    • We use agarose gel electrophoresis to characterize how the monovalent catioinic surfactant cetyltrimethylammonium bromide (CTAB) compacts double-stranded DNA, which is detected as a reduction in electrophoretic DNA velocity. The velocity reaches a plateau at a ratio R = 1.8 of CTAB to DNA-phosphate charges, i.e., above the neutralization point, and the complexes retain a net negative charge at least up to R = 200. Condensation experiments on a mixture of two DNA sizes show that the complexes formed contain only one condensed DNA molecule each. These CTAB-DNA globules were further characterized by time-resolved measurements of their velocity inside the gel, which showed that CTAB does not dissociate during the migration but possibly upon entry into the gel. Using the Ogston-model for electrophoresis of spherical particles, the measured in-gel velocity of the globule is quantitatively consistent with CTAB having two opposite effects, reduction of both the electrophoretic charge and DNA coll size. In the case of CTAB the two effects nearly cancel, which can explain why opposite velocity shifts (globule faster than uncomplexed DNA) have been observed with some catioinic condensation agents. Dissociation of the complexes by addition of anionic surfactants was also studied. The DNA release from the globule was complete at a mixing ratio between anionic and cationic surfactants equal to 1, in agreement with equilibrium studies. Circular DNA retained its supercoiling, and this demonstrates a lack of DNA nicking in the compaction-release cycle which is important in DNA transfection and purification applications.

    • Investigations of polluted brownfield sites and sample analyses are expensive, and the resulting data are often of poor quality. Efforts are needed, therefore, to improve the methods used in investigations of brownfield sites to both reduce costs and improve the quality of the results. One approach that could be useful for both of these purposes is the triad strategy, developed by the US Environmental Protection Agency, in which managing uncertainty is a central feature. In the investigations reported here, a field study was conducted to identify possible ways in which uncertainties could be managed in practice. One example considered involves optimizing the uncertainty by adjusting the sizes of samples and the efforts expended in analytical work according to the specific aims of the project. In addition, the potential utility of several toxicity assessment methods for screening sites was evaluated. As well as presenting the results of these assessments, in this contribution we discuss ways in which a flexible work strategy and screening methods inspired of the triad philosophy could be incorporated into the Swedish approach to remediate brownfield sites. A tiered approach taking advantage of field and screening methods is proposed to assess brownfield sites focusing on the response and acceptable uncertainty that are required for the task.

  • Long-term risk of cardiovascular disease in Hodgkin lymphoma survivors : retrospective cohort analyses and a concept for prospective intervention

    • Previous studies have shown increased cardiovascular mortality as late side effects in Hodgkin lymphoma (HL) patients. This study identifies stratifying risk factors for surveillance and defines concepts for a clinical feasible and noninvasive prospective protocol for intervention of cardiovascular side effects. HL patients diagnosed between 1965 and 1995 (n = 6.946) and their first-degree relatives (FDR) were identified through the Swedish Cancer Registry and the Swedish Multigeneration Registry. For the HL and FDR cohort, in-patient care for cardiovascular disease (CVD) was registered through the Hospital Discharge Registry, Sweden. Standard incidence ratios of developing CVD for the HL cohort were calculated. A markedly increased risk for in-patient care of CVD was observed in HL patients with HL diagnosed at age 40 years or younger and with more than 10 years follow-up. In the HL survivors, a family history of congestive heart failure (CHF) and coronary artery disease (CAD) increased the risk for these diseases. The Swedish Hodgkin Intervention and Prevention study started in 2007. In the pilot feasibility study for prospective intervention (47 patients), about 25% of the cases had side effects and laboratory abnormalities. These patients were referred to a cardiologist or general practitioner. In the prospective cohort, a positive family history for CHF or CAD could be a stratifying risk factor when setting up a surveillance model. The prospective on-going study presents an intervention model that screens and treats for comorbidity factors. This article also presents an overview of the study concept.

    • Förmågan att effektivt utnyttja och kritiskt förhålla sig till olika informationsresurser – ”information literacy” eller informationskompetens – är en central färdighet i det moderna samhället. Det utgör också en allt viktigare del av lärandet i högre utbildning. Syftet med detta paper är tvåfaldigt: 1) att problematisera relationen mellan informationskompetens som en generell färdighet och mer specifika ämnesorienterade kunskaper och färdigheter; 2) ge exempel på och utveckla redskap för lärare att på ett ämnesmässigt integrerat sätt arbeta med, utveckla och examinera studenternas informationskompetens. Ett huvudspår i framställningen är att synliggöra hur träningen i informationskompetens kan stimulera och hjälpa studenterna till fördjupade ämneskunskaper.

  • Long-term rearing of Arctic charr Salvelinus alpinus under different salinity regimes at constant temperature

    • Arctic charr Salvelinus alpinus of the Holar strain (mean+/- s. e. body mass= 152.1+/-3.1g) were reared at four different salinity regimes at a constant temperature of 7.4 degrees C. Two groups were given a three-month acclimation in salinity 18 before the salinity was increased to either 25 or 29 (groups called A25 and A29), and two groups were reared in salinities 25 or 29 over the full experimental period of 409 days (groups called F25 and F29). In the first 3months, the A25 and A29 groups had the highest growth rates. By October 2011, there were no significant differences (two-way nested ANOVA, P> 0.05) in the mean body masses among A25, F25 and F29 (c. 1450 g), whereas A29 had a lower mean mass (1282 g). The growth in the last period from October 2011 to January 2012 was reduced by sexual maturation in the highest salinity regimes (A29 and F29), whereas fish in groups A25 and F25 showed high growth throughout the study. Males in all salinity groups had higher growth rates than females for the most part of the study, but the divergence between the sexes was most pronounced in the highest salinity regimes. All salinity groups showed distinct changes in Na+, K+-ATPase activity, with high activity in spring and summer, and lower activity in the autumn. Plasma sodium (Na+) levels were stable indicating that none of the experimental groups had problems in maintaining hydromineral balance during the study. While plasma leptin levels were not affected by salinity regimes, it was noted that these levels were 13-30% higher in fish with empty guts compared with those having food in their gut at the time of sampling. This suggests a link between leptin levels and food intake, indicating that this hormone may play a role in food intake and energy allocation in fishes.

  • Samverkan för innovation : En fallstudie av mötet mellan akademi, industri och sjukvård

    • Samverkan kan leda till innovation, konkurrenskraftiga företag, förstklassig forskning samt välfungerande myndigheter och institutioner. I den politiska debatten idag finns en förväntan att Sverige ska upprätthålla sin konkurrenskraft och bemöta samhällets utmaningar genom innovation och att vägen till innovation går via samverkan. Avhandlingen bygger på en studie av ett samverkansprojekt vars syfte var att skapa innovation för att minska antalet vårdrelaterade infektioner i Sverige. Projektet som studerats ses som en transdisciplinär ansats med aktörer som representerade akademi, industri samt hälso- och sjukvård.Syftet med avhandlingen är att vidareutveckla kunskapen om interorganisatorisk samverkan för innovation. Detta görs genom ett tredelat bidrag, till teoribildningen kring samverkan för innovation som börjat växa fram, till den samverkande praktiken inom både privat och offentlig sektor samt till politiker och beslutsfattare som styr fördelning av statliga anslag till forskning och innovation.Fallstudien som ligger till grund för avhandlingen är baserad på en etnografiskt inspirerad studie. Empiriskt material samlades in och skapades tillsammans med aktörerna i projektet under drygt två års tid genom intervjuer och deltagande observation.Studien visar att interorganisatorisk samverkan består av flera dimensioner och kan förstås på flera nivåer. Interorganisatorisk samverkan innebär inte bara att det är olika organisationer som ska göra en gemensam ansträngning. Organisationerna består av olika människor med olika discipliner och professioner vilka bygger på olika utgångspunkter och sätt att se på världen. Samverkan kan ses som ett sätt att fylla mellanrummen mellan organisationer istället för att bygga broar över gränser. I de organisatoriska mellanrummen kan aktörer från olika organisationer, med olika discipliner och professioner mötas utan institutionaliserade roller, i en receptiv kontext där innovation kan skapas.
