection in dairy animals and risk factors associated with it. The cross-sectional study used multi-stage, random sampling in the states of Bihar and Assam in India. In total, 740 dairy animals belonging to 534 households of 52 villages were covered under this study. Serological testing was conducted by indirect enzyme-linked immunosorbent assay (iELISA). Animal-level Brucella seropositivity was found to be 15.9% in Assam and 0.3% in Bihar. Seropositivity in urban areas (18.7%) of Assam was found to be higher than in rural areas (12.4%). Bihar was excluded from the risk factor analysis, as only one Brucella seropositive sample was detected in the state. A total of 30 variables were studied for assessing risk factors, of which 15 were selected for multivariable regression analyses following a systematic process. Finally, only three risk factors were identified as statistically significant. It was found that animals belonging to districts having smaller-sized herds were less likely (p < 0.001) to be Brucella seropositive than animals belonging to districts having larger-sized herds. Furthermore, the chance of being Brucella seropositive increased (p = 0.007) with the increase in age of dairy animals, but decreased (p = 0.072) with the adoption of artificial insemination (AI) for breeding. We speculated that the identified risk factors in Assam likely explained the reason behind lower Brucella seropositivity in Bihar, and therefore any future brucellosis control program should focus on addressing these risk factors.

  • Analyses of Extended-Spectrum-β-Lactamase, Metallo-β-Lactamase, and AmpC-β-Lactamase Producing Enterobacteriaceae from the Dairy Value Chain in India

    • The consumption of milk contaminated with antibiotic-resistant bacteria poses a significant health threat to humans. This study aimed to investigate the prevalence of Enterobacteriaceae producing β-lactamases (ESBL, MBL, and AmpC) in cow and buffalo milk samples from two Indian states, Haryana and Assam. A total of 401 milk samples were collected from dairy farmers and vendors in the specified districts. Microbiological assays, antibiotic susceptibility testing, and PCR-based genotyping were employed to analyze 421 Gram-negative bacterial isolates. The overall prevalence of β-lactamase genes was 10% (confidence interval (CI) (7–13)), with higher rates in Haryana (13%, CI (9–19)) compared to Assam (7%, CI (4–11)). The identified β-lactamase genes in isolates were blaCMY, blaMOX, blaFOX, blaEBC, and blaDHA, associated with AmpC production. Additionally, blaCTX-M1, blaSHV, and blaTEM were detected as ESBL producers, while blaVIM, blaIMP, blaSPM, blaSIM, and blaGIM were identified as MBL producers. Notably, Shigella spp. were the dominant β-lactamase producers among identified Enterobacteriaceae. This study highlights the presence of various prevalent β-lactamase genes in milk isolates, indicating the potential risk of antimicrobial-resistant bacteria in dairy products. The presence of β-lactam resistance raises concern as this could restrict antibiotic options for treatment. The discordance between genotypic and phenotypic methods emphasizes the necessity for comprehensive approaches that integrate both techniques to accurately assess antibiotic resistance. Urgent collaborative action incorporating rational and regulated use of antibiotics across the dairy value chain is required to address the global challenge of β-lactam resistance.

    • The use and misuse of antibiotics in both humans and animals contributes to the global emergence of antimicrobial resistant (AMR) bacteria, a threat to public health and infection control. Currently, India is the world’s leading milk producer but antibiotic usage within the dairy sector is poorly regulated. Little data exists reflecting how antibiotics are used on dairy farms, especially on small-scale dairy farms in India. To address this lack of data, a study was carried out on 491 small-scale dairy farms in two Indian states, Assam and Haryana, using a mixed method approach where farmers were interviewed, farms inspected for the presence of antibiotics and milk samples taken to determine antibiotic usage. Usage of antibiotics on farms appeared low only 10% (95% CI 8–13%) of farmers surveyed confirmed using antibiotics in their dairy herds during the last 12 months. Of the farms surveyed, only 8% (6–11%) had milk samples positive for antibiotic residues, namely from the novobiocin, macrolides, and sulphonamide classes of antibiotics. Of the farmers surveyed, only 2% (0.8–3%) had heard of the term “withdrawal period” and 53% (40–65%) failed to describe the term “antibiotic”. While this study clearly highlights a lack of understanding of antibiotics among small-scale dairy farmers, a potential factor in the emergence of AMR bacteria, it also shows that antibiotic usage on these farms is low and that the possible role these farmers play in AMR emergence may be overestimated.

    • Animal products are highly nutritious, but also highly perishable. In India milk is an important source of animal protein, but problems with low quality of the milk, high degrees of adulterated milk on the market, high bacterial loads, and sometimes presence of zoonotic pathogens persist. Most dairy farmers in India are resource-poor small-holders, often with limited knowledge about the importance of food safety and hygiene. Milk quality problems including adulteration and bacterial contamination is common in the country.This paper describes a training intervention for improved food safety in Guwahati, Assam, India, conducted in 2009–2013. The training was designed to be short, simple and customized, cheap to deliver, easily accessible, and accompanied by incentives to bring change in knowledge, attitude and practices (KAP). In 2014 three outcomes were assessed: changed KAP; milk production; and, mastitis prevalence. Selected food safety hazards were also assessed, although their management had not been included in training. We found evidence of improved KAP among trained farmers, 14% higher milk production, and a tendency towards less mastitis, but no effects on food safety hazard levels.This study shows that a training intervention can have a medium-term impact, while the issue of food safety is more complex and cannot be assumed to automatically follow from even successful training.

    • ABSTRACTBackground: Food-borne diseases are contributing to health burdens globally, especially in developing countries. In India, milk production is important for nutrition security, but milk products are prone to contamination with pathogens. In Assam, a state in Northeast India, a novel hygiene intervention was conducted in 2009–2011, and the knowledge, attitudes and practices among milk producers, milk traders and sweet makers were assessed.Methods: The first survey was conducted in 2009 and included 405 producers, 175 traders and 220 sweet makers from 4 districts. The second survey was conducted in 2012 with 161 producers and 226 traders from 2 districts, both trained and untrained participants. In addition to questionnaires, observations on hygiene were done and samples were analysed for Escherichia coli.Results: In 2009 only 13.0%, 9.1%, and 33.1% of producers, traders and sweet makers respectively believed diseases could be transmitted by milk.     • Background: In India, milk production is important for both the economy and the provision of nutritious food. However, the productivity of the livestock is affected by circulating infectious diseases, and some zoonotic diseases, such as brucellosis, may cause a heavy impact on the farm as the disease cause abortions and reproductive failures in bovines, with chronic febrile illness in humans.Methods: 249 dairy farms in the state of Haryana, India, were interviewed, and collected raw milk from 81 were analyzed for antibodies towards Brucella abortus.Results: More samples were positive using milk ring test (MRT) (55.6%, 45/81) than using enzyme-linked immunosorbent assay (ELISA) (29.6%, 24/81), with all ELISA positive samples also positive in MRT. The ELISA results were used for risk factor analyses. Seropositive farms were significantly (p = 0.015) larger than seronegative, with an average 7.9 cattle, compared to 4.9. Seropositive farms were more likely to report stillbirth occurring the last year, and a significantly higher proportion of seropositive farms reported retained placenta (odds ratio 5.2).Conclusion: This study showed that Brucella seroprevalence is high among farms in Haryana, and a control program is needed to ensure improved human and animal health, as well as improved livestock productivity.

    • Brucellosis is one of the most important zoonotic diseases in the world and one of the most important causes of reproductive disorders in livestock. The disease is endemic in India. The This study aimed to assess seroprevalence, risk factors, and clinical predictors of Brucella infection in dairy animals, along with the relevant knowledge and practices of farmers and also the cost of reproductive disorders in dairy farming. A cross-sectional study was conducted in the states of Assam and Bihar, India through a primary survey of 534 randomly selected dairy farming households and serological investigation of 740 blood samples collected from their dairy animals (cattle and buffalo).  From laboratory analysis, animal-level Brucella seropositivity was 15.9% in Assam and 0.3% in Bihar. Three identified risk factors for Brucella seropositivity in Assam were geographical location (district) (p<0.001), age of dairy animals (p=0.008) and mating system (p=0.07).  Occurrence of retained placenta was the most important clinical symptom (OR 20.7) for predicting Brucella infection. Only a small percentage of farmers (3.4%, n=18) knew about brucellosis. Actions to prevent Brucella infection by the farming community were negligible. The estimated cost of reproductive disorders was USD 36.1 per dairy animal per year which represented approximately 4.1% of the mean value of dairy animals (USD 877). Reproductive disorders caused an estimated annual economic cost of USD 59.0 million in Assam and USD 453.9 million in Bihar. The findings help in identifying future research priorities and limitations and designing effective Brucella control programme in dairy farming in India and beyond.
