    • This paper presents an approach to automatically generate predetermined motion time system (PMTS) based assembly instruction lists from simulations within digital human modelling (DHM) software. A formal high-level instruction language is used to instruct an automated manikin to perform assembly operations. The manikin, the language and the assembled objects are composed into a discrete model. In a post-computational step, the model is used to construct PMTS instruction lists by mapping the manikin motions, assembled objects and the language instructions onto a set of PMTS keywords. The described approach is implemented in the DHM software IMMA and tested on industrial cases of manual assembly in the automotive industry.

    • The peroxisome proliferator-activated receptor (PPAR) alpha agonist WY 14,643 increased the secretion of apolipoprotein (apo) B-100, but not that of apoB-48, and decreased triglyceride biosynthesis and secretion from primary rat hepatocytes. These effects resulted in decreased secretion of apoB-100-very low density lipoprotein (VLDL) and an increased secretion of apoB-100 on low density lipoproteins/intermediate density lipoproteins. ApoB-48-VLDL was also replaced by more dense particles. The proteasomal inhibitor lactacystin did not influence the recovery of apoB-100 or apoB-48 in primary rat hepatocytes, indicating that co-translational (proteasomal) degradation is of less importance in these cells. Treatment with WY 14,643 made the recovery of apoB-100 sensitive to lactacystin, most likely reflecting the decreased biosynthesis of triglycerides. The PPAR alpha agonist induced a significant increase in the accumulation of pulse-labeled apoB-100 even after a short pulse (2-5 min). There was also an increase in apoB-100 nascent polypeptides, indicating that the co-translational degradation of apoB-100 was inhibited. However, a minor influence on an early posttranslation degradation cannot be excluded. This decreased co-translational degradation of apoB-100 explained the increased secretion of the protein. The levels of apoB-48 remained unchanged during these pulse-chase experiments, and albumin production was not affected, indicating a specific effect of PPAR alpha agonists on the co-translational degradation of apoB-100. These findings explain the difference in the rate of secretion of the two apoB proteins seen after PPAR alpha activation. PPAR alpha agonists increased the expression and biosynthesis of liver fatty acid-binding protein (LFABP). Increased expression of LFABP by transfection of McA-RH7777 cells increased the secretion of apoB-100, decreased triglyceride biosynthesis and secretion, and increased PPAR alpha mRNA levels. These findings suggest that PPAR alpha and LFABP could interact to amplify the effect of endogenous PPAR alpha agonists on the assembly of VLDL.

  • Numerical Techniques for Acoustic Modelling and Design of Brass Wind Instruments

    • Acoustic horns are used in musical instruments and loudspeakers in order to provide an impedance match between an acoustic source and the surrounding air. The aim of this study is to develop numerical tools for the analysis and optimisation of such horns, with respect to their input impedance spectra. Important effects such as visco-thermal damping and modal conversion are shown to be localised to different parts of a typical brass instrument. This makes it possible to construct hybrid methods that apply different numerical techniques in different parts of the instrument. Narrow and slowly flaring parts are modelled using a one-dimensional transmission line analogy, and the rapidly flaring bell is modelled using a two-dimensional finite-difference method. The connection between the different regions is done by the aid of impedance boundary conditions. The use of such boundary conditions is investigated with respect to the required number of degrees of freedom. Numerical shape optimisation is employed in order to design horns with desired impedance characteristics throughout a design frequency band. A loudspeaker horn is optimised with respect to its sound power output, and a brass instrument is optimised with respect to its intonation. The horns are modelled using the finite-element method and a transmission line analogy. In order to achieve rapid convergence of the optimisation, gradient based minimisation algorithms are used. A prerequisite for success is the ability to accurately and inexpensively compute the gradient of the objective function. The gradient for the finite-element method is computed by an adjoint equation technique, whereas for the transmission line analogy, it is derived by formal differentiation of the model. In order to find smooth solutions, a smoothing technique is used, where optimisation is done with respect to the right hand side of a Poisson type equation.

  • Thermal conductivity versus depth profiling of inhomogeneous materials using the hot disc technique

    • Transient measurements of thermal conductivity are performed with hot disc sensors on samples having a thermal conductivity variation adjacent to the sample surface. A modified computational approach is introduced, which provides a method of connecting the time-variable to a corresponding depth-position. This allows highly approximate—yet reproducible—estimations of the thermal conductivity vs. depth. Tests are made on samples incorporating different degrees of sharp structural defects at a certain depth position inside a sample. The proposed methodology opens up new possibilities to perform non-destructive testing; for instance, verifying thermal conductivity homogeneity in a sample, or estimating the thickness of a deviating zone near the sample surface (such as a skin tumor), or testing for presence of other defects
