  • Surface plasmon resonance aptasensor for Brucella detection in milk

    • A Surface Plasmon Resonance (SPR) aptasensor was developed for the detection of Brucella melitensis (B. melitensis) in milk samples. Brucellosis is a bacterial zoonotic disease with global distribution caused mostly by contaminated milk or their products. Aptamers recognizing B. melitensis were selected following a whole bacteria-SELEX procedure. Two aptamers were chosen for high affinity and high specificity. The high affinity aptamer (B70 aptamer) was immobilized on the surface of magnetic silica core-shell nanoparticles for initial purification of the target bacteria cells from milk matrix. Another aptamer, highly specific for B. melitensis cells (B46 aptamer), was used to prepare SPR sensor chips for sensitive determination of Brucella in eluted samples from magnetic purification since direct injection of milk samples to SPR sensor chips is known for a high background unspecific signal. Thus, we integrated a quick and efficient magnetic isolation step for subsequent instant detection of B. melitensis contamination in one ml of milk sample by SPR with a LOD value as low as 27 ± 11 cells.

    • In the present study, we succesfully synthesized electrospun MnFe2O4 nanofibers (NFs) from poly(N-vinylpyrrolidone)/manganese(II) nitrate composite by electrospinning and then as-spun NFs were calcined 450 degrees C for 2 h in air atmosphere to remove the polymer matrix and fabricate inorganic MnFe2O4 nanofibers. In order to investigate the sintering behavior of MnFe2O4 nanofibers in air atmosphere, the synthesized as-spun nanofibers were calcined with different calcination rates. Thus the effect of calcination rate on structure and morphology of nanofibers were discussed clearly. The structural, magnetic, morphological, spectroscopic and thermal characterizations were also done by XRD, VSM, TEM, SEM, FTIR and TG analysis. In the presence of slow calcination rate, only peaks of MnFe2O4 could be observed on other hand in the presence of rapid calcination rate, formation of an impurity was observed. Scanning electron microscope images revealed that MnFe2O4 nanorods possess a broader range size distribution with higher particle size. Also, magnetic properties were both size and shape dependent.

    • This research develops a multi-criteria framework and Quality Assurance checklist (QA) for Positive-Energy-Districts (PEDs). PEDs have a large potential to contribute to reach urban climate goals but require also changes of planning, development, and transformation processes of neighborhoods. Our understanding of PEDs has been broadened to include aspects of circularity and economic viability, ensuring that PED concepts not only contribute to environmental sustainability but are also economically feasible. In this enhanced perspective, Digital Twins serving as essential carriers of information and facilitators of communication, effectively bridging the gap between different dimensions of sustainable development and practical implementation and monitoring. This study is part of an ongoing European project, Digital Twin for PEDs (DT4PEDs), with Living Labs in three countries: Austria, Sweden, and Turkey. The presented study addresses the central research question, "In what phases can specific requirements be linked to the process to ensure PED development throughout the entire process using a Digital Twin?" within the context of the DT4PEDs project. Based on a participatory workshop with stakeholders from partner countries and a parallel literature review, the study encompasses three key areas. First, it seeks to consolidate property development practices across Austria, Sweden, and Turkey, including both new construction and retrofitting, into a unified framework. Second, it formulates a PED multi-criteria framework, complemented by a Quality Assurance (QA) checklist. Finally, the recommendations for digital twin are proposed to support energy related information flows and stakeholder dialogue.
